The development and maintenance of drug addiction triggered by psychoactive substances, such as methamphetamine (Meth), is strongly influenced by synaptic and morphological adaptations in the brain. The hippocampus has a key role in the formation of maladaptive drug-context associations and relapse. However, the mechanisms regulating this complex process are not completely understood. Recent studies highlight that the actin cytoskeleton and its regulatory proteins play a fundamental role in morphological and behavioral plasticity associated with drug use. Here, we show that a binge pattern of

Traumatic brain injury (TBI) induces neuronal death, inflammation, and neurological dysfunction. Although nerve growth factor (NGF) possesses neuroprotective potential, its clinical use is hindered by poor blood-brain barrier (BBB) permeability and insufficient neural targeting. Here, a neurophilic biomimetic lipoprotein for brain-targeted NGF delivery is developed: 1) a matrix-like core (Nc) that preserves NGF bioactivity; 2) an ApoE3-reconstituted high-density lipoprotein shell (Nc-rHDL) to enhance BBB penetration; 3) an αRDP peptide-modified version (Nc-rHDL@P) to improve neural targeting.

The persistence of deleterious substances at lesion sites severely impedes neuronal survival and axonal regeneration following central nervous system (CNS) injury or in neurodegenerative disorders. Therefore, clearing these harmful components and establishing a regeneration-permissive microenvironment are crucial for functional recovery. In this study, primary olfactory ensheathing cells (OECs) isolated from adult SD rats were pharmacologically treated with fisetin, a pharmacological agent. To model CNS injury conditions, neural debris was generated through mechanical disruption of primary neu

The MELAS syndrome primarily affecting the CNS is mainly caused by the m.A3243G mutation. The heteroplasmy in different tissues affects the phenotypic spectrum, yet the impact of various levels of m.A3243G heteroplasmy on CNS remains elusive due to the lack of a proper neuronal model harboring m.A3243G mutation. We generated induced neurons (iNs) through the direct reprogramming of MELAS patients, with derived fibroblasts harboring high (>95%), intermediate (68%), and low (20%) m.A3243G mutation. iNs demonstrated neuronal morphology with neurite outgrowth, branching, and dendritic spines. Complex cortical malformations (CCMs), such as hemimegalencephaly and polymicrogyria, are associated with drug-resistant epilepsy and developmental impairment. They share certain neuropathological characteristics including mammalian target of rapamycin (mTOR) activation and an atypical number of white matter neurons. To get a better understanding of the pathobiology of the lesion architecture, we investigated the role of neurite outgrowth inhibitor A (NogoA), a known regulator of neuronal migration. Epilepsy surgery specimens from 16 CCM patients were analyzed and compared with sections of foc

Corticothalamic axons express Contactin-2 (CNTN2/TAG-1), a neuronal recognition molecule of the immunoglobulin superfamily involved in neurogenesis, neurite outgrowth, and fasciculation. TAG-1, which is expressed transiently by cortical pyramidal neurons during embryonic development, has been shown to be fundamental for axonal recognition, cellular migration, and neuronal proliferation in the developing cortex. Although Tag-1 -/- mice do not exhibit any obvious defects in the corticofugal system, the role of TAG-1+ neurons during the development of the cortex remains elusive. We have generated

UNLABELLED: Activation of extracellular signal-regulated protein kinase (ERK), a member of the mitogen-activated protein kinase family, has been shown to mediate neurite outgrowth-promoting effects of various neurotrophic factors in vitro. Moreover, in vivo, ERK is activated in the primary sensory neurons and associated glial cells after nerve injury. However, the precise role of ERK in nerve regeneration remains unclear.
OBJECTIVE: This work was aimed to investigate the effects of ERK inhibition on axonal regeneration and neuronal loss after axotomy.
METHODS: Unilateral sciatic nerve crush

Peripheral neurons respond to structural, biochemical, and electrical cues to regenerate following injury. However, how these cues contribute individually and in combination to promoting neurite outgrowth remains incompletely understood. Here, we developed an aligned collagen fiber-based scaffold to examine how combinations of extracellular matrix proteins contribute to neurite outgrowth with and without electrical stimulus. Type I collagen fibers were fabricated by contact-drawing, a dry spinning process capable of producing collagen fibers that can be twisted into multifilaments that support

Spinal cord injury (SCI) releases a cascade of events that leads to the onset of an inhibitory milieu for axonal regeneration. Some of these changes result from the presence of repulsive factors that may restrict axonal outgrowth after trauma. The Eph receptor tyrosine kinase (RTK) family has emerged as a key repellent cue known to be involved in neurite outgrowth, synapse formation, and axonal pathfinding during development. Given the nonpermissive environment for axonal regeneration after SCI, we questioned whether re-expression of one of these molecules occurs during regenerative failure. W

Regeneration in the peripheral nervous system is poor after chronic denervation. Denervated Schwann cells act as a "transient target" by secreting growth factors to promote regeneration of axons but lose this ability with chronic denervation. We discovered that the mRNA for pleiotrophin (PTN) was highly up-regulated in acutely denervated distal sciatic nerves, but high levels of PTN mRNA were not maintained in chronically denervated nerves. PTN protected spinal motor neurons against chronic excitotoxic injury and caused increased outgrowth of motor axons out of the spinal cord explants and for

The organization, function and evolution of the brain depends centrally on the precise development of a wide diversity of distinct neuronal subtypes. Furthermore, given the heterogeneity of neuronal subtypes within the CNS and the complexity of their connections, attempts to functionally repair circuitry will require a detailed understanding of the molecular controls over differentiation, connectivity and survival of specific lineages. Toward these goals, we recently identified developmentally regulated transcriptional programmes for specific lineages of long-distance neocortical projection ne

Activity-dependent structural plasticity is essential for the growth and remodeling of synaptic connections. At the Drosophila melanogaster larval neuromuscular junction (NMJ), spaced stimulation induces the translation-dependent formation of ghost boutons (GBs), which are immature boutons that lack a corresponding postsynaptic structure. Calcium/calmodulin-dependent protein kinase II (CaMKII) has previously been implicated in GB formation and is locally translated at synapses, raising the possibility that activity-dependent CaMKII synthesis contributes to GB formation. We examined activity-in

Perineural invasion (PNI) is associated with worse prognosis in various malignancies. Targeting PNI may hinder the tumor metastasis in head and neck squamous cell carcinoma (HNSCC) and pancreatic ductal adenocarcinoma (PDAC). We investigated the role of nuclear receptor subfamily 4 group A2 (NR4A2/Nurr1) mediated neuronal invasion in HNSCC and PDAC tumor progression. Aberrant expression of NR4A2 was observed in these tumors, and high levels of NR4A2 were associated with worse survival. The whole genome chromatin immunoprecipitation (ChIP) sequencing with NR4A2 antibody illustrated several gene

The skin is densely innervated by peripheral sensory neurons that detect various stimuli through specialized nerve endings in the dermis and epidermis. In recessive dystrophic epidermolysis bullosa (RDEB), repeated skin injury disrupts epidermal nerve fibers, leading to neuropathic pain and reduced thermal sensitivity. Normally, keratinocyte-derived neurotrophic signals guide sensory fiber re-entry into healed epidermis. We hypothesized that impaired neurotrophic support underlies failed reinnervation in RDEB. To investigate the mechanisms behind failed reinnervation, we assessed neurotrophic

Neurodegenerative disorders pose a major global health challenge due to impaired electrical signaling in nerve fibers, yet effective regenerative therapies remain limited. Here, we report an injectable hydrogel that integrates intrinsic conductivity, antioxidative capacity, and strain-stiffening mechanics, to emulate key features of the neural extracellular matrix (ECM) and promote neuronal differentiation. We design peptide amphiphiles (Fcn, n = 5, 10) by anchoring a redox-active ferrocene (Fc) motif to a self-assembling peptide segment (NVFFAKKC) via methylene spacers (n). These nanofibers a

Chronic hepatitis B virus (HBV) infection remains a major global health burden. Despite the widespread availability of effective prophylactic vaccines, millions of people are at risk for complications of chronic HBV infection. Nucleoside polymerase inhibitors are highly effective at suppressing the production of infectious viral particles and decreasing the risk of cirrhosis; however, viral clearance remains very rare, which means that treatment is often lifelong. Alternative therapeutic agents with a new mode of action giving sustained activity of treatment are needed. We report a new thiochr

Adult hippocampal neurogenesis plays an important role in health and disease. Regulating neurogenesis may be a key mechanism in the pathophysiology and treatment of several neurobehavioral disorders such as schizophrenia, depression, autism spectrum disorders and Alzheimer's disease. Cytokines are known to affect adult neurogenesis, but conflicting studies have been reported with regard to their actual role. Interleukine-17 (IL-17), a potent pro-inflammatory cytokine, has been shown to inhibit proliferation of neuroprogenitors and thus reduce hippocampal neurogenesis, while other studies sugge

Familial dysautonomia (FD) is a sensory and autonomic neuropathy caused by mutations in elongator complex protein 1 (ELP1). FD patients have small trigeminal nerves and impaired facial pain and temperature perception. These signals are relayed by nociceptive neurons in the trigeminal ganglion, a structure that is composed of both neural crest- and placode-derived cells. Mice lacking Elp1 in neural crest derivatives ('Elp1 CKO') are born with small trigeminal ganglia, suggesting Elp1 is important for trigeminal ganglion development, yet the function of Elp1 in this context is unknown. We demons

In this study, serotonin (5-hydroxytryptamine; 5HT)-immunoreactive (5HT-IR) neuronal fibers were identified in the primary olfactory pathway of the sea lamprey. These neurons are likely part of a nonolfactory neural system that innervates the olfactory sac. Cell bodies with 5HT immunoreactivity predominated in the lamina propria of the rostral portion of the nasal cavity and were less prevalent adjacent to the olfactory epithelium. The 5HT-IR fibers were parallel to axons of the olfactory receptor neurons in the lamina propria of the olfactory mucosa and in the olfactory nerve. Serotonergic fi
