Gene transfer to human fetal pulmonary tissue developed in immunodeficient SCID mice.

Human fetal lung rudiments (8-12 weeks of development) undergo considerable growth upon microsurgical ectopic implantation in the xenograft-tolerant SCID mouse, and differentiate into a lung-like tissue that includes: (i) bronchial structures lined with pseudostratified, secretory, ciliated epithelium surrounded by smooth muscle and cartilage rings, (ii) submucosal glands, and (iii) alveolar sacs. Normal expression of the cystic fibrosis transmembrane conductance regulator (CFTR) protein was detected by immunostaining in those grafts, and similar differentiation was observed from either normal

Performance of American Indian children with fetal alcohol syndrome on the test of language development.

Alcohol is a teratogen known to have deleterious effects on the developing embryo and fetus. Language deficits secondary to central nervous system dysfunction are among the sequelae of Fetal Alcohol Syndrome (FAS). Limited information is available regarding the language deficits associated with FAS, particularly among the population of American Indians in whom FAS is highly prevalent. The TOLD-P and TOLD-I were administered to 27 American Indian children: 10 with FAS and 17 normally developing control subjects. Pure-tone screening, immittance measurement, and vision screening preceded language

Development and regulation of growth and differentiated function in human and subhuman primate fetal gonads.

We have attempted to summarize the research on primate fetal gonadal development that has occurred over the past three decades. Many similar … racrine agents. Although many parts of the puzzle are still missing it is probable that, similar to fetal development of other endocrine tissues and to events in postnatal gonads, these local regulators have important functions. Currently, primate fetal gonadal research is lacking in at least two key aspects: 1) the definition of paracrine and autocrine nonsteroida …

Analysis of the development of T cells by transferring precursors into cultured fetal thymus with a microinjector.

Development of T cells from precursors in the bone marrow or fetal thymus cells was investigated by transferring these cells into deoxyguanosine (dGuo)-treated organ cultured fetal thymus (FTOC) with a microinjector (micro i.t. method). Donor type T cells could be generated after transfer by the micro i.t. method of precursors present both in bone marrow and fetal thymus. The kinetics of the generation of T cells from these precursors was similar to that found in in vivo intra thymus transfer systems. As low as 300 fetal thymus cells were shown to be sufficient to repopulate a dGuo-FTOC. On th

Testes of XX in equilibrium with XY chimeric mice develop from fetal ovotestes.

The majority of XX in equilibrium with XY chimeric mice develop into fertile males. The sexual differentiation of the gonads in these animals has been examined on days 12-14 postocoitum to determine if their development parallels that of normal testes. It was found that 50% of chimeric fetuses, the proportion predicted to be XX in equilibrium with XY, had neither normal testes nor ovaries. Instead, ovotestes were present, with varying proportions of presumptive ovarian and testicular tissue. On day 12 the ovotestes were organized with testicular tissue in the central region and ovarian tissue

The effects of Love Canal soil extracts on maternal health and fetal development in rats.

The effects of a solvent extract of the surface soil of the Love Canal chemical dump site, Niagara Falls, New York, and of a natural extract, or leachate, which is drained from the canal for treatment, on the maternal health and fetal development were determined in rats. The solvent extract, which was contaminated with 2,3,7,8-tetrachlorodibenzo-p-dioxin (2, 3,7,8-TCDD) at 170 ppb and numerous other chlorinated organic compounds with the primary identified components being the isomers of benzenehexachloride (BHC), was dissolved in corn oil and administered by gavage to pregnant rats at 0,25,75…

Maternal ethanol consumption and fetal development in the rat: a comparison of ethanol exposure techniques.

Comparison of the polydipsia and liquid diet procedures for ethanol exposure were used in female rats, either before and/or during gestation. Autopsy of the fetuses on day 20 of gestation found a significant weight reduction in the polydipsia-exposed animals that could be attributed to ethanol. No such effect was found in the liquid diet-exposed animals. The data are discussed in relation to the human fetal alcohol syndrome.

The effect of progesterone on fetal and placental development in normal and protein-energy-restricted rats.

Maternal protein-energy restriction (25% of the ad libitum intake) during the first 10 days of pregnancy resulted in severely altered fetal growth rates. Fetal development was assessed by body weight, brain weight, brain DNA, and brain protein content on fetal days 16, 18, 20 and at term. The individual placentas were also examined (weight, DNA and protein content) on each of these fetal days. Progesterone was administered commencing with day 3 of pregnancy until the day Caesarian section was done, in an attempt to rehabilitate placental development. This treatment did not improve placental de…

Intrapartum fetal asphyxia: clinical characteristics, diagnosis, and significance in relation to pattern of development.

The clinical and fetal heart rates and acid-base characteristics and their sequelae have been reviewed in 587 patients. The relevant clinical factors in the asphyxia group were the preterm fetus, the intrauterine growth retarded fetus, maternal toxemia, and midforceps delivery. The duration of the developing metabolic acidosis in the asphyxia group ranged from terminal to the last two hours of labor. Marked patterns of total decelerations and moderate and marked patterns of late decelerations are of predictive value in the diagnosis of intrapartum fetal asphyxia with a trend to an increased in

Chronic Fetal Leucine Infusion Does Not Potentiate Glucose-Stimulated Insulin Secretion or Affect Pancreatic Islet Development in Late-Gestation Growth-Restricted Fetal Sheep.

BACKGROUND: Growth-restricted fetuses have attenuated glucose-stimulated insulin secretion (GSIS), smaller pancreatic islets, less pancreatic β-cells, and less pancreatic vascularization compared with normally growing fetuses. Infusion of leucine into normal late-gestation fetal sheep potentiates GSIS, as well as increases pancreatic islet size, the proportion of the pancreas and islet comprising β-cells, and pancreatic and islet vascularity. In addition, leucine stimulates hepatocyte growth factor (HGF ) mRNA expression in islet endothelial cells isolated from normal fetal sheep. OBJECTIVE:

Human placental lactogen and unconjugated estriol concentrations in twin pregnancy: monitoring of fetal development in intrauterine growth retardation and single intrauterine fetal death.

Human placental lactogen and unconjugated estriol concentrations in maternal serum were evaluated in 100 uneventful twin pregnancies, and these values were compared with those observed in 16 twin pregnancies associated with intrauterine growth retardation or single intrauterine fetal death. In pregnancies associated with intrauterine growth retardation (n = 8), human placental lactogen levels were at the lower limit of normal range for singleton pregnancies, whereas estriol levels were normal in most cases. When one of the fetuses had died before week 33 of pregnancy (n = 5), both human placen

Global epigenetic immunohistochemical markers reveal developmental connections of distinct cell types during fetal development.

PURPOSE: The prenatal development of the human retina is a complex process orchestrated by several epigenetic mechanisms. This study investigated some global epigenetic markers (histone H3, histone H4, 5-methylcytosine, 5-hydroxymethylcytosine) during the fetal development of the human retina in 34 human eyes from 11 to 38 weeks of gestation. METHODS: Immunohistochemical staining was performed to assess the distribution and intensity of the selected epigenetic markers across developmental stages. For the general retinal staining intensity, a score from 0 to 3 was used, whereas for the percent…

FNDC5 deficiency alters placental development, maternal body weight, and fetal-placental growth in mice.

UNLABELLED: Irisin, a secreted myokine cleaved from FNDC5, regulates energy homeostasis and vascular function, but its role in pregnancy remains poorly understood. Here, using a murine model, we tested how global FNDC5 deficiency affects maternal metabolism, placental development and fetal growth, extending our earlier evidence that irisin mitigates human placental oxidative stress, reduces anti-angiogenic signaling and promotes trophoblast differentiation-factors involved in preeclampsia. Global FNDC5 knockout (KO) and wild-type (WT) mice were subjected to timed-mating and maternal, fetal and

Maternal dietary citrulline supplementation increases fetal growth and programs pancreatic development in the lambs.

Intrauterine growth restriction (IUGR), caused by maternal undernutrition, impairs fetal growth and increases the risk for postnatal metabolic dysfunction. L-arginine can mitigate these effects; however, its use in sheep is limited by ruminal microbial degradation. Interestingly, L-citrulline, the precursor for arginine synthesis, bypasses ruminal catabolism and may be a practical alternative. This study evaluated if maternal L-citrulline supplementation to nutrient restricted ewes from gestational days (GD) 28 to 140 (term = 147) enhances fetal growth in lambs. Gestating ewes were fed 50% of

Evaluation of fetal brain development in growth restriction subtypes using brain MRI volume measurement.

BACKGROUND: This retrospective study aims to explore the value of brain MRI volume measurements in evaluating fetal brain development in fetal growth restriction (FGR) fetuses, by comparing 2 subtypes of FGR with appropriate gestational age (AGA) fetuses. METHODS: A total of 169 fetal brain MRI with suspected abnormal development, identified through ultrasound screening at this hospital between 2021 and 2025 were analyzed,11 cases were excluded. 98 cases were FGR fetuses (43 early-onset subtypes and 55 late-onset subtypes), and 60 were AGA fetuses, gestational age ranging from 26 to 37 weeks.

Tensor veli palatini muscle and palatine aponeurosis: Their independent fetal development and delayed meeting.

To demonstrate the prenatal morphology of the palatine aponeurosis (AP) and establish how the tendon of the tensor veli palatini (TVP) muscle meets the aponeurosis, we examined serial histological sections of 25 heads from human fetuses at approximately 9-16 weeks of gestational age (GA). At 9 weeks GA, the initial AP was identified as mesenchymal condensation that extended anteriorly to attach to the palatine bone. The TVP muscle extended inferiorly and curved along the greater palatine nerve; however, the TVP tendon does not reach the AP. At 10 weeks GA, the AP had developed as a tendinous o

Amniotic fluid as a novel delivery route of 3-hydroxybutyrate during fetal brain development.

Understanding how fetuses receive essential nutrients during pregnancy is crucial for optimal brain development. We observed significantly higher levels of 3-hydroxybutyrate (3HB) in the amniotic fluid surrounding human fetuses during healthy late pregnancy compared to maternal blood. 3HB concentrations in the maternal blood were 0.12 mmol/L, while those in the amniotic fluid were 0.56 mmol/L. The rate-limiting enzyme of ketogenesis, mitochondrial 3-hydroxy-3-methylglutaryl-CoA synthase 2 (HMGCS2), was highly expressed in the cells of the chorionic plate. The placenta likely plays a crucial ro
