Serum leptin and body weight in females with anorexia and bulimia nervosa.

In this study we hypothesized that there is a correlation between serum leptin levels and body mass indices within patients with anorexia nervosa or bulimia nervosa during a twelve weeks' course of in-patient treatment. We evaluated leptin levels weekly in female in-patients with anorexia (n = 17) or bulimia nervosa (n = 18). Only patients with anorexia nervosa were therapeutically encouraged to gain weight throughout the treatment episode. For the whole cohort, body mass indices and serum leptin levels were highly correlated upon admission (r = 0.89, p < 0.001). The median intra-individual co…

Inhibition of insulin secretion by leptin in normal rodent islets of Langerhans.

The recently discovered adipose cell-specific hormone called leptin decreases food intake and increases energy expenditure in rodents through a pathway involving hypothalamic leptin receptors, OB-R. In addition, leptin decreases insulin circulating levels independent of the reduction in food intake. Whether or not the hormone has a direct effect on pancreatic beta-cells is not clear, because previous in vitro studies have led to controversial results depending on the animal model used. The present study was designed to investigate the effects of leptin in islets of Langerhans isolated from nor…

The expression of leptin and its receptors in pre-ovulatory human follicles.

The expression of leptin and its receptors was examined by reverse transcriptase-polymerase chain reaction and immunofluorescence in granulosa and cumulus cells of pre-ovulatory follicles and in meiotically mature oocytes obtained from women undergoing in-vitro fertilization. Leptin concentrations were measured in newly aspirated follicular fluids and in maternal serum before and after the administration of an ovulatory dose of human chorionic gonadotrophin. The findings demonstrate leptin expression at the mRNA and protein levels by granulosa and cumulus cells, and the presence of leptin in m…

Do leptin receptors play a functional role in the endocrine pancreas?

It was recently speculated that leptin may exert a direct inhibitory effect upon insulin release from the pancreatic B-cell. This proposal meets, however, with two objections. First, although the message for leptin receptors is indeed detected in rat pancreatic islets, the short form of this receptor, for which no signalling function is known, represents the major species present in islet cells. Second, in the isolated perfused rat pancreas, leptin (1.0 nM) fails to affect the release of either insulin or glucagon.

Leptin in the Commissural Nucleus of the Tractus Solitarius (cNTS) and Anoxic Stimulus in the Carotid Body Chemoreceptors Increases cNTS Leptin Signaling Receptor and Brain Glucose Retention in Rats.

Background and Objectives: The commissural nucleus of the tractus solitarius (cNTS) not only responds to glucose levels directly, but also receives afferent signals from the liver, and from the carotid chemoreceptors (CChR). In addition, leptin, through its receptors in the cNTS, regulates food intake, body weight, blood glucose levels, and brain glucose retention (BGR). These leptin effects on cNTS are thought to be mediated through the sympathetic-adrenal system. How these different sources of information converging in the NTS regulate blood glucose levels and brain glucose retention remains…

Unravelling obesity: from leptin to glucagon-like peptide-1 receptor agonists.

The global obesity epidemic involves a multifaceted interplay between genetics, environment, and the interactions between them. The discovery of leptin in 1994 changed our understanding of body weight regulation and triggered extensive genetic studies into monogenic and polygenic obesity. Despite initial therapeutic hopes, leptin's efficacy in obesity was limited by leptin resistance. However, glucagon-like peptide-1, an incretin hormone, has become an important tool for managing obesity. Glucagon-like peptide-1 receptor agonists aid in weight loss and glycemic control while also providing met…

Leptin in Dairy Cows: Metabolic Adaptation, Reproductive Function, and Health Applications.

Leptin (LEP) is an adipocyte-derived cytokine that integrates nutritional status, metabolism, and reproduction in cattle, with particular relevance for modern high-producing dairy cows. In ruminants, LEP and its receptors are widely expressed in metabolic and reproductive tissues, including adipose tissue, liver, hypothalamus, pituitary, ovary, uterus, and placenta, where LEP modulates energy homeostasis, neuroendocrine function, and local tissue responses. Changes in circulating LEP concentrations during the transition period reflect changes in body fat reserve, insulin and GH-IGF-1 dynamics,…

Leptin-driven microglial activation in fibromyalgia: mechanistic insights and therapeutic implications.

Fibromyalgia (FM) is a chronic nociplastic pain disorder marked by widespread musculoskeletal pain, fatigue, sleep disturbances, and cognitive dysfunction. Central sensitization (CS) and sustained neuroimmune dysregulation are thought to contribute to disease persistence. Although aberrant microglial activation is increasingly recognized as a potential amplifier of nociceptive signaling in FM, the upstream immuno-metabolic drivers of this process remain incompletely defined. Emerging evidence suggests that leptin, an adipocyte-derived cytokine-like hormone, may act as an immunomodulator linkin…

Leptin Therapy Improves Metabolic Dysfunction in Immune Checkpoint Inhibitor-induced Lipodystrophy.

Immune checkpoint inhibitors (ICI) have transformed cancer therapy but can induce a spectrum of endocrine complications. Acquired generalized lipodystrophy (AGL) is a rare adverse effect, leading to severe metabolic derangements including insulin-resistant diabetes, dyslipidemia, and hepatic steatosis. Marked hypoleptinemia is noted in these patients, and leptin-replacement therapy is known to be beneficial in patients with other forms of AGL. Though rare, T-cell lymphoma has been reported in patients with AGL on leptin therapy, and there has been much debate on the association between hyperle…

Leptin antagonism improves Rett syndrome phenotype in symptomatic Mecp2-deficient mice.

Rett syndrome (RTT) is a severe X-linked neurodevelopmental disorder caused by mutations in MECP2. Elevated circulating levels of the adipocyte hormone leptin are consistently observed in patients and in mouse models, yet their contribution to disease progression has remained unclear. Here, we show that reducing leptin signaling-either pharmacologically or genetically-significantly alleviates RTT-like phenotypes in Mecp2-deficient mice. In males, these interventions preserved general health, prevented weight loss, and improved breathing and locomotor functions. At the neuronal level, they rest…

FAK regulates leptin-induced angiogenesis and vasculogenic mimicry in breast cancer.

Leptin, an adipokine primarily secreted by adipose tissue, has been implicated in tumor progression by regulating angiogenesis. Leptin also plays a key role in tumor vascularization by promoting vasculogenic mimicry (VM). During tumor progression, leptin activates several signaling pathways, including the focal adhesion kinase (FAK) pathway. This study aimed to determine whether leptin promotes and regulates angiogenesis and VM through the FAK pathway. We used the chick chorioallantoic membrane (CAM) as a model to evaluate blood vessel formation and to induce tumors to assess angiogenic expres…

Elevated seminal plasma leptin may correlate with varicocele presence and BMI.

OBJECTIVES: Male infertility affects 20-30 % of infertile couples worldwide, and varicocele being a common cause. The relationship between leptin, a hormone involved in energy homeostasis and reproduction, and male fertility parameters remains incompletely understood, particularly in the context of varicocele and obesity. Thus, the current study sought to assess the associations between seminal plasma leptin and other clinical and demographic data of infertile patients. METHODS: In this cross-sectional study, 90 men (29 with varicocele, 61 without) were evaluated from November 2023 to March 2…

Leptin/PPARγ interaction mediates obesity-driven Th17 differentiation in rheumatoid arthritis.

Rheumatoid arthritis (RA) is a chronic autoimmune disorder, with some studies suggesting that obesity may increase the risk of developing RA. However, the relationship between obesity and RA is complex, involving both epidemiological associations and, paradoxically, protective effects. The exact role of obesity in RA pathophysiology remains controversial. In this study, we investigated the impact of obesity on RA progression, focusing on the molecular mechanisms of immune regulation mediated by the adipokine leptin. We examined obese RA patients and employed high-fat diet (HFD)-induced obesity…

Leptin Improves Parameters of Brown Adipose Tissue Thermogenesis in Lipodystrophic Mice.

Lipodystrophy syndromes (LD) are a heterogeneous group of very rare congenital or acquired disorders characterized by a generalized or partial lack of adipose tissue. They are strongly associated with severe metabolic dysfunction due to ectopic fat accumulation in the liver and other organs and the dysregulation of several key adipokines, including leptin. Treatment with leptin or its analogues is therefore sufficient to reverse some of the metabolic symptoms of LD in patients and in mouse models through distinct mechanisms. Brown adipose tissue (BAT) thermogenesis has emerged as an important …

Factors affecting cord blood leptin levels in a consecutive birth cohort.

INTRODUCTION: The fetus that fails to meet its ideal growth trajectory has increased risks of poor health outcomes throughout life. "Gold standard" methods of anthropometric assessment such as measurement of percentage body fat can be difficult to apply across populations and other biomarkers such as serum concentration of umbilical cord blood leptin may be more effective for screening. This study reports cord blood leptin levels in a large prospective consecutive birth cohort and assesses the relationship between leptin and neonatal and maternal factors. METHODS: Venous umbilical cord blood …
