The brain's primary immune cells, microglia, are a leading causal cell type in Alzheimer's disease (AD). Yet, the mechanisms by which microglia can drive neurodegeneration remain unresolved. Here, we discover that a conserved stress signaling pathway, the integrated stress response (ISR), characterizes a microglia subset with neurodegenerative outcomes. Autonomous activation of ISR in microglia is sufficient to induce early features of the ultrastructurally distinct "dark microglia" linked to pathological synapse loss. In AD models, microglial ISR activation exacerbates neurodegenerative patho…

The effects of repeated cold stress on physiological, transcriptional, and metabolic processes was studied in Arabidopsis thaliana. An experiment included stages of initial chilling, acclimation, and repeated cold treatment. Repeated cold stress increased electrolyte leakage and decreased the chlorophyll a content, indicating a disruption of the membrane integrity and a restructuring of the photosynthetic apparatus. Expression analysis showed upregulation of the DREB1A, MYC2, COR15A, and COR15B genes, which are associated with the cold response and stress memory formation. Transcription of the

As sessile organisms, plants constantly face environmental stresses that disrupt reactive oxygen species (ROS) homeostasis. ROS function as critical signaling molecules orchestrating stress adaptation, while microRNAs (miRNAs) have emerged as master regulators of gene expression. Accumulating evidence reveals a bidirectional regulatory network between miRNAs and ROS. In this review, we first examine how miRNAs compartment-specific regulate ROS metabolism in the apoplast, mitochondria, and chloroplasts. We then delineate the multi-layered control of miRNA biogenesis by ROS, encompassing redox-s

Microplastic contamination in urban water systems poses emerging risks to human health, yet integrated frameworks linking sources, exposure pathways, and health outcomes remain limited. This study develops an integrated machine learning-Bayesian-structural equation modeling framework to quantify and predict microplastic-driven health risks across urban water systems. A systematic literature review screened 1,742 records, of which 47 studies met the predefined eligibility criteria and were included in the final analysis. Here, we integrate machine learning, Bayesian inference, and structural eq

Multifunctional hydrogels with integrated antioxidant and antibacterial activities are vital for modulating the wound microenvironment, mitigating oxidative stress, and preventing infection. However, efficient construction of such hydrogels with programmable responsiveness and structural integrity that can be tailored to therapeutic demands remains challenging. Herein, we developed a modular, functionally programmable hydrogel platform constructed via an in situ Schiff base reaction between aldehyde-functionalized polyphenolic nanoparticles and aminoglycoside antibiotics. The resulting nanocom

Degenerative musculoskeletal diseases are structural and functional failures of the musculoskeletal system, including osteoarthritis, osteoporosis, intervertebral disc degeneration (IVDD), and sarcopenia. As the global population ages, degenerative musculoskeletal diseases are becoming more prevalent. However, the pathogenesis of degenerative musculoskeletal diseases is not fully understood. Previous studies have revealed that endoplasmic reticulum (ER) stress is a stress response that occurs when impairment of the protein folding capacity of the ER leads to the accumulation of misfolded or un

Stress, a prevalent factor in modern life, profoundly impacts human health by disrupting immune function and intestinal homeostasis. This review elucidates the mechanisms through which stress dysregulates the hypothalamic-pituitary-adrenal (HPA) and sympathetic-adrenal medulla (SAM) axes, leading to altered glucocorticoid and catecholamine levels that impair immune responses and promote inflammation. Chronic stress exacerbates gut microbiota dysbiosis, reduces microbial diversity, and compromises intestinal barrier integrity, contributing to conditions like inflammatory bowel disease (IBD). St

Plants are increasingly subjected to concurrent heat stress and elevated CO2; however, how they perceive and integrate these combined stresses remains poorly understood. Here, we identify the β-carbonic anhydrase βCA2 as a central signaling hub in tomato that decodes these combined cues. We demonstrate that βCA2 physically interacts with the receptor kinase PSKR1 at the plasma membrane and that PSKR1-mediated phosphorylation of βCA2 is significantly enhanced under simultaneous elevated CO2 and heat stress. We further identify Ser231 as the crucial phosphorylation site on βCA2. Phosphorylation

Heat stress is a major environmental constraint limiting the productivity of perennial ryegrass (Lolium perenne), a widely cultivated forage and turfgrass. Here, through a genome-wide association study (GWAS) analysis of a diverse perennial ryegrass population, we identified LpbZIP41 as a major heat tolerance quantitative trait locus (QTL) for heat tolerance. Natural variation at this locus comprises three haplotypes showing differential thermotolerance, with evidence of positive selection during breeding. LpbZIP41 localizes to the nucleus and exhibits rapid heat-inducible expression. Function

BACKGROUND AND AIM: Cryopreservation of boar semen is widely used for genetic conservation and artificial insemination; however, it induces oxidative stress and molecular alterations that compromise sperm function. Epigenetic modifications, particularly DNA methylation, play a critical role in sperm integrity and fertilization capacity. This study aimed to investigate genome-wide DNA methylation changes and their association with oxidative stress during cryopreservation of Tibetan pig semen.
MATERIALS AND METHODS: Semen samples were collected from healthy Tibetan pigs and cryopreserved using

Recent research suggests an involvement of hippocampal neurogenesis in behavioral effects of antidepressants. However, the precise mechanisms through which newborn granule neurons might influence the antidepressant response remain elusive. Here, we demonstrate that unpredictable chronic mild stress in mice not only reduces hippocampal neurogenesis, but also dampens the relationship between hippocampus and the main stress hormone system, the hypothalamo-pituitary-adrenal (HPA) axis. Moreover, this relationship is restored by treatment with the antidepressant fluoxetine, in a neurogenesis-depend

Hypothermia is a serious condition marked by a significant decrease in core body temperature, posing considerable risks to biological systems. In response to thermal stress, cells activate protective mechanisms, often synthesizing heat shock proteins (HSPs). These highly conserved proteins are crucial in cellular stress responses, primarily functioning as chaperones. HSPs facilitate correct protein folding and prevent misfolding and aggregation, thereby protecting cellular integrity during adverse conditions. This paper explains how HSPs alleviate stress responses related to low body temperatu

The neutrophil-to-lymphocyte ratio (NLR) is a predictor of morbidity for a variety of medical conditions, but little is known about how variation in NLR arises. We examined variation in this measure in a sample of 4577 infant rhesus monkeys (54.8 % female), who participated in the BioBehavioral Assessment program at the California National Primate Research Center at 3-4 months of age. Lower values for NLR were seen for animals reared indoors, for animals that were raised to be free of specific pathogens, and for males. In addition lower NLR was associated with higher stress values of cortisol

Wsc1p and Mid2p are transmembrane signaling proteins of cell wall stress in the budding yeast Saccharomyces cerevisiae When an environmental stress compromises cell wall integrity, they activate a cell response through the Cell Wall Integrity (CWI) pathway. Studies have shown that the cytoplasmic domain of Wsc1p initiates the CWI signaling cascade by interacting with Rom2p, a Rho1-GDP-GTP exchange factor. Binding of Rom2p to the cytoplasmic tail of Wsc1p requires dephosphorylation of specific serine residues but the mechanism by which the sensor is dephosphorylated and how it subsequently inte

In mammals, stress elicits a stereotyped endocrine response that requires an increase in the activity of hypothalamic parvocellular neuroendocrine neurons. The output of these cells is normally constrained by powerful GABA-mediated synaptic inhibition. We found that acute restraint stress in rats released the system from inhibitory synaptic drive in vivo by down-regulating the transmembrane anion transporter KCC2. This manifested as a depolarizing shift in the reversal potential of GABA(A)-mediated synaptic currents that rendered GABA inputs largely ineffective. Notably, repetitive activation
