Morphine pharmacokinetics and pain assessment after intracerebroventricular administration in patients with terminal cancer.

Morphine pharmacokinetics and pain relief were evaluated after intracerebroventricular administration of morphine (0.4 +/- 0.11 mg) in seven patients with cancer suffering from intractable pain. Ventricular cerebrospinal fluid (CSF), lumbar CSF, and plasma morphine concentrations were analyzed by a specific morphine radioimmunoassay. A two-compartment model was sufficient to describe the kinetics of morphine in ventricular CSF. Morphine diffuses to the lumbar level, and the mean maximum concentration was 192 +/- 105 ng/ml at 4.5 +/- 1.3 hours. Ventricular and lumbar CSF morphine kinetics showe

Recirculation of cerebrospinal fluid through the tela choroidae is why high levels of melatonin can be found in the lateral ventricles.

Evidence is presented to support the hypothesis that cerebrospinal fluid (CSF) is transported through the tela choroidae and recirculated in the ventricular system. The concept that the CSF is resorbed by the arachnoid villus-superior sagittal sinus system is accepted as fact. The experimental studies on which the currently accepted theory is based were published in 1914 by Dr Lewis Weed. Weed used a low pressure (near physiologic) method and a high pressure (non-physiologic) method. His observations with the high pressure method are a basis for the theory of arachnoid villus absorption. On th

The role of atrial natriuretic factor in dehydration and schedule-induced drinking behaviour.

Dehydration-induced drinking (DID) has been defined as a type of homeostatic behaviour controlled by factors related to water balance, whereas schedule-induced polydipsia (SIP) is considered to be a type of nonhomeostatic drinking subsequent to a general increase in motor excitability. In this study, we have attempted to assess the role of atrial natriuretic factor (ANF) in both models to elucidate the mechanisms controlling water intake. Intracerebroventricular injection of ANF (2-8 nmol) caused a dose related suppression of water intake in both DID and SIP, but intravenous injection with a h

Neostigmine-induced hyperglycemia is mediated by central muscarinic receptor in fed rats.

We previously reported that neostigmine injected into the third cerebral ventricle stimulated adrenal secretion of epinephrine, secretion of glucagon from the pancreas, and direct neural innervation of the liver, resulting in hepatic venous plasma hyperglycemia in anesthetized fed rats. However, receptor type of these 3 mechanisms is not known. Therefore, we examined the effects of intraventricularly injected cholinergic or adrenergic antagonists on neostigmine-induced catecholamines in intact rats, glucagon secretion which is mediated by direct neural innervation of pancreas in bilateral adre

5-Iodo-2-deoxyuridine administered into the lateral cerebral ventricle as a radiosensitizer in the treatment of disseminated glioma.

A rat brain tumor model (Fischer 344 rats) with the clinical and pathological features of dissemination via the cerebrospinal fluid (CSF) pathways was used to demonstrate the efficacy of 5-iodo-2-deoxyuridine (IUDR) as a radiosensitizer when it is administered directly into the CSF. Stereotaxic implantation of 9L gliosarcoma cells (5 X 10(5) into the CSF of the lateral cerebral ventricle resulted in widespread dissemination and median survival of 18.5 and 20 days (range, 10-22) in two experiments. A continuous 7-day infusion of IUDR into the CSF starting on the day of tumor implantation did no

Hypertonic saline resuscitation is prevented by intracerebroventricular saralasin but not by captopril.

Hypertonic saline resuscitation (HR, 7.5% NaCl, 4 ml/kg) effectively reverts severe hemorrhage, but a central neural component is probably involved in the survival response. This experiment examines the role of central angiotensinergic pathways in hemorrhage-hypertonic resuscitation interaction. Severely bled (43 +/- 2 ml/kg) pentobarbital-anesthetized dogs with chronically implanted cerebral ventricular cannulae were resuscitated with 4 ml/kg 7.5% NaCl, iv 10 min after intracerebroventricular injection of 0.5 ml normal saline (CT), 150 micrograms saralasin (in 0.5 ml saline, SR), or 10 mg cap

Ablation of the area postrema and emesis.

The emetic action of dopamine, norepinephrine, epinephrine, nicotine, dimethylphenyl-piperazinium (DMPP), and 4-m-chlorophenylcarbamoyloxy-2-butynyltrimethylammonium (McN-A-343) injected intracerebroventricularly (i.c.v.) to the unanesthetized cat was investigated and compared. ED50 values (mg) were as follows: nicotine, 0.011; epinephrine, 0.047; norepinephrine, 0.57; DMPP, 0.9; dopamine, 1.66; and McN-A-343, 4.42. The most potent was nicotine, whereas the least active McN-A-343. On the other hand, DMPP produced the longest emetic response, about 30 min, while McN-A-343-induced emesis lasted

Modification by physostigmine of the cardiovascular responses to intracerebroventricular administration of 5-hydroxytryptamine in rats.

In rats the effect of inhibition of the brain cholinesterase activity on the pressor and heart rate responses to 5-hydroxytryptamine (5-HT), administered into the lateral cerebral ventricle (l.c.v.) was examined. After administration of physostigmine (twice in a small dose of 2.5 micrograms l.c.v., 20 and 15 min before the second injection of 5-HT), the pressor effect of 5-HT (5 micrograms) was strongly reduced or almost abolished, its pure tachycardia was reduced or reversed into a bradycardia and its pure bradycardia was diminished or reversed into a tachycardia. Effects of intraventricular histamine and H2 receptor antagonists on intraocular pressure.

Severe ocular hypertension has been reported in a chronic glaucoma patient following use of histamine H2 receptor antagonists for treatment of peptic ulcer. Subsequent studies, however, have failed to demonstrate a significant action of topical or intravenously administered H2 blockers on intraocular pressure (IOP) in humans. In this study, cimetidine and ranitidine were administered into the cerebral ventricles of unanesthetized New Zealand White rabbits. Both drugs caused prolonged increases in IOP at a dose of 1 umol. Maximal elevations of IOP occurred approximately 20 min after drug inject

Comparison of the action of prostaglandin with endotoxin on thermoregulatory response thresholds.

Prostaglandin E2 (PGE2) and lipopolysaccharide (LPS) derived from E. coli were injected into the lateral cerebral ventricle of rabbits at 30 degrees C ambient temperature. The threshold core temperatures for ear cutaneous vasoconstriction (Thv) and shivering (Thsh) were determined by whole-body cooling with an intestinal thermode. Each threshold, as determined at the plateau phase of LPS fever and PGE2 hyperthermia respectively, were compared with the control values before LPS and PGE2 injection. Thsh was not changed by the injection of LPS, while Thv was increased. After PGE2 injection both T

Prosencephalic afferents to the mediodorsal thalamic nucleus.

The afferent projections from the prosencephalon to the mediodorsal thalamic nucleus (MD) were studied in the cat by use of the method of retrograde transport of horseradish peroxidase (HRP). Cortical and subcortical prosencephalic structures project bilaterally to the MD. The cortical afferents originate mainly in the ipsilateral prefrontal cortex. The premotor, prelimbic, anterior limbic, and insular agranular cortical areas are also origins of consistent projections to the MD. The motor cortex, insular granular area, and some other cortical association areas may be the source of cortical co

Induction of hyperphagia in rats by intracerebroventricular infusion of sodium pentobarbital. A method for testing anorexigenic compounds.

In rats hyperphagia could be reversibly induced by intracerebroventricular infusion of sodium pentobarbital. Hyperphagia was elicited in a dose dependent manner, the optimal infusion rate being 1000 micrograms/h sodium pentobarbital. At this rate, feeding behaviour was induced selectively, with only minor side effects being apparent. Systemic application of d-methamphetamine or propylhexedrine given prior to infusion resulted in a dose dependent reduction of hyperphagia. The method, therefore, could be useful for testing putative anorexigenic compounds in hyperphagic rats.

Effects of intraventricular beta-endorphin on GABA system in some areas of chick brain.

In chicks with cannulae chronically implanted into the III cerebral ventricle, the effects of a single dose (10 micrograms) of beta-endorphin on GABA and free glutamic acid content, GAD and GABA-T activities in the diencephalon, brain-stem and brain hemispheres were studied at the time of maximal behavioural stuporous state and analgesia. A significant decrease in GABA concentration both in the diencephalon and brain-stem, accompanied by a significant increase in GABA-T activity in the same areas, was shown to occur. No changes were observed in GAD activity and in glutamic acid content in the

Central and peripheral adrenergic mechanisms regulating gastric secretion in the rat.

Subcutaneous injection of sympathomimetic agents reduced gastric secretion in pylorus-ligated rats, and the decreasing order of activity was: isoproterenol, norepinephrine, naphazoline and phenylephrine. The effect of naphazoline and phenylephrine was antagonized with 4 mg/kg s.c. of phentolamine, and that of isoproterenol with 4 mg/kg s.c. of propranolol. Thus there exist separate, alpha and beta adrenergic receptors which control gastric secretion in the rat. Intracerebroventricular (i.c.v.) injection of the sympathomimetic agents also reduced secretion, naphazoline being the most potent. Th

Rifamycin in neonatal flavobacteria meningitis.

Three newborn infants with meningitis due to Flavobacterium meningosepticum were treated with rifamycin administered parenterally and directly into the cerebral ventricles. Antibiotic concentrations of blood and cerebrospinal fluid (CSF) were monitored during treatment. There was rapid sterilization of the CSF after this antibiotic. Jaundice was the only toxicity noted. All 3 infants developed hydrocephalus and are shunt dependent. Two of them are otherwise free of neurological complications and are developing normally. Rifamycin is a safe and effective antibiotic in this form of neonatal meni

Transcavernous Sinus Transtuber Cinereum Approach for Pure Third Ventricle Craniopharyngioma Resection.

BACKGROUND AND OBJECTIVES: Craniopharyngiomas confined to the third ventricle present formidable surgical challenges owing to their deep location and proximity to critical neurovascular structures. This study evaluates the transcavernous sinus transtuber cinereum (TCSTC) approach, a technically advanced route that enables direct access to the third ventricle while preserving surrounding anatomy. METHODS: Ten cadaveric heads were meticulously prepared with vascular injections, and the right side was dissected under magnification using a surgical microscope. High-resolution imaging and digital

Reelin supplementation recovers synaptic plasticity and cognitive deficits in a mouse model for Angelman syndrome.

The Reelin signaling pathway is implicated in processes controlling synaptic plasticity and hippocampus-dependent learning and memory. A single direct in vivo application of Reelin enhances long-term potentiation, increases dendritic spine density and improves associative and spatial learning and memory. Angelman syndrome (AS) is a neurological disorder that presents with an overall defect in synaptic function, including decreased long-term potentiation, reduced dendritic spine density, and deficits in learning and memory, making it an attractive model in which to examine the ability of Reelin

Globus pallidus plays a critical role in neurotrophic factor induced functional improvements in hemiparkinsonian monkeys.

This study was designed to test the hypothesis that the integrity of the globus pallidus (GP) is critical for neurotrophic factor, such as glial-derived neurotrophic factor (GDNF), induced functional changes in rhesus macaques with MPTP-induced parkinsonism, because our previous studies demonstrated that the GP was one of the most affected areas as assessed by the levels of dopamine (DA) and its metabolites. A group of eight hemiparkinsonian monkeys with pallidal lesions, which positively responsed to intraventricular (ICV) injections of GDNF prior to the lesions, and a group of eight hemipark
