[Evaluation of the activity of the inflammatory process in ankylosing spondyloarthritis].

A total of 139 patients with ankylosing spondyloarthritis (AS) (65 with the central form of the disease and 74 with the peripheral one) were examined to clarify the signs corresponding to the activity of the inflammatory process. To determine the degree of the inflammatory process activity in AS irrespective of its clinical forms the same laboratory indices can be used. With the central form of the disease some indices directly correlate with the markedness of pain and constraint in the spine. The activity of the process in this variant clearly reflects the level of circulating immune complexe

A quantitative approach to the determination of antigen in immune complexes.

Immune complexes are present in the circulation of healthy individuals and the formation of such complexes is part of a normal immune process. During some pathological conditions, significant amounts of immune complexes are formed and deposited in the kidney and other tissues, causing severe injury. Since the levels of immune complexes can provide valuable prognostic information, dozens of methods have been developed to detect and quantify these complexes. However, many of these methods are non-specific, not quantitative, and give false-positive results. Methods based on detecting the antigen

Erythema elevatum diutinum in a patient with Crohn's disease.

A possible association between erythema elevatum diutinum and Crohn's disease is described. To our knowledge, erythema elevation diutinum has never been reported in a patient with Crohn's disease, nor has the disorder been considered one of the cutaneous manifestations of inflammatory bowel disease. Erythema elevatum diutinum is believed to be mediated by the deposition of circulating immune complexes in the dermal perivascular spaces. Our patient showed clinical and histopathologic findings typical of both erythema elevatum diutinum and Crohn's disease. The patient also had a positive Raji ce

Interferon lambda in anti-viral defense and cancer: dual roles, mechanism and therapeutic potential.

BACKGROUND: The type III interferon family, known as interferon lambda (IFN-λ), consists of four isoforms (IFN-λ1, IFN-λ2, IFN-λ3, and IFN-λ4) and plays essential roles in immune responses. It mediates anti-viral and anti-tumoral activities and is distinguished from type I and type II interferons (IFNs) by its interaction with a unique receptor complex involving IFN-λR1 and IL-10 Rβ predominantly expressed on epithelial cells and selected immune cell populations. This interaction results in activation of the JAK-STAT signaling pathway and transcription of interferon-stimulated genes (ISGs). M

Immune-mediated disease caused by climate change-associated environmental hazards: mitigation and adaptation.

Global warming and climate change have increased the pollen burden and the frequency and intensity of wildfires, sand and dust storms, thunderstorms, and heatwaves - with concomitant increases in air pollution, heat stress, and flooding. These environmental stressors alter the human exposome and trigger complex immune responses. In parallel, pollutants, allergens, and other environmental factors increase the risks of skin and mucosal barrier disruption and microbial dysbiosis, while a loss of biodiversity and reduced exposure to microbial diversity impairs tolerogenic immune development. The r

Pulmonary and extrapulmonary sarcoidosis in relation to circulating immune complexes: a quantification of immune complexes by two radioimmunoassays.

Serum specimens from 53 patients with pulmonary sarcoidosis were examined for the presence of immune complexes by 2 methods, the Raji cell and the monoclonal rheumatoid factor radioimmunoassays. We found increased concentrations of immune complexes in the sera of 27 patients by one or both techniques. A significant association was found between increased concentrations of immune complexes and stage III sarcoidosis. Seventeen of 23 patients with stage III sarcoidosis and 10 of 50 with stage I or II disease had increased concentrations of immune complexes. Eight of the 10 patients with stage I o

Circulating immune complexes in carcinoma of the pancreas.

Circulating immune complexes were detected by the Clq binding assay in 45% of patients with pancreatic carcinoma. 75% of patients with primary biliary cirrhosis and 60% of patients with sclerosing cholangitis. The immune complexes present in most sera from these three groups were not reactive in the Raji cell radioimmune assay or monoclonal rheumatoid factor radioimmunoassay for soluble immune complexes. Thus, it was not possible to differentiate among these diseases on the basis of circulating immune complexes.

Lessons learned from clinical trials of thymic stromal lymphopoietin (TSLP) inhibition.

Type 2 (T2) immune-mediated epithelial-driven diseases are characterized by dysregulated immune responses at epithelial barrier surfaces. A key mediator of these diseases is thymic stromal lymphopoietin (TSLP), an epithelial cytokine that acts as a regulator of both T2 and non-T2 inflammation. TSLP activates dendritic cells and other immune cells, promoting the release of proinflammatory cytokines, including interleukin (IL)-4, IL-5, and IL-13. Given its central role in initiating and amplifying T2 inflammation, TSLP has emerged as a promising therapeutic target in multiple epithelial-driven i

Mucosal-Associated Invariant T Cells in Health and Disease.

Mucosal-associated invariant T (MAIT) cells are a population of evolutionarily conserved, unconventional innate-like T lymphocytes that are restricted by major histocompatibility complex Class I-related (MR1) molecules. They can be activated either in a T-cell receptor-MR1- or cytokine-dependent manner. Activated MAIT cells play crucial roles in maintaining homeostasis and the fine regulation of the immune system. This review presents a systematic exploration of the development, biological characteristics, and multiple effects of MAIT cells in health and various diseases. In healthy individual

High throughput screening identifies repurposable drugs for modulation of innate and acquired immune responses.

A balanced immune system is essential to maintain adequate host defense and effective self-tolerance. While an immune system that fails to generate appropriate response will permit infections to develop, uncontrolled activation may lead to autoinflammatory or autoimmune diseases. To identify drug candidates capable of modulating immune cell functions, we screened 1200 small molecules from the Prestwick Chemical Library for their property to inhibit innate or adaptive immune responses. Our studies focused specifically on drug interactions with T cells, B cells, and polymorphonuclear leukocytes

Environmentally driven immune imprinting protects against allergy.

Allergic diseases are caused by overexuberant type II immune responses mounted against environmental antigens1. The allergic state is typified by the presence of allergen-reactive immunoglobulin E (IgE), which triggers mast cell degranulation upon allergen encounter, manifesting in pruritis, oedema and, in severe cases, anaphylaxis. Over the past century, the prevalence of allergic diseases has increased markedly, suggesting that environmental rather than genetic factors are mediating this change2. Although many hypotheses connecting environment to allergy exist3-6, the biological mechanisms t

Mast cells in autoimmune disease: Unveiling their multifaceted roles and therapeutic potential.

Mast cells play a crucial role in developing autoimmune diseases by influencing immune responses, contributing to inflammation, and disrupting immune tolerance. This review examines the involvement of mast cells in various autoimmune disorders, including multiple sclerosis (MS), rheumatoid arthritis (RA), systemic lupus erythematosus (SLE), and type 1 diabetes mellitus (T1DM). The mechanisms through which mast cells mediate immune dysregulation are explored, focusing on their release of cytokines, role in antigen presentation, and interactions with other immune cells. Additionally, we discuss

Allergic diseases in the lungs.

Four basic immune reactions in the lung are fundamental to our understanding of allergic lung diseases. The first, immediate hypersensitivity, principally involves airway obstruction because of hypersecretion and bronchospasm. Resulting roentgenographic changes are those of large and small airway disease. The second reaction, cytotoxic antibody-mediated disease, produces air space filling by hemorrhage. The third form, immune complex-mediated hypersensitivity, causes mixed patchy consolidation and interstitial roentgenographic patterns caused by inflammation stimulated by antigen-antibody comp

Light quantity and photosystem function mediate host susceptibility to Turnip mosaic virus via a salicylic acid-independent mechanism.

Evidence going as far back as the early part of the 20th century suggests that both light and chloroplast function may play key roles in host susceptibility to viruses. Despite the long history of such work, confirmation of these phenomena and a determination of the underlying mechanisms remain elusive. Here, we revisited these questions using modern imaging technologies to study the susceptibility of Nicotiana benthamiana to Turnip mosaic virus (TuMV). We found that both light deficiency and photosystem impairment increased the susceptibility of N. benthamiana to TuMV infection. Time-lapse ph

Advancements in Understanding the Role of Oxylipins in Liver Injury and Liver Failure.

End-stage liver disease (ESLD) is characterized by a dramatic deterioration of liver function, frequently accompanied by systemic inflammatory storms and multiple organ failures. Central to the onset and progression of ESLD, systemic inflammation arises from complex interactions among various inflammatory signaling molecules and immune cells within and beyond the liver. As key inflammatory modulatory molecules, bioactive oxylipins have been increasingly recognized for their complex molecular mechanisms implicated in various diseases. This review aims to summarize recent findings regarding the

[Detection of circulating immune complexes using a laser-nephelometric assay (author's transl)].

Using Laser nephelometric measurement a method is described to detect immune complexes, which seems to be suitable for routine studies as well as for specific complex characterisation. Tetanus-Antitetanus complexes are used as standard thus the result is expressed as tetanus complex units (TCU). The method is simple and permits the direct measurement of complexes in native sera. Furthermore it is possible to characterise the complexes either by the immuno-globuline class of the antibody involved or the antigen respectively. Complexes were found with this method in various frequency in all but

The role of emotional distress in disfiguring skin diseases: A view from brain-skin axis.

Disfiguring skin diseases refer to skin diseases that significantly affect one's appearance. The development of disfiguring skin diseases is multifactorial, such as genetic, immune, and endocrine factors. However, recent research suggests that emotional distress (such as anxiety, depression, and sleep disorders) has been implicated as a key contributing factor to the onset and progression of disfiguring skin diseases, including acne, rosacea, and vitiligo; in turn, the psychosocial burden imposed by these conditions exacerbates emotional disturbances, thereby creating a self-perpetuating vicio

Proteomics focusing on immune markers in psoriatic arthritis.

The immune-pathogenesis of psoriatic arthritis represents a subject of intense research, as a still unknown factor can trigger the chronic inflammation that, upon a defective immune terrain, generates this auto-immune/auto-inflammatory condition. The pathogenesis complexity of psoriatic arthritis resides in the psoriatic synovitis milieu, where intricate immune relations are emerging during disease development. Innate immune response generates inflammatory cytokines driving effectors functions for immune and non-immune cells that sustain the chronical character of the synovitis. Herein, we rev

Immune complex glomerulonephritis in baboons (Papio cynocephalus) with indwelling intravascular catheters.

Baboons with long term, indwelling, intravascular catheters developed clinical signs of renal and hepatic impairment. These included proteinuria and hypoalbuminemia without edema, and albumin to globulin ratios were reversed. Serum IgM, IgG, rheumatoid factor, and liver enzyme concentrations were above normal. Immunofluorescent staining of renal glomerular capillary loops was positive for IgG, IgM, B1c, and C4. Major microscopic lesions were membranoproliferative glomerulonephritis, chronic active hepatitis, degenerative arthritis, and chronic sialoadenitis. Electron microscopy of renal glomer
