In Situ CO2-Induced Hydrogels for Enhancing Carbon Sequestration in Saline Aquifers.

Carbon sequestration in saline aquifers underpins global net‑zero emission goals, yet existing CO2 storage capacity remains inadequate, and conventional approaches exhibit limited adaptability to reservoir environment. Here, we introduce smart, injectable soft materials based on CO2‑triggered hydrogels integrated with tailored deep eutectic solvents (DESs) to boost sequestration. The hydrogel forms upon CO2‑triggered protonation of erucamine (a C22‑tailed tertiary amine) within its aqueous dispersion, which promotes electrostatic interactions and hydrophobic self‑assembly. The DESs (choline ch…

Thermosensitive Liposome-Loaded Hydrogels Enable Mechano-Chemical Cascade Therapy for Promoting Angiogenesis.

Pharmacological activation of selective biophysical or chemical signaling pathways is known to regulate angiogenesis; however, whether these cues can be modulated in a temporally coordinated, cascade-like manner to generate stable and functional vascular networks remains unclear, particularly in the ischemic microenvironment. Here, we demonstrate a mechanochemical cascade in which TRPV4 (transient receptor potential vanilloid 4)-mediated mechanosensing initiates endothelial activation, followed by platelet-derived growth factor-BB (PDGF-BB) signaling that drives vascular formation and maturati

Multifunctional PNIPAM Hydrogels Based on Supercritical CO2-Assisted PEDOT:PSS/SWCNT Hybrid Structure.

We report herein the typical conductive polymer poly(3,4-ethylenedioxythiophene):polystyrenesulfonate (PEDOT:PSS) to be successfully modified onto single-walled carbon nanotubes (SWCNTs) using a supercritical carbon dioxide (SC CO2) antisolvent-induced polymer epitaxy method, and the role of the experimental temperature of SC CO2 on the modification process was systematically investigated. PEDOT:PSS/SWCNTs composites with optimal modification prepared at 80 °C and 15 MPa were used as functional fillers, and functional temperature-sensitive composite hydrogels of PEDOT:PSS/SWCNTs/poly-N-isoprop…

Injectable pH- and Temperature-Responsive Hydrogels for Scaffold Applications in Tissue Engineering.

Injectable hydrogels offer promising alternatives for scaffold-based tissue engineering due to their minimally invasive delivery and in situ forming capability. In this study, we reported the first development of an injectable hydrogel scaffold combining carboxymethyl cellulose (CMC), poly(ethylene glycol) (PEG), and poly(ε-caprolactone) (PCL) into a single system. This novel approach integrated the biocompatibility of CMC, tunable responsiveness of PEG, and mechanical robustness/degradability of PCL, which had not been previously reported. A pH- and temperature-responsive carboxymethyl cellul…

Storage and Stability of AAV-Containing Fibrin Hydrogels for Retinal Gene Therapy.

Subretinal and intravitreal injection of retinal gene therapy is associated with serious adverse events and poor efficacy. We sought to improve retinal gene therapy delivery by developing fibrin hydrogel encapsulated adeno-associated virus (FE-AAV). Here we investigate conditions for storage and stability for FE-AAV. FE-AAV containing 1.9 × 109 genome copies of AAV2/2-CMV-GFP was manufactured using fibrinogen reconstituted in 0.01 M sodium citrate, pH 7 (NaC) or phosphate-buffered saline containing 0.001% (v/v) Pluronic F68 (F68). Samples were stored at either -80 °C or 4 °C for up to 16 weeks

Photodegradable Hydrogels for On-Demand Modeling of Age-Related Spatiotemporal ECM Deformation.

The extracellular matrix (ECM) plays a crucial role in regulating tissue behavior through a dynamic interplay of spatial and temporal cues. Dynamic materials capable of modulating these cues at relevant scales are essential for tackling current challenges in tissue engineering and addressing fundamental biological questions. In vision research, there is a notable lack of suitable in vitro systems to study ECM dynamics. To help fill this gap, we developed an easy-to-use, photosensitive poly(ethylene glycol)-based hydrogel that can deform on demand to simulate ECM bulging, known as drusen, assoc

Bioactive injectable sulfated alginate-based hydrogels containing desferrioxamine for diabetic wound healing.

Diabetic wounds are prone to bacterial infection and difficult to heal due to chronic inflammation and hyperglycemic microenvironment. Here we developed a novel bioactive hydrogel with multifunctional tunable properties, including biocompatibility, pH-response, anti-inflammatory activity, and the ability to promote diabetic wound healing. The injectable and self-healing hydrogel was prepared by cross-linking sulfated oxidized sodium alginate (S-OSA) with adipic dihydrazide (ADH) and subsequently loading with the angiogenic drug desferrioxamine (DFO). Under acidic microenvironment in diabetic w

The Intersecting Physical Mechanisms That Regulate Cell Viability in 3D Synthetic Hydrogels.

Hydrogels restrict protein transport to different extents, with nanoporous synthetic polymer networks providing far less protein permeability compared to microporous biopolymer networks. To evaluate whether reduced permeability was a driving factor in reduced cell viability in synthetic hydrogels, we compared poly(ethylene glycol) vinyl sulfone (PEG-VS) hydrogels with Matrigel to quantify the influences of modulus, transport, and confinement on encapsulated cells. We observed extensive reductions in cell viability when encapsulated in PEG-VS gels compared to Matrigel. In transwell experiments …

Biomineralization-Inspired Hydrogels Enable Thermodynamically Enhanced CO2 Hydration for Efficient CO2 Sequestration.

The massive emissions of carbon dioxide (CO2) have triggered serious ecological crises, presenting severe challenges for CO2 sequestration. Mineral carbon sequestration is a promising approach for the integration of carbon capture, utilization, and storage (CCUS). However, its reaction thermodynamic process is universally sluggish, and its in situ crystallization regulation strategies are insufficient. To address the above limitations, inspired by the rapid biomineralization of eggshells catalyzed by carbonic anhydrase (CA), we innovatively introduced amino-functionalized zeolitic imidazolate

Hierarchical stabilization of bioactive hydrogels by multi-arm peptide-polymer supramolecular staples.

Supramolecular peptide hydrogels offer attractive bioactivity and dynamic mechanical behavior for three-dimensional cell culture and tissue engineering. However, their broader use is often limited by slow gelation and insufficient mechanical stability. Here, we introduce a molecular design strategy in which a tryptophan zipper pendant multiarm poly (ethylene glycol) (Trpzip-PEG) conjugate is incorporated into Trpzip nanofibrillar hydrogels to facilitate hierarchical tuning of materials properties. Trpzip peptides self-assemble into entangled nanofiber networks, while the addition of Trpzip-PEG…

Formulation and in-vitro characterization of okra mucilage-based controlled release hydrogels.

BACKGROUND: Certain drugs at therapeutic doses can cause gastrointestinal toxicity, which controlled-release formulations help to address. Natural polymers are widely used in drug delivery due to their non-toxic, biodegradable, and biocompatible properties. Okra mucilage powder (OMP) is especially promising because of its strong gelling ability and pH-sensitive behavior. OBJECTIVES: This study aimed to develop OMP-based hydrogels for the controlled release of methotrexate (MTX) over extended periods via free-radical polymerization. METHODS: Acrylic acid (AA) was used as the monomer with OMP …

Stiffness-engineered hydrogels drive neural regeneration via the Man2b1- lysosomal mechanotransduction axis.

Peripheral nerve repair requires biomaterials capable of dynamically guiding neural cells. While stiffness-tunable hydrogels hold promise, how their mechanical properties translated into pro-regenerative intracellular signals remains poorly understood. To address gap, we developed a library of polyacrylamide/chitosan hydrogels with tunable stiffness (1.5∼39 kPa) to mimic the native neural microenvironment. Using this platform, we identified an optimal stiffness (∼13.75 kPa) that maximizes PC12 cell adhesion, spreading, neurite outgrowth, and migration. When fabricated into nerve guidance condu…

Bioinspired Keratin-Based Eutectic Hydrogels for Intelligent Health Monitoring and Photothermal Therapy.

The development of epidermal sensors that integrate high mechanical strength, excellent sensing performance, self-healing capability, and monitoring and therapeutic functions into conductive hydrogels remains a major challenge. In response to this challenge, a PDA@Ag/PAA/PSBMA/FK eutectic hydrogel was synthesized via a facile one-pot method. The resulting hydrogel exhibited remarkable stretchability (1900%), high ionic conductivity (0.17 S·m-1), excellent low-temperature tolerance, and autonomous self-healing properties. The fabricated sensor exhibited a wide sensing range (2%-1000%) and can e…

Dynamic hydrogels for bone tissue engineering: modulating the fate of resident MSCs.

Different tissues exhibit distinct mechanical properties, including stiffness and viscoelasticity. Both static and dynamic mechanical cues modulate cellular behaviour and induce phenotypic changes. To leverage this regulatory mechanism, researchers have engineered dynamic hydrogels through physical interactions and dynamic covalent bonds (DCBs). Prior studies demonstrate that viscoelastic hydrogels effectively direct mesenchymal stem cell (MSC) behaviour, making them promising candidates for bone tissue engineering (BTE). This review systematically summarizes (i) dynamic hydrogel crosslinking …

Biomimetic Gradient Hydrogels Regulate Osteochondral Regeneration Microenvironment Remodeling via Spatiotemporal Programming Engineering.

Osteochondral repair remains a critical challenge owing to conventional scaffolds' inadequate dynamic mechanical adaptability, poor interfacial integration, inefficient cell recruitment, and lack of spatiotemporal regulation of bioactive cues. Inspired by native tissue hierarchical gradients and endogenous healing mechanisms, we developed an electric field-driven continuous gradient hydrogel (GHZF4) via spatiotemporal programming. Integrating nanofiber-reinforced self-adaptive matrix, electric field-induced nanofiber alignment, and ZIF-8 nanocarrier-mediated bioactive release, GHZF4 constructs
