Pharmacology of appetite suppression: implication for the treatment of obesity.

Given the current global epidemic of obesity there is a demand for new anti-obesity drugs to overcome the problem. Many pharmacological agents reduce food intake and significantly decrease body mass when administered to animals but affect feeding behaviour in a profoundly different way indicating the variety of biological mechanisms by which such agents act (appetite verses non-appetite). More limited clinical data demonstrates that some of the same drugs produce decreases in food intake and weight loss in humans. A few of these drugs do so by modifying the functioning of the appetite system a

Normal-weight individuals experience an increased drive to eat and greater meal-satiety response in response to weight-loss: results from the NUTRILEX project.

While there is an increasing prevalence of individuals with "normal weight" engaged in diet-induced weight loss, few studies have examined t … our profile by validated questionnaires. Food reward was assessed in a fasted state using the Leeds Food Preferences Questionnaire. Subjective appetite sensations were measured before and after a standardized meal using visual analogue scales and satiety quotients were calculated. Fasting hunger and desire to eat increased at T1 (p < 0.01) and T2 (p < 0.001) while …

Cultural determinants of food attitudes in anosmic patients.

The decrease in food enjoyment is a major factor why patients suffer from depression when having anosmia, or total loss of smell. While we have some knowledge about how food preferences and attitudes change with dysosmia, these findings are limited because other factors such as culture are not factored in. It is likely that the culture in which an anosmic patient identifies with will influence how their smell loss impacts their relationship with food. This study examined the current attitudes within the United States and Germany towards foods, focusing on the comparison between anosmic patient…

Nutritional considerations of a paediatric gluten-free food guide for coeliac disease.

The gluten-free (GF) diet is the only treatment for coeliac disease (CD). While the GF diet can be nutritious, increased reliance on processed and packaged GF foods can result in higher fat/sugar and lower micronutrient intake in children with CD. Currently, there are no evidence-based nutrition guidelines that address the GF diet. The objective of this cross-sectional study was to describe the methodological considerations in forming a GF food guide for Canadian children and youth (4-18 years) with CD. Food guide development occurred in three phases: (1) evaluation of nutrient intake and diet

Knowing Is Not Eating: Child-Reported Food Perception Versus Actual Dietary Intake in Southern Peru.

Background/Objectives: Understanding whether children's perceptions of food, that is, what a child recognizes as healthy, are associated with their actual consumption is key to designing effective nutritional interventions, but this has rarely been directly tested in the children themselves. This study aimed to determine whether children's food perceptions are associated with their actual recent intake. Methods: As secondary objectives, the cultural salience of foods perceived as healthy and unhealthy was characterized in 129 schoolchildren (52 girls, 77 boys; ages 8-12) at a school in Moquegu

Repeated semaglutide treatment attenuates cocaine-vs-food choice in male and female rats.

The present study determined the effectiveness of the glucagon-like peptide 1 agonist semaglutide to attenuate cocaine-vs-food choice in male and female rats. Repeated 5-day semaglutide treatment decreased cocaine choice and significantly reduced body weight. These preclinical results support the clinical evaluation of semaglutide as a candidate cocaine use disorder medication.

Transection of the glossopharyngeal nerve reduces energy and sugar intake but does not affect fat intake or meal patterns in rats offered a palatable cafeteria diet.

Taste information is transmitted to the brain via branches of cranial nerves VII (chorda tympani, CT, and greater superficial petrosal, GSP), IX (glossopharyngeal, GL), and X (superior laryngeal branch of the vagus) with each nerve innervating relatively segregated fields of taste buds on the tongue, palate, and laryngeal epithelium. Approximately 70% of taste buds reside on the tongue and are innervated by the CT or GL. Previously we found that rats offered a 5-choice cafeteria diet after combined transection of the CT and GL (2NX) ate more fat, less sugar, and larger, but fewer meals each da

Immersive virtual contexts, engagement, and emotions: How do these factors influence sensory acceptance?

Although virtual reality (VR) holds great potential in sensory research, further investigations are needed to assess the impact of immersion on consumer evaluations. Previous studies have limitations, such as a small number of evaluators, inconsistent graphical quality of virtual environments, lack of comparisons with traditional laboratory settings, and insufficient data on consumer engagement during virtual assessments. To address these gaps, this study evaluated consumer engagement using a presence scale, along with acceptance and emotions evoked during the consumption of an industrial beer

Causal analysis of dietary preferences and the risk of endometriosis using large-scale population data.

Dietary factors have recently been recognized as potentially influential in the pathogenesis of endometriosis (EM), yet studies on the causal relationship between dietary preferences and EM are limited. The present study aimed to explore the causal relationships between 187 dietary preferences and EM using Mendelian randomization (MR) methods. This study utilised genome-wide association study data from over 500,000 European participants for dietary preferences and 64,658 EM patients from Finland. Dietary preferences with potential causal relationships to EM were identified using two-sample MR

Aging decreases preferences for salts, but not for sucrose, and alters morphology of fungiform taste pores in mice.

Mice are commonly used for laboratory research, due in large part to the widespread advancement in the genetic toolbox, such as reporters, knock-in, and knockout mice. The effects of aging on the taste system in mice have been largely unstudied. The aim of this study was to examine whether taste preferences to sucrose, NaCl, and NH4Cl were associated with aging and changes in ultrastructural characteristics of fungiform taste pores using scanning electron microscopy. Thirty-minute 2-bottle preference tests in wild-type mice indicated that preferences for NaCl and NH4Cl, but not sucrose, were s

Food choice and neural reward systems in adolescents with anorexia nervosa and atypical anorexia nervosa.

BACKGROUND: Adolescence is a critical developmental period for the study of anorexia nervosa (AN), an illness characterized by extreme restriction of food intake. The maturation of the reward system during adolescence combined with recent neurobiological models of AN led to the hypothesis that early on in illness, restrictive food choices would be associated with activity in nucleus accumbens reward regions, rather than caudate regions identified among adults with AN. METHODS: Healthy adolescents (HC, n = 41) and adolescents with AN or atypical AN (atypAN, n = 76) completed a Food Choice Task

Gut-brain circuits for fat preference.

The perception of fat evokes strong appetitive and consummatory responses1. Here we show that fat stimuli can induce behavioural attraction even in the absence of a functional taste system2,3. We demonstrate that fat acts after ingestion via the gut-brain axis to drive preference for fat. Using single-cell data, we identified the vagal neurons responding to intestinal delivery of fat, and showed that genetic silencing of this gut-to-brain circuit abolished the development of fat preference. Next, we compared the gut-to-brain pathways driving preference for fat versus sugar4, and uncovered two

Goal Directed and Self-Control Systems in Bulimia Nervosa: An fMRI Study.

BACKGROUND: Binge eating is apparently the opposite of the strict control over food intake typically set by "maladaptive dieters". Using functional magnetic resonance imaging (fMRI), we investigated the role of goal-directed behaviors, and the related use of self-control, in binge-related food choices in patients with Bulimia Nervosa (BN). METHOD: While undergoing fMRI, women aged 18-35 with BN (N = 35) and healthy control women (N = 26) rated foods for healthiness and tastiness and then made food choices on a 5 points Likert scale between two conflicting options: one food with lower healthin

The Protein Status of Rats Affects the Rewarding Value of Meals Due to their Protein Content.

BACKGROUND: Protein status is controlled by the brain, which modulates feeding behavior to prevent protein deficiency. OBJECTIVE: This study tested in rats whether protein status modulates feeding behavior through brain reward pathways. METHODS: Experiments were conducted in male Wistar rats (mean ± SD weight; 230 ± 16 g). In experiment 1, rats adapted for 2 wk to a low-protein (LP; 6% of energy) or a normal-protein (NP; 14% of energy) diet were offered a choice between 3 cups containing high-protein (HP; 50% of energy), NP, or LP feed; their intake was measured for 24 h. Development of novel tools to measure food neophobia in children.

BACKGROUND: The main tool currently used to measure food neophobia (the Food Neophobia Scale, FNS, developed by Pliner & Hobden, 1992) may not remain optimal forever. It was developed around 25 years ago, and the perception and availability of "novel" and "ethnic" foods may have changed in the meantime. Consequently, there is a need for developing updated tools for measuring food neophobia. OBJECTIVE: To develop novel tools to measure food neophobia in children. DESIGN: Based on a review of 13 designs to assess food neophobia and willingness to try unfamiliar foods, a Food Neophobia Test Too

Temperamental approach/withdrawal and food neophobia in early childhood: Concurrent and longitudinal associations.

The purpose of the present study was to investigate whether temperamental approach/withdrawal processes were concurrently and longitudinally associated with parent ratings and behavioral observations of food neophobia at 4.5 years of age. Additionally, maternal feeding practices were examined as potential moderators of the association between toddler temperament and food neophobia. Data were drawn from a longitudinal study following individuals (n = 82) from infancy through early childhood. At 18 months of age, toddlers were observed in an unfamiliar laboratory setting with an experimenter and

Testosterone prevents but not reverses anhedonia in middle-aged males and lacks an effect on stress vulnerability in young adults.

Middle-aged male rats are more vulnerable than young adult ones to develop anhedonia when exposed to chronic mild stress (CMS). Clinical studies support the idea that in aged subjects the low testosterone (T) levels are related with their higher stress vulnerability and that this hormone possesses antidepressant-like actions. In this study we evaluated the role of gonadal hormones--mainly T--on the depressive-like behavior of middle-aged and young adult male rats submitted to CMS. In middle-aged rats we analyzed the effect of T restitution (at the levels of young adult animals) given 3 weeks b

Mosquito (Diptera: Culicidae) bloodmeal sources during a period of West Nile virus transmission in Puerto Rico.

Host bloodmeals of indigenous Caribbean mosquitoes have not been studied previously. We identified vertebrate DNA in 90 blood-engorged mosquitoes belonging to four genera (Aedes, Culex, Deinocerites, and Uranotaenia) and 12 species that were collected in Puerto Rico within a geographic and temporal focus of West Nile virus transmission in 2007. It was found that 62 (68.8%) bloodmeals were from reptiles, 18 (20.0%) from birds, and 10 (11.1%) from mammals. Only one bloodmeal of 18 derived from Culex (Culex) species was passerine, suggesting a preference for nonpasserine birds and other vertebrat
