Calcium activates erythrocyte AMP deaminase [isoform E (AMPD3)] through a protein-protein interaction between calmodulin and the N-terminal domain of the AMPD3 polypeptide.

Erythrocyte AMP deaminase [isoform E (AMPD3)] is activated in response to increased intracellular calcium levels in Tarui's disease, following exposure of ionophore-treated cells to extracellular calcium, and by the addition of calcium to freshly prepared hemolysates. However, the assumption that Ca(2+) is a positive effector of isoform E is inconsistent with the loss of sensitivity to this divalent cation following dilution of erythrocyte lysates or enzyme purification. Ca(2+) regulation of isoform E was studied by examining in vitro effects of calmodulin (CaM) on this enzyme and by monitorin

Dietary glucoraphenin ameliorates obesity and steatotic liver disease associated with gut dysbiosis induced by a Western diet in mice.

Western-style diets promote obesity, gut dysbiosis, and metabolic dysfunction-associated steatotic liver disease (MASLD). In this study, glu … sed by WD and recovered serum branched-chain amino acids and microbiota-derived purine metabolites (inosine and hypoxanthine). These findings indicate that GRE mitigates WD-induced obesity and MASLD via coordinated regulation of lipid metabolism and the gut-liver axis.

Metabolomic Changes Underpinning Permethrin Resistance in the Anopheles gambiae Vector from Cameroon.

Insecticide resistance reduces the effectiveness of current malaria-vector-control interventions. To understand the mechanisms underlying permethrin resistance in field-derived Anopheles gambiae vectors collected from Cameroon, we used ultra-high-performance liquid chromatography coupled with high-resolution tandem mass spectrometry (UHPLC-HRMS/MS) to comprehensively analyze metabolic changes in resistant and susceptible samples to gain insights into mechanisms driving permethrin resistance. Resistant mosquitoes exhibited upregulated levels of inosine, nicotinic acid, dipeptides, amino acids, …

Differential A-to-I editing of SINE B2 RNAs unveils an epitranscriptome response to Aβ neurotoxicity.

Adenosine-to-inosine (A-to-I) RNA editing is a major epitranscriptomic mechanism, yet its contribution to non-coding RNA regulation during neurodegeneration is largely unknown. SINE B2 RNAs represent the dominant editing substrates in mice and have been shown to regulate gene expression. Here, we introduce and validate a repeat-aware bioinformatics framework that enables position-specific quantification of A-to-I editing within SINE RNAs, which has been challenging using standard genome-based pipelines. Applying this approach, we identify discrete editing hotspots in mouse SINE B2 RNAs that ar…

Mycophenolic acid exerts dichotomous regulation of hepatic lipogenesis in a metabolic context-dependent manner.

With the increasing global burden of fatty liver disease (FLD), liver transplantation (LT) recipients face heightened risks of steatosis rec … doxically inhibiting lipid accumulation in steatotic hepatic models. Mechanistically, we identified inosine monophosphate dehydrogenase 2 (IMPDH2) as the molecular switch governing this dichotomy. Notably, significantly increased interaction between IMPDH2 and PPARγ was observed following MPA treatment. Our findings highlight the intricate role of MPA in …

A single-cell transcriptome atlas reveals paradoxical monoallelic expression of RNA editing and heterozygous SNPs.

Both genomic mutations and RNA editing contribute to functional complexity and drive adaptive evolution. Single-cell profiling offers deep insight into the cis-regulatory mechanisms underlying these variations. Using 13 025 single-cell Smart-Seq libraries from whole-body Drosophila melanogaster, we unexpectedly found that 94.0% of adenosine-to-inosine RNA editing sites and 92.8% of heterozygous single nucleotide polymorphismss (SNPs) with sufficient "unique fragment support" exhibit binary expression (0 or 1) in a single cell. The genotypes of representative heterozygous SNPs were validated by…

ADAR1 as a Placental Innate Immune Rheostat Sustaining the Homeostatic Balance of Intrinsic Interferon Response at the Maternal-Fetal Interface.

The mechanisms that balance a robust intrinsic antiviral defense at the maternal-fetal interface with fetal development remain elusive. Here, it is delineated that ADAR1, an adenosine-to-inosine (A-to-I) editor, fine-tunes intrinsic interferon (IFN)-mediated integrates stress response (ISR) in the mouse placenta, thereby orchestrating embryonic development and antiviral defense. Placental Adar1 deletion (Adar1PKO) trigger spatially resolved overwhelming IFN responses, which impair the differentiation of IFN hyper-responsive junctional zone (JZ) progenitors and functions of the placental JZ, ul…

Enhancing RNA base editing on mammalian transcripts with small nuclear RNAs.

Endogenous uridine-rich small nuclear RNAs (U snRNAs) form RNA-protein complexes to process eukaryotic pre-mRNA into mRNA. Previous studies have demonstrated programmable U snRNA guide-targeted exon inclusion and exclusion. Here we investigated whether snRNAs can also enhance RNA base editing over state-of-the-art RNA-targeting technologies in human cells. Compared with adenosine deaminase acting on RNA (ADAR)-recruiting circular RNAs, we find that guided A>I snRNAs consistently increase adenosine-to-inosine editing for higher exon count genes, perturb substantially fewer off-target genes and …

Uncovering a role for METTL13 in malignant transformation of human hematopoietic stem cells and in the progression of pediatric leukemia.

Post-transcriptional RNA modifications, such as N6-methyladenosine (m6A) methylation and adenosine to inosine (A-to-I) editing, are critical regulators of hematopoietic stem cell (HSC) self-renewal and differentiation, yet their precise contributions to malignant transformation are not fully elucidated. In this study, we uncovered the epitranscriptomic landscape caused by knockdown of genes from the methyltransferase (METTL)-family in hematopoietic stem and progenitor cells (HSPCs). We identified both converging and distinct roles of METTL3 and METTL14, known members of the m6A writer complex,…

Malignant A-to-I RNA editing by ADAR1 drives T-cell acute lymphoblastic leukemia relapse via attenuating dsRNA sensing.

Leukemia initiating cells (LICs) are regarded as the origin of leukemia relapse and therapeutic resistance. Identifying direct stemness determinants that fuel LIC self-renewal is critical for developing targeted approaches to eliminate LICs and prevent relapse. Here, we show that the RNA editing enzyme ADAR1 is a crucial stemness factor that promotes LIC self-renewal by attenuating aberrant double-stranded RNA (dsRNA) sensing. Elevated adenosine-to-inosine (A-to-I) editing is a common attribute of relapsed T-ALL regardless of molecular subtypes. Consequently, knockdown of ADAR1 severely inhibi…

ADAR2 increases in exercised heart and protects against myocardial infarction and doxorubicin-induced cardiotoxicity.

Exercise training benefits the heart. The knowledge of post-transcription regulation, especially RNA editing, in hearts remain rare. ADAR2 is an enzyme that edits adenosine to inosine nucleotides in double-stranded RNA, and RNA editing is associated with many human diseases. We found that ADAR2 was upregulated in hearts during exercise training. AAV9-mediated cardiac-specific ADAR2 overexpression attenuated acute myocardial infarction (AMI), MI remodeling, and doxorubicin (DOX)-induced cardiotoxicity. In vitro, overexpression of ADAR2 inhibited DOX-induced cardiomyocyte (CM) apoptosis. but it …

ADAR1-mediated RNA editing is required for thymic self-tolerance and inhibition of autoimmunity.

T cells play a crucial role in the adaptive immune system, and their maturation process is tightly regulated. Adenosine deaminase acting on RNA 1 (ADAR1) is the enzyme responsible for adenosine-to-inosine RNA editing in dsRNAs, and loss of ADAR1 activates the innate immune sensing response via melanoma differentiation-associated protein 5 (MDA5), which interprets unedited dsRNA as non-self. Although ADAR1 is highly expressed in the thymus, its role in the adaptive immune system, especially in T cells, remains elusive. Here, we demonstrate that T cell-specific deletion of Adar1 in mice causes a…

ADAR1 Activation Drives Leukemia Stem Cell Self-Renewal by Impairing Let-7 Biogenesis.

Post-transcriptional adenosine-to-inosine RNA editing mediated by adenosine deaminase acting on RNA1 (ADAR1) promotes cancer progression and therapeutic resistance. However, ADAR1 editase-dependent mechanisms governing leukemia stem cell (LSC) generation have not been elucidated. In blast crisis chronic myeloid leukemia (BC CML), we show that increased JAK2 signaling and BCR-ABL1 amplification activate ADAR1. In a humanized BC CML mouse model, combined JAK2 and BCR-ABL1 inhibition prevents LSC self-renewal commensurate with ADAR1 downregulation. Lentiviral ADAR1 wild-type, but not an editing-d…
