Activation of the G-protein coupled estrogen receptor 1 (GPER1) reduces transient receptor potential vanilloid 1 (TRPV1) activity and human iPSC-derived nociceptive neuron firing.

Chronic pain affects approximately one fifth of people worldwide and displays a huge burden for health care institutions. Some chronic pain conditions have a higher prevalence in women than men. Previous studies related these sex-specific differences to sex-hormone receptor activation and interaction with pain-related receptors like the transient receptor potential vanilloid 1 (TRPV1). Here, especially estrogen seems to play a crucial role. While the role of estrogen receptor alpha and beta in modulating TRPV1 has already been studied, the interplay of the membrane-bound and G-protein -coupled

Tissue-selective estrogen complex for women who experience breast discomfort or vaginal bleeding when on hormone therapy.

OBJECTIVE: The objective of this study was to evaluate the effects of switching from hormone therapy to tissue-selective estrogen complex (TSEC) in women who experience vaginal bleeding or breast discomfort. METHODS: This retrospective cohort study included 82 postmenopausal women who received TSEC after switching from another hormone therapy due to adverse events. Changes in symptoms and imaging after switching to TSEC were evaluated. RESULTS: The mean age was 56.9 years. The women were switched to TSEC due to vaginal bleeding in 56.1% and breast discomfort in 47.6% (multiple choices were a

Molecular pathways: digoxin use and estrogen-sensitive cancers--risks and possible therapeutic implications.

Digoxin, a phyto-estrogen, binds with estrogen receptors (ER) and can cause gynecomastia. Among women currently using digoxin, breast and uterus cancer incidences are significantly increased (approximate risk ratios, 1.3-1.5). Both cancers are often estrogen sensitive. In contrast, ovary and cervix cancers are relatively estrogen insensitive, and incidence is unaffected by digoxin exposure. When digoxin use stops, incidence rapidly reverts to that in nonusers. These patterns parallel those of estrogen, suggesting that digoxin works via ER-stimulated proliferation of ductal and/or acinar cells,

Estrogen's master switch: NFκB-NLRP3, SIRT1-HMGB1, and TLR4 triad tamed for brain rescue in ageing and neurodegeneration.

Estrogen operates as a pleiotropic steroidal, neuroendocrine modulator to combat accelerated brain ageing and neurodegeneration by addressing a convergent inflammatory-metabolic trio. Estrogen receptor-dependent neural cellular signalling reduces TLR4-mediated immune priming and NF-κB activation, preventing NLRP3 inflammasome assembly and pro-inflammatory cytokine release. Concurrently, estrogen increases SIRT1 activity, restoring metabolic and epigenetic equilibrium while inhibiting HMGB1 acetylation, translocation, extracellular release, and activation of the stress-response pathway. Coordin

Decoding estrogen receptor and GPER biology: structural insights and therapeutic advances in ERα-positive breast cancer.

Classical estrogen receptors, ERα and ERβ, along with the membrane-bound G-protein-coupled estrogen receptor (GPER), play critical roles in driving ERα-positive breast cancer (BC). Clinical management of this subtype relies on endocrine therapy (ET), which targets ER signaling through selective estrogen receptors modulators (SERMs), degraders (SERDs), and aromatase inhibitors (AIs). While ET has significantly reduced recurrence and mortality rates, acquired resistance remains a major therapeutic challenge. Activating ESR1 mutations, which encode constitutively active ERα variants, are detected

Oral selective estrogen receptor degraders (SERDs) for the treatment of hormone receptor-positive, HER2-negative breast cancer title.

INTRODUCTION: Endocrine therapy (ET) remains the backbone of treatment for hormone receptor-positive (HR+), HER2-negative breast cancer. However, resistance, frequently driven by acquired ESR1 mutations detected in up to 40% of patients progressing on first-line ET, represents a major clinical challenge. Oral selective estrogen receptor degraders (SERDs) have been developed to overcome the limitations of first-generation agents and achieve more potent ER targeting. AREAS COVERED: This review summarizes the mechanism of action of oral SERDs compared with prior endocrine therapies and evaluates

A Novel Membrane Estrogen Receptor Activated by STX Induces Female Sexual Receptivity through an Interaction with mGluR1a.

Membrane initiated estradiol signaling has been shown to be vital for multiple physiological processes. Several receptors have been proposed to mediate the actions of estradiol at the membrane. Here, we examined the ability of STX, an agonist of a novel putative membrane estrogen receptor, to activate sexually receptive behavior in the female rat. Infusions of STX into the arcuate nucleus of the hypothalamus resulted in the activation and internalization of μ-opioid receptors in the medial preoptic nucleus, an action that is required for lordosis behavior. Indeed, STX was able to augment sexua

Ovariectomy Exacerbates Acute Ethanol-Induced Tachycardia: Role of Nitric Oxide and NMDA Receptors in the Rostral Ventrolateral Medulla.

Ethanol consumption influences cardiovascular functions. In humans, acute consumption of ethanol causes dose-dependent tachycardia. Our previous study showed that ethanol-induced tachycardia might involve decreased nitric oxide (NO) signaling in the brain's medulla. NMDA receptors, another important target of ethanol, are one of the upstream signals of nitric oxide. Reports showed the modulation of NMDA receptor function by estrogen or estrogen receptors. The present study aims to examine the hypothesis that depletion of estrogen by ovariectomy (OVX) might modulate ethanol-induced tachycardia

Effects of 17β-estradiol on leptin signaling in anterior pituitary of ovariectomized rats.

Leptin is secreted predominantly by adipocytes and exerts its role mainly by interaction with the long form of leptin receptor (LEPR_V2). It has been identified that LEPR_V2 is widely distributed in various tissues, including the anterior pituitary. Cross-talk between leptin and estrogens has been indentified. Estrogen is known to modulate the tissue-specific expression of LEPR_V2 and leptin in ovariectomized (OVX) rats, a model of postmenopausal condition. Our previous data showed that 17β-estradiol (E2) up-regulated the expression of LEPR_V2 protein and mRNA in rat dorsal root ganglion (DRG)

GPER1: A Hidden Key in Cardio-Renal Axis-Unveiling its Therapeutic Potential for Heart and Kidney Disease.

Cardiovascular and kidney diseases are tightly interconnected through a bidirectional cardio-renal axis and exhibit pronounced sex-specific differences, implicating estrogen-associated signaling networks as critical modulators of disease susceptibility and progression. Among these, G protein-coupled estrogen receptor 1 (GPER1,GPR30) has emerged as a rapid signaling hub integrating vascular function, inflammatory responses, oxidative stress, and tissue remodeling across the cardiac, renal, and vascular tissues. However, interpretation of the extensive phenotype literature-largely derived from s

Bazedoxifene exhibits antiestrogenic activity in animal models of tamoxifen-resistant breast cancer: implications for treatment of advanced disease.

PURPOSE: There is compelling evidence to suggest that drugs that function as pure estrogen receptor (ER-α) antagonists, or that downregulate the expression of ER-α, would have clinical use in the treatment of advanced tamoxifen- and aromatase-resistant breast cancer. Although such compounds are currently in development, we reasoned, based on our understanding of ER-α pharmacology, that there may already exist among the most recently developed selective estrogen receptor modulators (SERM) compounds that would have usage as breast cancer therapeutics. Thus, our objective was to identify among av…

Nuclear receptors: potential biomarkers for assessing physiological functions of soy proteins and phytoestrogens.

Soy consumption is associated with decreased incidence of chronic diseases, including cardiovascular diseases, atherosclerosis, diabetes, osteoporosis, and certain types of cancers. However, consumption of high amounts of soy isoflavones may adversely influence endocrine functions, such as thyroid function and reproductive performance, because of their structural similarity to endogenous estrogens. Nuclear receptors are a group of transcription factors that play critical roles in the regulation of gene expression and physiological functions through direct interaction with target genes. Modulat

Hypothyroidism Alters Uterine Kisspeptin System and Activity Modulators in Cyclic Rats.

Hypothyroidism causes ovarian dysfunction and infertility in women and animals and impairs the hypothalamic expression of kisspeptin (Kp). However, kisspeptin is also expressed in the genital system, and the lack of the Kp receptor (Kiss1r) in the uterus is linked to reduced implantation rates. This study investigated the impact of hypothyroidism on the uterine expression of Kp and Kiss1r in female rats throughout the estrous cycle and the associated changes in uterine activity modulators. Hypothyroidism was induced through daily administration of propylthiouracil (PTU) over a period of 14 day

Identification and characterization of novel estrogen receptor-beta-sparing antiprogestins.

The steroid hormones estrogen and progesterone together regulate the development and maintenance of the female reproductive system. The actions of these two hormones are mediated by their respective nuclear receptors located within overlapping cell populations in target organs. The molecular mechanism of action of these two hormones has been defined to a large extent using estrogen receptor (ER) and progesterone receptor (PR) antagonists. In the case of ER, the available antagonists are highly receptor selective. With respect to PR, however, the available antiprogestins also interact with the

Disruption of estrogen signaling by developmental exposure to BPA and TBT causes long term functional deficits in zebrafish retina.

Bisphenol A (BPA) and tributyltin (TBT) are two endocrine disrupting compounds (EDC) that have opposite effects on estrogen signaling. BPA is an estrogen agonist that binds to all estrogen receptor types. TBT is an aromatase inhibitor that binds to the enzyme aromatase, preventing the synthesis of estrogen from testosterone. Both estrogen receptors and aromatase are localized to the retina and estrogen signaling is required for proper eye and retinal neurogenesis. Abnormal eye growth and retinal changes are reported immediately after developmental exposure to either EDC consistent with the rol
