Targeting ferroptosis by trimetazidine and probiotics to attenuate 5-fluorouracil-induced intestinal mucositis in mice.

Intestinal mucositis is a common side effect in cancer patients receiving chemotherapy, yet effective treatment options are lacking. This study aimed to investigate the potential protective effects of trimetazidine and probiotics combination in a 5-FU-induced intestinal mucositis model. We hypothesized that these agents would attenuate mucositis through anti-inflammatory and antioxidant effects associated with modulation of ferroptosis-related pathways, particularly through Nrf2-associated regulatory mechanisms. 5-fluorouracil in a dose of 50 mg/kg/day i.p. for 6 consecutive days starting from…

Isoquercitrin Alleviates 5-Fluorouracil-Induced Intestinal Mucositis in Mice by Modulating Inflammation and Oxidative Stress.

This study aimed to evaluate the role of isoquercitrin (ISO) in 5-fluorouracil-induced intestinal mucositis. The animals (6 animals/group) were divided into: vehicle (2% DMSO), 5-fluorouracil (5-FU) and groups receiving ISO (10, 50, or 100 mg/kg). ISO at 100 mg/kg (ISO-100) (232.5 ± 3.8 µm) attenuated the reduction in villus height induced by 5-FU (125.3 ± 1.5 µm) and the reduction in crypt depth in the intestine (ISO-100: 101.7 ± 3.5 µm vs 5-FU: 70.8 ± 1.6 µm; p < 0.05). The jejunum was chosen for assays involving oxidative stress and inflammation. ISO-100 attenuated the mastocytosis (ISO-100…

Probiotic preparations in mitigating chemotherapy-induced oral mucositis: therapeutic efficacy, mechanisms, and clinical translation potential.

Chemotherapy-induced oral mucositis (CIOM) is a prevalent toxic side effect of cancer treatment, severely compromising patients' quality of life, nutritional intake, and treatment adherence. Its pathogenesis has evolved from the traditional model of simple epithelial damage to a complex pathological process involving the interplay of chemotherapy toxicity, host immunity, and oral microbiota. Research indicates that chemotherapy can disrupt the oral microbiota, promoting the proliferation of pathogenic bacteria and exacerbating damage to the mucosal barrier and local inflammatory responses. Cur…

Lenalidomide for the Treatment of Chronic Oral Mucositis and Soft Tissue Necrosis After Radiation Therapy.

Radiation-induced oral mucositis is a complication of head and neck cancer therapy that may persist beyond the expected timelines and become refractory to conventional management, adversely affecting patients' quality of life. Radiation therapy for infiltrative tumors can also result in soft tissue necrosis, which may complicate post-treatment recovery. There are currently no therapies approved by the U.S. Food and Drug Administration for persistent radiation-induced soft tissue injury. Herein, we describe 2 patients with head and neck cancer with recalcitrant radiation therapy-associated oral…

New insights into the mechanisms of modified Pulsatilla decoction in alleviating chemotherapy-induced intestinal mucositis.

Chemotherapy-induced intestinal mucositis (IM) is a prevalent complication affecting up to 80% of cancer patients undergoing treatment. Current therapies focus on symptomatic relief rather than addressing the underlying mechanism. Recent advances in integrative medicine highlight the potential of traditional Chinese medicine formulations as alternatives or adjuncts to existing therapies. In this context, this editorial discusses the recent results of a study published by Qiu et al, which investigates the multifaceted potential of modified Pulsatilla decoction (PD), a formulation of PD with lic…

Severe pneumonia with Mycoplasma pneumoniae-induced raѕh and mucositis in an 8-year-old boy.

Mycoplasma pneumoniae is a common cause of community-acquired pneumonia (CAP) in children. While M. pneumoniae infection is typically mild and self-limiting, individuals of all ages may develop severe CAP or extrapulmonary manifestations such as M. pneumoniae-induced rash and mucositis (MIRM). Severe M. pneumoniae pneumonia with concurrent MIRM in an 8-year-old boy is reported. The clinical course was complicated by persistent fever, severe mucositis and bilateral multi-lobar pneumonia requiring advanced respiratory support. Initial antibiotic and supportive therapy yielded little improvement,…

Narrative review of the management of oral mucositis during chemoradiation for head and neck cancer.

Oral mucositis (OM) can be a significant problem for patients undergoing radiation or chemoradiation for head and neck cancer. In modern clinical trials, grade 3-4 OM can be seen in over 40% of patients and can cause a significant impact on their quality of life (QOL). Despite this fact, strategies for the prevention and treatment of OM vary widely, with options including both lifestyle modifications and pharmaceuticals. Here we evaluate and summarize the current clinical interventions for the management of radiation-induced OM. The majority of the current evidence focuses on reducing OM relat…
