Ecology of the free-living stages of Trichostrongylid parasites of sheep.

The prevalence of gastrointestinal nematodes (GINs) is dictated by environmental conditions which influence the survival, development, and m … es ranging from 0° to 40°C and nominal target FWCs ranging from 0% to 80% for 8 days. The different life cycle stages (eggs, pre-infective larvae, intra-pellet infective larvae and extra-pellet infective larvae) were enumerated each day using a modified McMaster technique. Multiple linear regression analysis was used to assess the impact of environmental effects on …

Defining Stage-Specific Activity of Potent New Inhibitors of Cryptosporidium parvum Growth In Vitro.

Cryptosporidium parvum and Cryptosporidium hominis have emerged as major enteric pathogens of infants in the developing world, in addition to their known importance in immunocompromised adults. Although there has been recent progress in identifying new small molecules that inhibit Cryptosporidium sp. growth in vitro or in animal models, we lack information about their mechanism of action, potency across the life cycle, and cidal versus static activities. Here, we explored four potent classes of compounds that include inhibitors that likely target phosphatidylinositol 4 kinase (PI4K), phenylala

Genetic Characterization of Coenzyme A Biosynthesis Reveals Essential Distinctive Functions during Malaria Parasite Development in Blood and Mosquito.

Coenzyme A (CoA) is an essential universal cofactor for all prokaryotic and eukaryotic cells. In nearly all non-photosynthetic cells, CoA bi … out the cellular functions of the other enzymes of the CoA biosynthesis pathway in malaria parasite life cycle stages. All enzymes of this pathway were targeted for deletion or deletion/complementation analyses by knockout/knock-in plasmid constructs to reveal their essential roles in P. yoelii life cycle stages. Inhibition of PfCLK3 interferes with malaria parasite RNA splicing and explains the mechanism of action of a new class of antimalarial candidates.

Emerging resistance to front-line antimalarials means there is an urgent need to discover new drugs with novel mechanisms of action that rem … the affected genes were involved in numerous essential parasite processes associated with multiple life cycle stages and revealed transcripts and introns particularly susceptible to inhibition. Our study supports the role of PfCLK3 as an important regulator of spliceosome activity and establishes the distinct mechanism of parasiticidal activity of PfCLK3 inhibitor …

Leishmania major: common antigen responsible for induction of delayed-type hypersensitivity response in guinea pigs.

The cellular response to Leishmania major (L. major) was evaluated in vivo by delayed-type hypersensitivity (DTH) skin test reaction using leishmanin as antigen. Our previous study had shown the development of species-specific DTH reaction in sensitized guinea pigs by application of a single purified antigen from promastigotes and filtered culture supernatants of L. major. This study has shown that purified antigen is common in both stages of the life cycle and filtered culture supernatant of L. major. The common antigen was purified and analyzed from soluble Leishmania antigen (SLA) of amasti

Extracellular like-gregarine stages of Cryptosporidium parvum.

The present study confirms the existence of extracellular stages of Cryptosporidiumparvum during in vitro culture on MDCK, HCT 8 and Vero cells as well as alveolar macrophages, by optic, Nomarski and transmission electron microscopy images. Extracellular trophozoite/gamont, stages in syzygy, zygotes and spores with eight sporozoites were seen in the supernatant of the cultures. The first ultrastructural images of extracellular stages of C. parvum are shown in this study. The morphology of these stages, which have characteristics similar to those of some gregarines, support the contention that

Dietary Micronutrients from Zygote to Senility: Updated Review of Minerals' Role and Orchestration in Human Nutrition throughout Life Cycle with Sex Differences.

Micronutrients such as selenium, fluoride, zinc, iron, and manganese are minerals that are crucial for many body homeostatic processes supplied at low levels. The importance of these micronutrients starts early in the human life cycle and continues across its different stages. Several studies have emphasized the critical role of a well-balanced micronutrient intake. However, the majority of studies looked into or examined such issues in relation to a specific element or life stage, with the majority merely reporting the effect of either excess or deficiency. Herein, in this review, we will loo

Establishment of the complete life cycle of Spirometra (Cestoda: Diphyllobothriidae) in the laboratory using a newly isolated triploid clone.

Methods to maintain the life cycle of pathogenic organisms become powerful tools for studying molecular and cellular bases of infectious diseases. Spirometra erinaceieuropaei is a parasitic tapeworm that causes sparganosis in humans. Because S. erinaceieuropaei has a complex life cycle with different stages and host species requirements, there have been no reports to establish the complete life cycle in the laboratory. In this study, using Cyclops as the first intermediate host, mouse as the experimental second intermediate host, and dog as the final host, we succeeded in maintaining S. erinac

From Genes to Transcripts, a Tightly Regulated Journey in Plasmodium.

Over the past decade, we have witnessed significant progresses in understanding gene regulation in Apicomplexa including the human malaria parasite, Plasmodium falciparum. This parasite possesses the ability to convert in multiple stages in various hosts, cell types, and environments. Recent findings indicate that P. falciparum is talented at using efficient and complementary molecular mechanisms to ensure a tight control of gene expression at each stage of its life cycle. Here, we review the current understanding on the contribution of the epigenome, atypical transcription factors, and chroma

Toxoplasma gondii merozoite gene expression analysis with comparison to the life cycle discloses a unique expression state during enteric development.

BACKGROUND: Considerable work has been carried out to understand the biology of tachyzoites and bradyzoites of Toxoplasma gondii in large part due to in vitro culture methods for these stages. However, culturing methods for stages that normally develop in the gut of the definitive felid host, including the merozoite and sexual stages, have not been developed hindering the ability to study a large portion of the parasite's life cycle. Here, we begin to unravel the molecular aspects of enteric stages by providing new data on merozoite stage gene expression. RESULTS: To profile gene expression d

Reproduction and love: strategies of the organism's cellular defense system?

A novel view is presented which states that primordial germ cells and their descendants can be regarded as 'cancerous cells' which emit signals that activate a whole array of cellular defensive mechanisms by the somatoplasm. These cells have become unrestrained in response to the lack of typical cell adhesion properties of epithelial cells. From this point of view: (1) the encapsulation of oocytes by follicle cells, vitelline membrane and egg shell; (2) the suppression of gonadal development in larval life; (3) the production of sex steroid hormones and of vitellogenin; and (4) the expulsion o

Mapping diversity in African trypanosomes using high resolution spatial proteomics.

African trypanosomes are dixenous eukaryotic parasites that impose a significant human and veterinary disease burden on sub-Saharan Africa. Diversity between species and life-cycle stages is concomitant with distinct host and tissue tropisms within this group. Here, the spatial proteomes of two African trypanosome species, Trypanosoma brucei and Trypanosoma congolense, are mapped across two life-stages. The four resulting datasets provide evidence of expression of approximately 5500 proteins per cell-type. Over 2500 proteins per cell-type are classified to specific subcellular compartments, pr

Optimal outbreeding is shaped during larval life history in the splash pool copepod Tigriopus californicus.

Inbreeding and outbreeding depression are dynamic forms of selection critical to mating system evolution and the efficacy of conservation biology. Most evidence on how the relative severity and timing of these forces are shaped is confined to self-fertilization, distant outcrossing, and intermediate "optimal outcrossing" in hermaphrodites. We tested the notion that closed population demographics may reduce and delay the costs of inbreeding relative to distant outbreeding in an intertidal copepod with separate sexes and a biphasic larval/post-metamorphic life history (Tigriopus californicus). A

Fasciola hepatica miracidia are dependent on respiration and endogenous glycogen degradation for their energy generation.

It is generally accepted that free-living stages of parasitic helminths are dependent on aerobic degradation of endogenous energy sources for their energy generation. This concept, however, is not the result of extensive experimental evidence, but originated mainly intuitively as oxygen is widely available in their habitat and these stages generally have a small size. Schistosoma mansoni, the sole parasitic helminth whose energy metabolism has been studied throughout its life-cycle indeed has aerobically functioning free-living stages. However, large differences exist in energy metabolism betw

Maternal and Cold-Stratification Warming Outweigh Germination-phase Warming in Regulating Seed Germination of Alpine Plants.

BACKGROUND AND AIMS: Global warming impacts on key plant life-history stages represent a central focus in ecology. Seed germination, the critical starting point in the plant life cycle, lacks a systematic understanding of its response mechanisms to climate warming. METHODS: Here, we examined how warming at the maternal, cold stratification, and germination stages affects seed germination in ten alpine plant species. KEY RESULTS: Cold stratification (absence vs. presence) is decisive in regulating seed dormancy, but its effect is modulated by temperatures experienced during both the maternal

Comparative Analysis of the Humoral Immune Response to the EBV Proteome across EBV-Related Malignancies.

BACKGROUND: Epstein-Barr virus (EBV) is linked to multiple cancers, including classical Hodgkin lymphoma (cHL), endemic Burkitt lymphoma (eBL), nasopharyngeal carcinoma (NPC), and extranodal natural killer/T-cell lymphoma (NKTCL). METHODS: Anti-EBV IgG and IgA antibody responses targeting 202 sequences from 86 EBV proteins were measured using the same EBV whole proteome array across four case-control studies investigating EBV-positive cHL, eBL, NPC, and NKTCL (407 cases/620 controls). We grouped EBV-targeted antibodies into pathways by immunoglobulin type (IgA and IgG) and life-cycle stage (l
