Metabolomic analyses reveal lipid abnormalities and hepatic dysfunction in non-human primate model for Yersinia pestis.

INTRODUCTION: Pneumonic plague is caused by the aerosolized form of Yersinia pestis and is a highly virulent infection with complex clinical consequences, and without treatment, the fatality rate approaches 100%. The exact mechanisms of disease progression are unclear, with limited work done using metabolite profiling to study disease progression. OBJECTIVE: The aim of this pilot study was to profile the plasma metabolomics in an animal model of Y. pestis infection. METHODS: In this study, African Green monkeys were challenged with the highly virulent, aerosolized Y. pestis strain CO92, and

Postnatal changes in serotonergic innervation to the hippocampus of methyl-CpG-binding protein 2-null mice.

Rett syndrome is a progressive neurodevelopmental disorder caused by mutations in the methyl-CpG-binding protein 2 (MeCP2) gene. Previous reports have revealed serotonergic function to be altered in the medullas of patients with Rett syndrome and in an animal model of the disease. However, it has remained unclear whether a genetic loss of MeCP2 disrupts serotonergic innervation to the forebrain. In this study, we measured levels of monoamines by high-performance liquid chromatography with electrochemical detection in selected regions of the forebrains of Mecp2-null mice (Mecp2-/y) and wild-typ

Host range and zoonotic potential linked to P-like fimbrial (PLF) adhesin specificity in avian pathogenic Escherichia coli.

Fimbrial adhesins are surface-associated bacterial proteins that contribute to colonization, tissue tropism, and biofilm formation. Genomic analysis of the avian pathogenic Escherichia coli (APEC) strain QT598 identified a plasmid-localized fimbrial operon, termed plf which encodes P-like (PL) fimbriae. Herein, we investigated the role of P-like (PL) fimbriae, encoded on the ColV plasmid of APEC strain QT598, in a natural host turkey respiratory infection model. We determined that deletion of the plf genes reduced colonization in the lungs of turkeys. The PlfG class II fimbrial adhesin from AP

Sex chromosome effects unmasked in angiotensin II-induced hypertension.

Sex differences in mean arterial pressure (MAP) are reported in many experimental models of hypertension and are ascribed to gonadal sex based on studies showing that gonadectomy and gonadal hormone replacement affect MAP. The interpretation of these studies, however, has been confounded by differences in the sex chromosome complement (XX versus XY). To investigate the sex chromosome complement independent of gonadal sex, we used the 4 core genotype mouse model in which gonadal sex is separated from the sex chromosome complement enabling comparisons among XX and XY females and XX and XY males.

Number of chromocentres in the nuclei of mouse Sertoli cells in relation to the strain and age of males from puberty to senescence.

Nuclei of mouse Sertoli cells were examined on air-dried toluidine blue-stained preparations to analyse factors influencing the aggregation of heterochromatin into chromocentres. For the CBA strain males tested at 1.3, 2, 3, 6, 9 and 12 months of age, mean numbers of heterochromatin bodies were 4.8, 2.4, 2.1, 1.8, 1.6 and 1. 4, respectively; two chromocentres predominated from 2 to 9 months of age. In the KE strain, heterochromatin aggregation was significantly accelerated; nuclei containing only one chromocentre were predominant from 6 months. The number of chromocentres did not change with t

Role of sex and gender in autoimmune thyroid diseases: mechanisms and knowledge gaps.

Autoimmune thyroid diseases (AITDs), mainly Hashimoto's thyroiditis and Graves' disease, are the most common organ-specific autoimmune disorders and display one of the strongest sex biases in autoimmunity, with a marked female predominance. This review examines how biological sex and gender-related determinants shape AITD susceptibility, expression, and clinical heterogeneity, and their relevance to personalized care. PubMed/MEDLINE and Scopus were searched from inception to June 2026, with Web of Science used for forward citation tracking. Human studies, reviews and meta-analyses, and selecte

Limited early transcriptome-wide mucosal response to the bacterial pathogen Yersinia ruckeri in Atlantic salmon (Salmo salar) alevins.

Atlantic salmon larvae possess immune structures, effector cells, and molecules, and their mucosal organs serve as critical barriers against pathogens. But despite these defences, fish larvae show limited immune response capabilities, and the early transcriptomic responses of different mucosal surfaces remain poorly understood. Therefore, this study investigated the transcriptomic landscape of Atlantic salmon (Salmo salar) alevins exposed to the pathogen Yersinia ruckeri, analysing three mucosal organs (skin, gills, and tongue) over 72 h post-exposure. RNA-Seq analysis revealed distinct basal

Lacking P2X7-receptors protects substantia nigra dopaminergic neurons and hippocampal-related cognitive performance from the deleterious effects of high-fat diet exposure in adult male mice.

BACKGROUND: Dietary fat consumption, involved in the pathogenesis of insulin resistance and impaired glucose metabolism, is linked with decline in cognitive functions, dementia, and development of Parkinson's disease and Alzheimer's disease. Mature IL-1β, requiring the activation of the P2X7 receptor (P2X7R)-inflammasome complex, is an important mediator of neuroinflammation. The aim of the study was to test whether P2X7R activation might interfere with systemic and cerebral metabolic homeostasis. METHODS: We treated WT and P2X7R KO mice with a high-fat diet (HFD) for 16 weeks, evaluating the

Sex chromosome heteromorphism and the Fast-X effect in poeciliids.

Fast-X evolution has been observed in a range of heteromorphic sex chromosomes. However, it remains unclear how early in the process of sex chromosome differentiation the Fast-X effect becomes detectible. Recently, we uncovered an extreme variation in sex chromosome heteromorphism across poeciliid fish species. The common guppy, Poecilia reticulata, Endler's guppy, P. wingei, swamp guppy, P. picta and para guppy, P. parae, appear to share the same XY system and exhibit a remarkable range of heteromorphism. Species outside this group lack this sex chromosome system. We combined analyses of sequ

The gut microbiota reduces leptin sensitivity and the expression of the obesity-suppressing neuropeptides proglucagon (Gcg) and brain-derived neurotrophic factor (Bdnf) in the central nervous system.

The gut microbiota contributes to fat mass and the susceptibility to obesity. However, the underlying mechanisms are not completely understood. To investigate whether the gut microbiota affects hypothalamic and brainstem body fat-regulating circuits, we compared gene expression of food intake-regulating neuropeptides between germ-free and conventionally raised (CONV-R) mice. We found that CONV-R mice had decreased expression of the antiobesity neuropeptide glucagon-like peptide-1 (GLP-1) precursor proglucagon (Gcg) in the brainstem. Moreover, in both the hypothalamus and the brainstem, CONV-R

Neurogenic inflammation in periimplant and periodontal disease: A case-control split-mouth study.

OBJECTIVE: Although the regulatory effects of substance-P (SP), neurokinin-A (NKA), calcitonin gene-linked peptide (CGRP) and neuropeptide-Y (NPY) on periodontal inflammatory responses have been described, the effects of these neuropeptides on healthy and diseased periimplant tissues are not clearly defined. MATERIALS AND METHODS: Thirty-nine implants loaded at least for 12 months with their symmetrically matching teeth were evaluated and compared by a split-mouth study design. Six study groups were created in this regard as follows: group 1 (healthy periodontal tissues), group 2 (healthy per

Plac1 (placenta-specific 1) is widely expressed during fetal development and is associated with a lethal form of hydrocephalus.

BACKGROUND: Plac1 is an X-linked gene essential for normal placental development. Plac1 ablation is associated with placentomegaly and intrauterine growth retardation. Additionally, Plac1-null mice exhibit reduced postnatal viability. This study was conducted as part of our ongoing examination of the postnatal phenotype associated with Plac1 ablation. METHODS: Plac1 was deleted in murine embryonic stem cells and bred against a C57BL/6 background. Mutant mice were genotyped using a polymerase chain reaction-based strategy. Embryonic Plac1 expression was assessed by quantitative real-time polym

Unravelling the hsp70 gene in snubnose pompano (Trachinotus blochii): insights into stress response mechanisms.

Heat shock proteins (HSPs) are molecular chaperones that play a crucial role in fish physiology from stress-induced damage by stabilizing proteins and enhancing survival under environmental challenges. This study characterizes the stress-responsive hsp70 (tb-ihsp70) gene in Trachinotus blochii, a prime marine aquaculture species, from an evolutionary perspective and links its expression to salinity and pathogen stress. The tb-ihsp70 encodes a 639 amino acid protein with conserved ATPase and substrate-binding domains critical for protein conformation under stress. Phylogenetic analysis grouped

PRAMEY enhances sperm-egg binding and modulates epigenetic dynamics in bovine embryogenesis.

Infertility and subfertility are significant reproductive challenges in cattle, often linked to genetic factors. Among these genetic factors, the bovine Y-linked gene family, PRAMEY, has emerged as a candidate due to its involvement in germ cell formation, fertilization, and embryonic development. This study investigates PRAMEY's role in sperm-egg binding, acrosome integrity, and epigenetic modifications during fertilization and early embryogenesis. Using IVF with bovine spermatozoa treated with either PRAMEY antibody (ab) or rabbit IgG control, we assessed sperm-egg binding and acrosome integ

Deep phenotypic analysis of psychiatric features in genetically defined cohorts: application to XYY syndrome.

BACKGROUND: Recurrent gene dosage disorders impart substantial risk for psychopathology. Yet, understanding that risk is hampered by complex presentations that challenge classical diagnostic systems. Here, we present a suite of generalizable analytic approaches for parsing this clinical complexity, which we illustrate through application to XYY syndrome. METHOD: We gathered high-dimensional measures of psychopathology in 64 XYY individuals and 60 XY controls, plus additional interviewer-based diagnostic data in the XYY group. We provide the first comprehensive diagnostic description of psychi

Rivastigmine Reverses the Decrease in Synapsin and Memory Caused by Homocysteine: Is There Relation to Inflammation?

Elevated levels of homocysteine (Hcy) in the blood, called hyperhomocysteinemia (HHcy), is a prevalent risk factor for it has been shown that Hcy induces oxidative stress and increases microglial activation and neuroinflammation, as well as causes cognitive impairment, which have been linked to the neurodegenerative process. This study aimed to evaluate the effect of mild hyperhomocysteinemia with or without ibuprofen and rivastigmine treatments on the behavior and neurochemical parameters in male rats. The chronic mild HHcy model was chemically induced in Wistar rats by subcutaneous administr

Involvement of IGF2 and H19 imprinting in choriocarcinoma development.

Complete hydatidiform mole is an abnormal pregnancy characterized by grossly swollen villi in the absence of a fetus (Kajii and Ohama 1997, Wake et al., 1978, Jacobs et al., 1980). It is well known that this abnormal pregnancy product is androgenetic in origin. The entire genome of the molar conceptus is paternally derived. The majority of moles result from fertilization of an empty egg by haploid sperm. The paternally derived haploid set then duplicated without cytokinesis and restores diploidy. Whole-genome sequencing identifies a potentially causative homozygous splicing variant in RAD54 like in non-obstructive azoospermia.

BACKGROUND: Non-obstructive azoospermia (NOA) is a severe form of male infertility characterized by impaired spermatogenesis. Although chromosomal abnormalities and Y-chromosome microdeletions are known causes, many cases remain idiopathic, suggesting that other genetic factors may be involved. RAD54 like (RAD54L) is a key component of the homologous recombination pathway required for DNA double-strand break repair and meiotic progression. OBJECTIVE: To identify the potential genetic cause of NOA using whole-genome sequencing (WGS) in affected individuals from consanguineous families. MATERI
