Epidermal Langerhans cells (LC) belong to the dendritic cell family and represent the major APC within the skin. LC capture epicutaneous Ag, migrate into regional lymph nodes, and present Ag to T cells, thereby initiating primary immune response. Guniea-pigs were sensitized by percutaneous application of dinitrochlorobenzene and exposed to UVB (280-320 nm) radiation. The exposure to radiation diminished the response to an elicitation dose of the hapten administered 14 days later within the site of irradiation. The exposure dose of radiation required to produce this effect resulted in a marked erythemal response, but this response did not conceal the contact allergic reaction. The site of elicitation of the allergic response had to be included in the exposure field.

The ability of closely related platinum group metal salts (PGMS) to cross-react with the principal sensitising agent ammonium hexachloroplatinate IV was investigated in refinery workers. Selected subjects were screened by skin prick test, specific RAST, RAST inhibition, and primate PCA tests. Skin tests of delayed hypersensitivity were performed on thirty-eight patients of leprosy with a bacterial antigen (tuberculin), a hapten (dinitrochlorobenzene) and allogenic lymphocytic transplantation. VP 16-213 (etoposide, abbr. to VP), an oncostatic drug, and VP-protein conjugates were examined for their antigenic properties using male Hartley guinea pigs. The results of systemic anaphylaxis test, passive cutaneous anaphylaxis test, passive hemagglutination test and Schultz-Dale reaction revealed that VP possessed neither antigenic nor haptenic properties. The dose levels of VP employed in the present experiment were confirmed not to suppress immune reactions.

Cardiolipin, the primary lipid hapten in the antigen suspension used for the detection of antitreponemal antibodies in the sera of syphilitic patients, was successfully coupled to glucose oxidase, peroxidase, and some other enzymes using different crosslinking agents. These complexes were used to replace the pure uncomplexed cardiolipin for the preparation of the antigen suspension. When these suspensions were allowed to react with serum that contained anticardiolipin antibodies the activity of the enzyme was inhibited. In the absence of antibody, no enzyme inhibition was observed.

Repeated exposure to the marine bryozoan, Alcyonidium gelatinosum, frequently provokes an eczematous allergic contact dermatitis known as the "Dogger Bank Itch". The dermatitis, representing a severe occupational disease, is especially widely distributed among trawlermen working in the Dogger Bank area in the North Sea. The allergy is shown to belong to the type of cell-mediated hypersensitivity. The hapten has been identified as the (2-hydroxyethyl)dimethylsulfoxonium ion. The isolation, structure determination and synthesis are discussed.

We describe the use of gas chromatography with mass spectrometry (GC-MS) in the investigation of a case of allergic contact dermatitis of the lip margins caused by a lip liner. Patch testing identified sensitization to para-tertiary-butylphenol-formaldehyde resin (PTBP-FR), a resin glue frequently used in leather work but rarely found in cosmetics. Investigation specified the para-tertiary-butylphenol (PTBP) component of the resin as the hapten responsible for the allergic contact dermatitis, as well as for associated depigmentation.

We report here the use of an alternative carrier diphtheria toxoid (DT) in human subjects to overcome antigen-specific unresponsiveness upon immunization with a hapten/ligand-carrier conjugate. In the phase I clinical trial of a birth control vaccine using gonadotrophin subunits linked to tetanus toxoid, some of the subjects failed to evoke a booster antibody response to human chorionic gonadotrophin (hCG). Presentation of the ligand on DT instead in subsequent immunizations restored anti-hCG response.
