Placental biology links genetic, epigenetic, ancestral, and social determinants to maternal-fetal health inequities.

Black and African American women in the United States experience disproportionately high rates of maternal and infant mortality and morbidity, including placenta-mediated complications such as preeclampsia, preterm birth, and miscarriage. These inequities arise from the interaction of social and environmental conditions with biological pathways that become embedded over the life course, with the placenta serving as a central mediator. In this review, we examine how placental biology integrates genetic variation, epigenetic regulation, paternal contributions, immune processes, and lived social

Genetic Determinants of Hematopoietic Toxicity Risk in Thai Pediatric Patients Undergoing 6-Mercaptopurine Treatment.

The nucleoside diphosphate-linked moiety X-type motif 15 (NUDT15) has been identified as a key genetic determinant of 6-mercaptopurine (6-MP)-induced hematopoietic toxicity in populations with a high frequency of NUDT15 variants but a low frequency of thiopurine S-methyltransferase (TPMT) variants. However, evidence remains limited in populations where both genetic variants are prevalent. This study aimed to characterize the impact of NUDT15 and TPMT polymorphisms on 6-MP-induced hematopoietic toxicity in Thai pediatric patients, a population with a high prevalence of both variants. A total of

Identification of Murine Rotavirus Virulence Determinants Using Bidirectional Selective Passaging and a Reverse Genetics System.

Live-attenuated rotavirus (RV) vaccines are the most effective interventions for preventing RV gastroenteritis (RVGE) in young children. However, the molecular basis of attenuation remains not well understood. Here, we describe a compact but comprehensive strategy to identify RV virulence determinants by combining low-passage bidirectional selection, sequence analysis, and segment-level phenotype testing via a reverse genetics infectious system. Using the virulent murine RV strain EW, virulence was quantified by diarrhea severity/duration and body-weight gain. Serial passaging in cell culture

Comparative functional genomics of Saccharomyces cerevisiae reveals genetic determinants of stress tolerance and ethanol fermentation.

Saccharomyces cerevisiae strains from diverse origins exhibit distinct phenotypic traits, providing valuable diversity and adaptability for industrial applications. Here, we conducted a comparative analysis of phenotypic and genomic features across 41 S. cerevisiae strains with clear industrial niche associations, aiming to identify genetic determinants underlying stress resistance and ethanol fermentation efficiency. These strains displayed niche-specific growth advantages under fermentation-related stress conditions, yet none showed broad tolerance. During ethanol fermentation using wheat an

Coagulation Pathways as Determinants of Acute Subdural Hematoma Resolution: Genetic Evidence From Human Data.

BACKGROUND: Acute subdural hematoma (ASDH) is a severe complication of traumatic brain injury, with high mortality and disability. Spontaneous hematoma resolution is an important determinant of functional recovery, but the biological mechanisms underlying this process remain poorly understood. Coagulopathy, common in ASDH, may influence hematoma dynamics, but its causal role remains uncertain. METHODS: We conducted a two-sample Mendelian randomization (MR) study to investigate the causal effects of coagulation traits on hematoma resolution. Genetic instruments for fibrinogen isoforms, coagula

Genetic and lifestyle determinants of lipid profiles in Japanese young adults: evidence from MBOAT7 rs641738.

BACKGROUND: Early prevention of dyslipidemia is critical for reducing the future onset of atherosclerosis and atherosclerotic cardiovascular diseases. Although lifestyle interventions have been recommended, the interactions between common genetic variants and modifiable habits in young populations remain unclear. Membrane-bound O-acyltransferase domain-containing 7 (MBOAT7) has been implicated in lipid metabolism and steatotic liver disease; however, its role in early atherosclerosis is poorly understood. Associations between the MBOAT7 rs641738 genotype, lifestyle habits, and atherogenic lipi

Sex determination and genetic screening of equine embryos with whole genome amplification and real-time PCR.

Reliable pre-implantation sex determination and genetic screening enables informed embryo transfer decisions in equine breeding while avoiding later interventions. We developed a streamlined workflow that couples rapid whole genome amplification (WGA) with a multiplex real-time PCR targeting ETSTY5 as a Y-specific marker and UBC as an autosomal control. On purified equine DNA, sex was correctly assigned down to 10 pg gDNA and to a single fibroblast cell. Direct testing of embryo biopsies without WGA yielded inconsistent results, whereas introducing a short WGA step produced tight allelic-discr

Phenome-wide screening of the putative causal determinants of bipolar affective disorder using genetic data.

Bipolar affective disorder (BPAD) is a chronic psychiatric condition with high heritability (60-85%) and significant phenotypic variability, complicating its etiological understanding. Genome-wide association studies (GWAS) have revealed genetic overlaps with diverse traits, suggesting pleiotropy. This study aimed to identify shared genetic factors and infer causal relationships using large-scale GWAS data.BPAD GWAS summary statistics were obtained from the UK Biobank via the Complex Trait Genetics Virtual Lab (CTG-VL) platform. Genetic correlations with 1,504 traits were estimated using linka

IL5 rs2069812 and IL13 rs1800925 Genetic variants as key determinants of clinically relevant asthma phenotypes.

Type 2-high airway inflammation in adults with asthma is heavily driven by the cytokines interleukin (IL)-4, IL-5, and IL-13. While genetic variations in these cytokines are known to influence asthma pathogenesis, their specific impacts on clinically relevant phenotypes remain to be fully elucidated. This study aimed to investigate the associations between inflammatory cytokine gene polymorphisms, clinical asthma phenotypes, inflammation cell subtypes, and lung function. A cross-sectional study was conducted involving 125 adults with asthma. Genotyping was performed for the following single-nu

Genetic background determines allele-specific Pfkelch13 -mediated artemisinin tolerance and persistence in Plasmodium falciparum.

Artemisinin-based combination therapies are the frontline treatment for Plasmodium falciparum malaria, but their efficacy is threatened by Artemisinin partial resistance (ART R ). ART R in Plasmodium falciparum is mediated by Pfkelch13 ( k13 ) mutations but remains rare in West Africa. A critical question is whether short-term drug survival intrinsically predicts long-term evolutionary persistence. Here, using CRISPR-Cas9 editing, and nanopore-assisted allele tracking in a contemporary Senegalese isolate, we evaluated ART R receptivity using the ring-stage survival assay (RSA), a proxy for cli

Phenotypic and genetic determinants of quinolone resistance in Escherichia coli at the human-swine interface of an agroecological farm.

Quinolone-resistant Escherichia coli are increasing in humans and livestock; however, their distribution and underlying mechanisms in shared environments remain incompletely characterized. We analyzed quinolone resistance in E. coli isolates from swine (n = 100) and humans (n = 65) associated with an agroecological farm in Morelos, Mexico. Antimicrobial susceptibility testing was performed for quinolones [nalidixic acid (NAL) and ciprofloxacin (CIP)], and the minimum inhibitory concentrations were determined. Phylogenetic grouping and genotypic analyses targeting gyrA mutations and plasmid-med

Investigating PXR (NR1I2) genetic polymorphisms as possible determinants of the magnitude of the clindamycin-rifampicin pharmacokinetic interaction.

PURPOSE: Rifampicin is known to induce clindamycin metabolism via CYP3A4/5, but the magnitude of this interaction varies greatly from one patient to another. Because this metabolic induction results from rifampicin activation of the nuclear pregnane X receptor (PXR), the genetic polymorphisms of its NR1I2 gene and that of CYP3A4/5 might predict the risk of clindamycin underdosage when combined with rifampicin. We therefore conducted a study to determine the possible influence of NR1I2 and CYP3A4/5 gene polymorphisms on baseline and rifampicin-induced clindamycin clearance. METHODS: Eighty-fiv

Genetic determinants of gene amplifications alter frequency and evolutionary trajectory of antibiotic resistance in Staphylococcus aureus.

Gene amplifications are thought to be common in bacterial populations, providing a rapid reversible mode of adaptation to diverse stresses, including the acquisition of antibiotic resistance. We previously showed that the opportunistic pathogen Staphylococcus aureus evolves resistance to the dual-targeting fluoroquinolone delafloxacin (DLX) that inhibits both the DNA gyrase and DNA topoisomerase IV via gene amplifications of an efflux pump encoding gene sdrM. However, the pathways that control the formation or selection of gene amplifications, and consequently adaptive trajectories, remain und
