Tissue and Salivary NMR Metabolomics in Reticular-Type Oral Lichen Planus.

Oral lichen planus (OLP) is a chronic T-cell-mediated autoimmune disease, with low potential for malignant transformation. Its etiology remains unclear, necessitating immunohistochemical and molecular-level studies to enhance diagnosis and management. Thirteen patients diagnosed with OLP and 13 healthy controls (HCs) were enrolled from three centers. Mucosal tissue samples collected during diagnostic biopsies and unstimulated whole saliva samples were analysed. A comprehensive approach was taken, with high-resolution magic angle spinning (HR-MAS) 1H-NMR spectroscopy performed on biopsies and l

Untargeted Plasma Metabolomics Extends the Biomarker Profile of Mitochondrial Neurogastrointestinal Encephalomyopathy.

Mitochondrial neurogastrointestinal encephalomyopathy (MNGIE) is caused by pathogenic mutations in the nuclear TYMP gene, which encodes the cytosolic enzyme thymidine phosphorylase. In addition to the systemic accumulation of thymidine and deoxyuridine, several case studies have reported abnormalities in a range of other metabolites in patients with MNGIE. Since metabolites are intermediates or end-products of numerous biochemical reactions, they serve as highly informative indicators of an organism's metabolic activity. This study aimed to perform an untargeted metabolomic profiling to determ

Insights from Metabolomics Profiling of MSUD in Pediatrics Toward Disease Progression.

BACKGROUND: Maple syrup urine disease (MSUD) is a genetic disorder caused by mutations in the branched-chain α-ketoacid dehydrogenase (BCKDH) complex, leading to toxic buildup of branched-chain amino acids (BCAAs) and their ketoacid derivatives. While newborn screening (NBS) and molecular testing are standard diagnostic tools, they face challenges such as delayed results and false positives. Untargeted metabolomics has emerged as a complementary approach, offering comprehensive metabolic profiling and potential for novel biomarker discovery. We previously applied untargeted metabolomics to neo…

Targeted metabolomics and transcript profiling of methyltransferases in three coffee species.

Coffee plants contain well-known xanthines as caffeine. Three Coffea species grown in a controlled greenhouse environment were the focus of this research. Coffea arabica and C. canephora are two first principal commercial species and commonly known as arabica and robusta, respectively. Originating in Central Africa, C. anthonyi is a novel species with small leaves. The xanthine metabolites in flower, fruit and leaf extracts were compared using both targeted and untargeted metabolomics approaches. We evaluated how the xanthine derivatives and FQA isomers relate to the expression of biosynthetic…

Metabolomics-based analysis of nitric oxide regulation of ginseng herb quality.

BACKGROUND: Ginsenosides, the primary active ingredients in Panax ginseng, are secondary metabolites. However, their content varies significantly across batches due to differences in environmental conditions and production methods. Ecological stress can increase the levels of reactive oxygen species (ROS) in plants, and ROS can enhance secondary metabolism. Nitric oxide (NO) can promote the production of O2 ·- and H2O2. This study utilized physiological and non-targeted metabolomics to investigate how NO regulates ginseng quality and how P. ginseng adapts to adversity. RESULTS: Sodium nitropr…

PyINETA: Open-source platform for INADEQUATE-JRES integration in NMR metabolomics.

Annotating compounds with high confidence is a critical element in metabolomics. 13C-detection NMR experiment INADEQUATE (incredible natural abundance double-quantum transfer experiment) stands out as a powerful tool for structural elucidation, whereas this valuable experiment is not often included in metabolomics studies. This is partly due to the lack of community platform that provides structural information based INADEQUATE. Also, it is often the case that a single study uses various NMR experiments synergistically to improve the quality of information or balance total NMR experiment time,…

Advanced progress of spatial metabolomics in head and neck cancer research.

Head and neck cancer ranks as the sixth most prevalent malignancy, constituting 5 % of all cancer cases. Its inconspicuous onset often leads to advanced stage diagnoses, prompting the need for early detection to enhance patient prognosis. Currently, research into early diagnostic markers relies predominantly on genomics, proteomics, transcriptomics, and other methods, which, unfortunately, necessitate tumor tissue homogenization, resulting in the loss of temporal and spatial information. Emerging as a recent addition to the omics toolkit, spatial metabolomics stands out. This method conducts i…
