Oral DNA vaccines based on CS-TPP nanoparticles and alginate microparticles confer high protection against infectious pancreatic necrosis virus (IPNV) infection in trout.

Infectious pancreatic necrosis virus (IPNV) is the etiological agent of a contagious viral disease causing remarkable mortalities in different fish species. Despite the availability of commercial vaccines against IPN, the disease still constitutes one of the main threats to the aquaculture industry worldwide. In this study, we developed a DNA vaccine encoding the VP2 gene of IPNV and evaluated its ability to induce protective immunity in rainbow trout fry (3 g) at doses of 10 and 25 μg/fish and boosting with the same doses two weeks later through the oral route using chitosan/tripolyphosphate

Chitosan-based nanoparticles as a hepatitis B antigen delivery system.

The design of antigen delivery systems, particularly for mucosal surfaces, has been a focus of interest in recent years. In this chapter, we describe the preparation of chitosan-based particles as promising antigen delivery systems for mucosal surfaces already tested by our group with hepatitis B surface antigen. The final proof of the concept is always carried out with immunization studies performed in an appropriate animal model. However, before these important studies, it is advisable that the delivery system should be submitted to a variety of in vitro tests. Among several tests, the chara

Antimicrobial and anti-inflammatory activity of chitosan-alginate nanoparticles: a targeted therapy for cutaneous pathogens.

Advances in nanotechnology have demonstrated potential application of nanoparticles (NPs) for effective and targeted drug delivery. Here we investigated the antimicrobial and immunological properties and the feasibility of using NPs to deliver antimicrobial agents to treat a cutaneous pathogen. NPs synthesized with chitosan and alginate demonstrated a direct antimicrobial activity in vitro against Propionibacterium acnes, the bacterium linked to the pathogenesis of acne. By electron microscopy (EM) imaging, chitosan-alginate NPs were found to induce the disruption of the P. acnes cell membrane

Encapsulated mesenchymal stromal cells for in vivo transplantation.

Immunomodulatory human mesenchymal stromal cells (hMSC) have been incorporated into therapeutic protocols to treat secondary inflammatory responses post-spinal cord injury (SCI) in animal models. However, limitations with direct hMSC implantation approaches may prevent effective translation for therapeutic development of hMSC infusion into post-SCI treatment protocols. To circumvent these limitations, we investigated the efficacy of alginate microencapsulation in developing an implantable vehicle for hMSC delivery. Viability and secretory function were maintained within the encapsulated hMSC p

Construction of growth plate organoids via a layered induction based in vitro 3D cultivation system.

Serious injury to the growth plate often leads to bony bridge formation, resulting in halted long bone growth, angular deformities, and limb length discrepancies. These problems persist unaddressed in the clinic. In this study, we engineered a four-layered growth plate organoid by integrating three-dimensional culture with layer-specific induction techniques. A gelatin/alginate hydrogel scaffold was utilized to recapitulate the architecture of the native growth plate. In the three cartilage zones, bone marrow derived mesenchymal stem cells (BMSCs) and chondrocytes were co-cultured at a 3:1 rat

The effects of β-D-mannuronic acid (M2000), as a novel NSAID, on COX1 and COX2 activities and gene expression in ankylosing spondylitis patients and the murine monocyte/macrophage, J774 cell line.

Ankylosing spondylitis (AS) is a debilitating chronic inflammatory disease with genetic predisposition, which is characterized by the involvement of spine and sacroiliac joints. Due to the relatively unsuccessful treatments, we designed β-D-mannuronic (M2000) with the beneficial effects in various experimental models as a novel non-steroidal anti-inflammatory drug (NSAID). The aims of our present study were: first, to compare the therapeutic effects of M2000, as a novel designed NSAID, with naproxen and placebo in Iranian patients with AS during 12 weeks; second, to evaluate the effect of M200

Core-shell hydrogel beads co-encapsulating kunitz-type trypsin inhibitor: A synergistic strategy for enhanced stability of oral active peptides.

This study repurposed Kunitz-type trypsin inhibitor (KTI) as a synergistic component for oral peptide co-delivery to overcome the burst release and enzymatic degradation of liraglutide. A pH-responsive chitosan (CS)‑sodium alginate (ALG) core-shell hydrogel co-delivery system co-encapsulating KTI and liraglutide, named ACKL (ALG-CS-KTI-liraglutide), was accordingly constructed. The optimized microspheres, with a 3% sodium tripolyphosphate (TPP)-crosslinked core and 0.8% ALG shell, exhibited uniform morphology, a distinct core-shell structure, and excellent mechanical properties. Effects of plant food potassium salts (citrate, galacturonate or tartrate) on acid-base status and digestive fermentations in rats.

Potassium (K) organic anion salts, such as potassium citrate or potassium malate in plant foods, may counteract low-grade metabolic acidosis induced by western diets, but little is known about the effect of other minor plant anions. Effects of K salts (chloride, citrate, galacturonate or tartrate) were thus studied on the mineral balance and digestive fermentations in groups of 6-week-old rats adapted to an acidogenic/5 % inulin diet. In all diet groups, substantial amounts of lactate and succinate were present in the caecum, besides SCFA. SCFA were poorly affected by K salts conditions. The K

Ketoprofen-eluting biodegradable ureteral stents by CO2 impregnation: In vitro study.

Ureteral stents are indispensable tools in urologic practice. The main complications associated with ureteral stents are dislocation, infection, pain and encrustation. Biodegradable ureteral stents are one of the most attractive designs with the potential to eliminate several complications associated with the stenting procedure. In this work we hypothesize the impregnation of ketoprofen, by CO2-impregnation in a patented biodegradable ureteral stent previously developed in our group. The biodegradable ureteral stents with each formulation: alginate-based, gellan gum-based were impregnated with

Nanoemulsions coated with alginate/chitosan as oral insulin delivery systems: preparation, characterization, and hypoglycemic effect in rats.

This study aimed to prepare nanoemulsions coated with alginate/chitosan for oral insulin delivery. Uncoated nanoemulsions were prepared by homogenization of a water in oil in water (w/o/w) multiple emulsion that was composed of Labrafac(®) CC, phospholipid, Span™ 80 and Cremorphor(®) EL. Coating of the nanoemulsions was achieved based on polyelectrolyte cross-linking, with sequential addition of calcium chloride and chitosan to the bulk nanoemulsion dispersion that contained alginate. The particle size of the coated nanoemulsions was about 488 nm and the insulin entrapment ratio was 47.3%. Cir

Chitotriosidase and gene therapy for fungal infections.

Chitotriosidase secreted by activated human macrophages has been implicated in the defence against chitin-bearing pathogens. The antifungal properties of human chitotriosidase were investigated here following retroviral vector-mediated gene transfer of the open reading frame of the chitotriosidase gene into Chinese hamster ovary cells. A chitinase assay confirmed that the engineered cells secreted recombinant chitotriosidase constitutively. Two dimensional gel electrophoresis and western blotting indicated that the recombinant protein is the major, chitin-binding, fifty kilodalton isoform. Cul

3D-printed bioactive scaffolds with alginate hydrogel and stromal vascular fraction differentiated into osteoblasts applied to bone repair.

Bone injuries present enormous medical challenges worldwide. Most scaffolds used in tissue engineering do not possess an appropriate combination of properties inherent to bone tissue, such as elasticity, rigidity, biocompatibility, osteoinductivity, and antimicrobial properties. In this study, we constructed a bioprinted scaffold using polycaprolactone (PCL), polyethylene glycol (PEG), and alginate cellular carrier hydrogels. This scaffold is associated with the stromal vascular fraction (SVF), a heterogeneous cell population rich in adipose-derived stem cells and contains osteogenic different

Antimicrobial dressings for the prevention of catheter-related infections in newborn infants with central venous catheters.

BACKGROUND: Central venous catheters (CVCs) provide secured venous access in neonates. Antimicrobial dressings applied over the CVC sites have been proposed to reduce catheter-related blood stream infection (CRBSI) by decreasing colonisation. However, there may be concerns on the local and systemic adverse effects of these dressings in neonates. OBJECTIVES: We assessed the effectiveness and safety of antimicrobial (antiseptic or antibiotic) dressings in reducing CVC-related infections in newborn infants. Had there been relevant data, we would have evaluated the effects of antimicrobial dressi

Addition of fibronectin to alginate matrix improves peripheral nerve regeneration in tissue-engineered conduits.

Schwann cell (SC) transplantation has been proposed to encourage peripheral nerve regeneration, but an optimal SC-carrying matrix would be needed. The aim of this study was to characterize how the addition of fibronectin to alginate would affect the outcome of nerve regeneration promoted by Schwann cells embedded in this matrix. Genetically labeled rat SCs were obtained by lacZ gene transduction. SCs were suspended in alginate hydrogel matrix with/without addition of liquid fibronectin, and their viability and growth in the different types of matrices were assessed in vitro by AlamarBlue assay

Biodegradation of diclofenac and paracetamol using immobilized laccase on novel polyacrylamide alginate gel chips and chitosan beads for mitigation of micropollutants in hospital effluent.

Excessive use of pharmaceuticals, especially antibiotics and non-steroidal anti-inflammatory drugs (NSAIDs) like diclofenac (DCF) and paracetamol (PCM), has led to increased levels in water sources. This study examined the production of laccase from the white-rot fungus (WRF) Megasporoporia minor, known for its high laccase output, for biodegrading DCF and PCM using innovative polyacrylamide gel chips or cryogel (PAG) and chitosan beads. These materials were characterized through fourier transform infrared spectroscopy (FTIR) and scanning electron microscopy (SEM). Results from immobilization
