Restoration of gamma center frequency via personalized entrainment marks cognitive preservation in early Alzheimer's disease.

Gamma entrainment shows promise for Alzheimer's disease (AD) treatment in preclinical models, but human trials have yielded heterogeneous results. We hypothesized that the clinical efficacy of gamma entrainment depends on individual neurophysiological receptivity, specifically the capacity for neural circuit plasticity. In this open-label pilot study, we screened 37 individuals and enrolled 16 participants with early AD (CDR 0.5-1.0, amyloid-positive) who completed 12 weeks of home-based flickering light stimulation at individually optimized gamma frequencies (32-40 Hz). Pre- and post-interven

Regulation of survival, growth, and metabolism by neuronal mTOR.

Reducing activity of the mechanistic/mammalian target of rapamycin (mTOR) with rapamycin extends lifespan and healthspan in many species. The mechanisms by which mTOR regulates lifespan and healthspan, however, are still unknown. Understanding how mTOR signaling in different cell types regulates lifespan and aspects of healthspan is urgently needed if we are to harness the potential individual and societal benefits of healthspan extension by mTOR attenuation. mTOR kinase can form two complexes, mTOR complex 1 (mTORC1) and mTOR complex 2 (mTORC2). The regulatory associated protein of mTOR (Rapt

Accelerated aging as a key mechanism linking diet quality to cardiovascular disease: a prospective mediation analysis in the UK Biobank.

We aimed to examine the mediating role of accelerated aging in the association between diet quality and CVD incidence and mortality. Data were obtained from the UK Biobank cohort. Diet quality was evaluated using four established indices: the Healthy Eating Index-2020, the Alternative Healthy Eating Index, the Dietary Approaches to Stop Hypertension index, and the Mediterranean Diet Score. Accelerated aging was assessed by the Phenotypic Age Acceleration and Biological Age Acceleration. Outcomes included incident CVD and CVD mortality. Multivariable linear regression model, Cox proportional ha

Genetic associations with longevity in a Calabrian cohort: an exploratory genome-wide study.

Human longevity is a complex trait shaped by genetic background and population-specific factors. Calabria, a region in Southern Italy with a high prevalence of centenarians and relative genetic isolation, is a valuable model for investigating the genetic architecture of extreme survival. Here, we performed a genome-wide association study of longevity in 705 Calabrian individuals, comparing long-lived subjects to younger controls using a mixed-model approach that accounts for relatedness and population structure. We identified 22 suggestive risk loci, among which one reached genome-wide signifi

The dual diversity crisis in alzheimer's disease research: why neuroimaging biomarkers and clinical trials keep failing.

Alzheimer's disease clinical trials have failed at a rate exceeding 99% over two decades, and neuroimaging biomarkers developed in discovery cohorts have repeatedly disappointed in validation and clinical application. Standard explanations address symptoms rather than structural causes. I argue that Alzheimer's disease research is caught in a Dual Diversity Crisis: the simultaneous neglect of population diversity and individual neurobiological heterogeneity as compounding validity threats. The first dimension concerns the systematic over-reliance on Western, young, healthy, and university-affi

Apolipoprotein(a)-rich electronegative HDL accelerates vascular aging and atherosclerosis by inducing endothelial senescence.

The limited benefits of high-density lipoprotein (HDL)-targeted therapies have shifted the research focus to HDL quality; some HDL subtypes may be pro-atherogenic and may counteract the potential benefits of HDL therapies. This study investigated the pathological mechanism of H5, a highly electronegative and compositionally abnormal HDL subfraction characterized by impaired cholesterol efflux. We used anion-exchange chromatography to separate HDL into five subfractions (H1-H5) from blood from 131 asymptomatic individuals (95 with metabolic syndrome and 36 without [controls]). Plasma H5 levels

Association between whole-blood NAD+ concentration and frailty in community-dwelling older adults: the Itabashi Longitudinal Study on Aging.

Nicotinamide adenine dinucleotide (NAD⁺) is central to energy metabolism, redox reactions, and cellular signaling. However, human evidence linking circulating NAD⁺ concentrations to frailty remains limited. We examined the association of whole-blood NAD⁺ concentration with frailty in community-dwelling older adults. This cross-sectional analysis included community-dwelling adults aged ≥ 65 years from the 2025 Itabashi Longitudinal Study on Aging. Frailty was assessed using the revised Japanese Cardiovascular Health Study Criteria. Multivariable linear and logistic regression models adjusted fo

Spatiotemporal profiling of white matter lesions and their contribution in the pathologies of Parkinson's disease animal models.

Increasing evidence has identified significant white matter lesions (WMLs) in Parkinson's disease (PD) patients. However, the complex relationships between WMLs and the neuropathological changes of PD remain unclear. In this study, we comprehensively elucidated the spatiotemporal dynamics of WMLs in rotenone-, 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP)-induced PD models, and α-synuclein (α-Syn) (A53T) transgenic mice. The results showed that WMLs occurred across multiple brain regions and gradually aggravated as PD models progressed. Notably, WMLs emerged as early pathological events

Two-wave status patterns of handgrip strength asymmetry and low handgrip strength in relation to all-cause mortality in middle-aged and older Chinese adults.

Handgrip strength asymmetry (HGSA) and low handgrip strength (LHGS) reflect distinct aspects of muscle health, but their changes over time have been less studied. We examined whether two-wave status patterns of HGSA and LHGS were associated with all-cause mortality in middle-aged and older Chinese adults. We analyzed 8924 participants aged ≥ 45 years from the China Health and Retirement Longitudinal Study. HGSA and LHGS were assessed in 2011 and 2013 and classified as normal at both waves, newly abnormal at Wave 2, normalized at Wave 2, or abnormal at both waves. The 2013 assessment served as

Low-dose tamoxifen ameliorates ovariectomy-induced metabolic and immune dysfunction.

The decline in estrogen following menopause is a major driver of metabolic and immune dysfunction in aging females. While hormone replacement therapy improves many of these outcomes, its clinical use remains limited due to concerns regarding estrogen-sensitive malignancies. Tissue-selective estrogen complexes (TSECs), which combine estrogens with selective estrogen receptor modulators (SERMs) such as tamoxifen (TAM), represent a promising strategy to preserve the metabolic and immunological benefits of estrogen while reducing oncogenic risk. However, the systemic effects of TAM under condition

Reduced sensorimotor lactate concentration and improved sleep efficiency following resistance training in older adults.

As populations age worldwide, pathological neurodegeneration with subsequent cognitive decline has emerged as a pressing public health challenge. As effective treatments remain limited once neurodegeneration has occurred, prevention and delay of decline onset are crucial. While physical exercise is associated with improved cognition, exercise-specific mechanisms remain unclear. We therefore examined the effects of 12 weeks explosive lower-body resistance training (RT) in community-dwelling older adults on proposed markers associated with brain metabolism and sleep. Thirty-six participants (64-

Impact of caloric restriction on biological aging: insights from a composite biomarker index in older adults.

Caloric restriction (CR) extends lifespan and delays age-related diseases in model organisms, yet its effects on biological aging in humans remain unclear. We pooled data from seven randomized CR trials (n = 829) and examined randomization to CR and change in weight with a biomarker index composed of CRP, IL-6, cystatin C, insulin, GDF-15, and TNF-R1. CR improved the composite biomarker index. The effect of CR decreased from -2.2 to -1.2 (95% CI -2.0 to -0.3) after adjustment for weight, while the effect of weight loss decreased from 0.22 to 0.16 (95% CI 0.09 to 0.23) when CR was included in t

Combating Alzheimer's through sleep and exercise (CASE): A study protocol.

Dementia affects over 55 million people worldwide, with Alzheimer's disease (AD) comprising most cases. Because curative therapies remain unavailable, there is an urgent need for prevention strategies that target modifiable risk factors. Behavioral interventions-particularly sleep improvement and physical activity-can independently benefit cognition, brain function, and cardiometabolic health, yet their combined (potentially synergistic) effects and scalable delivery models remain underexplored. This exploratory, hypothesis-generating, three-arm randomized controlled trial will evaluate: (1) w

Diet-dependent, beneficial and adverse effects of rapamycin on life span of Drosophila melanogaster.

Rapamycin supplementation has been reported to extend life spans in numerous species, including Drosophila melanogaster, but a recent study in this laboratory revealed a marked life-shortening effect in y w male flies. The experiments reported here were performed to characterize conditions under which rapamycin could extend or shorten life spans in five fly strains. On a cornmeal/torula yeast-based medium, rapamycin extended life in one experiment, had no effect in six and was harmful in 19. On a nutrient-rich brewer's yeast medium favored by 4/5 strains for egg-laying, rapamycin was beneficia

Structural and mitochondrial dendritic degenerations in old hypoglossal motor neurons.

Hypoglossal motor neurons (MNs) within the medullary hypoglossal nucleus innervate the striated muscles of the intrinsic and extrinsic tongue. Dysfunction of the control of the tongue muscles may lead to problems such as dysphagia, dysphonia and the increased risk of aspiration pneumonia in the elderly. In the human and Fischer 344 (F344) rat motor systems, age-related muscle weakness and behavioural dysfunctions are contemporaneous to MN death. In other neurons, dendritic and mitochondrial degenerations are fundamental pathophysiological components preceding neuronal death. We aimed to determ

Three-dimensional trace element profile reflecting aging, disease, and frailty.

Older age combined with chronic disease increases the risk of malnutrition and frailty, impacting disease recovery and overall clinical outcomes. Serum concentrations of several trace elements and their respective biomarkers have not yet been investigated with regard to frailty in older adults with disease, although these patients most likely have an altered trace element profile owing to both inflammatory conditions and inadequate dietary intake. This cross-sectional study investigated trace element profiles in relation to age, disease, and frailty status in geriatric patients (n = 198) as we

Motor speech markers of Alzheimer's risk.

Motor changes are increasingly recognized as part of the Alzheimer's disease (AD) phenotype, yet motor speech function and its clinical utility for early AD risk stratification remain underexplored. This cross-sectional study investigated whether measures of motor speech performance in home-based rapid syllable repetition (diadochokinesis [DDK]) tasks are associated with the established AD risk indicators of subjective cognitive decline (SCD), apolipoprotein E ε4 (APOE ε4) carriage, and plasma phosphorylated tau 181 (p-tau181). A two-step model of age-associated autoreactivity: B cell-intrinsic aging meets inflammaging.

Aging is associated with declining protective immunity and increased autoreactivity, yet the mechanisms linking these processes remain incompletely understood. B lymphocytes are central to this paradox because they integrate antigen recognition, innate signaling, and differentiation under tightly regulated tolerance checkpoints. This review advances the hypothesis that aging appears to drive a progressive, B cell-intrinsic relaxation of central and peripheral tolerance, leading to the accumulation of autoreactive clones. These defects are reinforced by transcriptional, epigenetic, metabolic, a

Association of biological age acceleration with muscle aging phenotype in young and middle-aged adults: a prospective cohort study.

Loss of muscle mass and quality (myopenia and myosteatosis) are hallmarks of aging and predictors of disability, yet their relationship with biological aging in younger adults is unclear. This prospective cohort study included 165,015 adults (mean age 39.9) who underwent low-dose chest computed tomography (CT) between 2010 and 2020. A longitudinal subset of 61,669 participants had repeated scans (median follow-up 4.1 years). Sex-specific circadian and behavioral phenotypes in aged growth hormone receptor knockout mice.

Growth hormone (GH) exerts complex effects on metabolism, cognition, and overall physiological well-being, and its secretion is highly regulated by the circadian clock. The Growth Hormone Receptor Knockout (GHR-KO) mouse, a model of GH insensitivity, characterized by chronically elevated circulating GH, provides a unique opportunity to examine the consequences of disrupted GH signaling during aging. Given the close relationship between circadian rhythmicity and GH secretion, we investigated sex- and genotype-specific differences in circadian activity, neuromuscular, and neurocognitive performa
