A toxin-like gene in rainbow trout: cloning, expression, and gene organization.

The 'three-finger' protein family consists of structurally related proteins which display a variety of biological functions. Snake venom alpha-neurotoxins, Ly-6 alloantigens, urokinase-type plasminogen activator receptor (uPAR) and the complement regulatory protein CD59 are members of this protein family. Their characteristic feature is the disposition of four disulfide bridges forming a hydrophobic core. Here it is reported the cloning and characterization of a toxin-like gene (toxin-1) in the rainbow trout (Oncorhynchus mykiss). Phylogenetic analysis showed that trout toxin-1 classified into…

Secretion of methoxyindoles from trout pineal organs in vitro: indication for a paracrine melatonin feedback.

Synthesis and release of the pineal hormone melatonin is in all vertebrates primarily regulated by the light/dark cycle. In pineal organs of teleost fish, like in other non-mammalian vertebrates, melatonin formation is regulated by a direct photoreception of the pineal organ. We performed measurements in explanted, perifused pineal organs of the rainbow trout, Oncorhynchus mykiss, to examine whether melatonin can influence its own production. For this purpose we have continuously perifused isolated pineal organs under light- and dark-adapted conditions and measured the release of melatonin and…

Cytotoxic and endocrine-disrupting potential of atrazine, diazinon, endosulfan, and mancozeb in adrenocortical steroidogenic cells of rainbow trout exposed in vitro.

An in vitro bioassay for detection and quantitative assessment of chemicals with the capacity to disrupt adrenal steroidogenesis has been developed and used to compare the cytotoxic and endocrine-disrupting potential of four pesticides. Enzymatically dispersed adrenocortical cells of rainbow trout (Oncorhynchus mykiss) were exposed in vitro to atrazine, diazinon, endosulfan, and mancozeb, and cortisol secretion in response to ACTH or dibutyryl-cAMP (dbcAMP) and cell viability were determined. The effective concentration, EC50 (concentration that inhibits cortisol secretion by 50%), the median …

Action of gamma-aminobutyric acid (GABA) in the isolated photosensory pineal organ.

The effect of GABA (gamma-aminobutyric acid), its agonists (muscimol, baclofen) and antagonist (bicuculline) on pineal ganglion cells of the luminosity type were studied in the isolated, superfused pineal organ of the rainbow trout, Oncorhynchus mykiss. Extracellular recordings revealed that GABA added through the superfusion medium caused a clear alteration of the activity of projecting neurons, which transmit luminosity responses to the brain. Spontaneous discharges of ganglion cells were predominantly suppressed by GABA (33 neurons out of 48), but 10 neurons were clearly excited. Similar ef…

Fipronil contamination impairs cardiorespiratory control of rainbow trout during aerobic exercise.

Fipronil is a widely detected phenylpyrazole insecticide in aquatic ecosystems that acts as a GABA receptor antagonist in arthropods, but its effects on vertebrate cardiorespiratory physiology are not well understood. In this study, we investigated whether environmentally relevant concentrations of fipronil (0.5 μg l-1, 96 h) impair the cardiovascular and ventilatory responses of rainbow trout (Oncorhynchus mykiss) during aerobic exercise. Using dorsal aortic and buccal cannulations, we continuously recorded heart rate (fH), mean arterial pressure (MAP), ventilatory rate (fV) and amplitude (VA…

The telencephalon is a neuronal substrate for systemic inflammatory responses in teleosts via polyamine metabolism.

Systemic inflammation elicits sickness behaviors and fever by engaging a complex neuronal circuitry that begins in the preoptic area of the hypothalamus. Ectotherms such as teleost fish display sickness behaviors in response to infection or inflammation, seeking warmer temperatures to enhance survival via behavioral fever responses. To date, the hypothalamus is the only brain region implicated in sickness behaviors and behavioral fever in teleosts. Yet, the complexity of neurobehavioral manifestations underlying sickness responses in teleosts suggests engagement of higher processing areas of t

Behavioural traits of rainbow trout and brown trout may help explain their differing invasion success and impacts.

Animal behaviour is increasingly recognised as critical to the prediction of non-native species success and impacts. Rainbow trout and brown trout have been introduced globally, but there appear to be differences in their patterns of invasiveness and ecological impact. Here, we investigated whether diploid rainbow trout and diploid and triploid brown trout differ among several key behavioural measures linked to invasiveness and impact. We assessed activity, boldness, aggression, and feeding, using open field, novel object, shelter, mirror, feeding, and functional response experiments. We also

Dermal injuries caused by purse seine capture result in lasting physiological disturbances in coho salmon.

Fish vitality can be measured by classifying reflex impairments (i.e., a visual impression of the ability to respond to induced stimuli) and visible injuries. These metrics can predict survival probability following release from fisheries, and monitoring physiological disturbances following capture can help understand mechanisms of mortality. To test the hypothesis that severity of injury and reflex impairment influences the time course of physiological recovery, coho salmon (Oncorhynchus kisutch) were held for up to 84-h following capture by purse seine. We classified reflex impairments and v…

Sodium chloride treatment effects on rainbow trout suffering from proliferative kidney disease caused by Tetracapsuloides bryosalmonae.

The aim of the present study was to evaluate the effect of a long-term sodium chloride bath on rainbow trout Oncorhynchus mykiss naturally infected by Tetracapsuloides bryosalmonae. A total of 106 infected fish were divided into 2 groups. One group was left untreated and the other was treated with sodium chloride in increasing doses up a concentration of 0.8%. After 14 d, treatment was stopped and for a further 7 d the fish response to the sodium chloride bath was observed. Cumulative mortality was significantly lower in the treated group (19.2%) compared to the untreated group (31.5%) after 2…

Water supply and feed as sources of antimicrobial-resistant Enterococcus spp. in aquacultures of rainbow trout (Oncorhyncus mykiss), Portugal.

The role of European fish farms in the spread of antimicrobial-resistance in the environment and food chain, as well as possible sources of their contamination by clinically relevant antimicrobial-resistance bacteria is scarcely known. This study aimed to assess the contribution of Portuguese rural trout farms on dispersion of Enterococcus with antimicrobial-resistance and putative virulence genes in the environment and food chain, as well as to identify farms contamination sources. We also assessed the presence of Enterococcus with low-levels of antimicrobial-resistance using epidemiological

Viral haemorrhagic septicaemia virus (VHSV Id) infections are detected more consistently using syndromic vs. active surveillance.

The eradication of viral haemorrhagic septicaemia virus (VHSV Id) from Finnish brackish-water rainbow trout Oncorhynchus mykiss farms located in the restriction zone in the Province of Åland, Baltic Sea, failed several times in the 2000s. The official surveillance programme was often unable to find VHSV-positive populations, leading to the misbelief in the fish farming industry that virus eradication could be achieved. The ability of 3 other surveillance programmes to detect infected fish populations was compared with the official programme. One programme involved syndromic surveillance based …

A matter of potential concern: natural organic matter alters the electrical properties of fish gills.

Natural organic matter (NOM) is an important constituent of aquatic environments; however, its influence on aquatic biota remains poorly studied. In the current study, NOM was isolated from nine different sites in southern Ontario, Canada, by the on-site treatment of water by reverse osmosis, followed by cation exchange. NOM from each site was reconstituted to 10 mg of C/L and pH 7.0 and exposed to either adult rainbow trout implanted with indwelling catheters or to in vitro primary cultures of the gill epithelium grown on semipermeable membranes. In both the in vivo and in vitro preparations,

Apolipoprotein AI could be a significant determinant of epithelial integrity in rainbow trout gill cell cultures: a study in functional proteomics.

The freshwater fish gill forms a barrier against an external hypotonic environment. By culturing rainbow trout gill cells on permeable supports, as intact epithelia, this study investigates barrier property mechanisms. Under symmetrical conditions the apical and basolateral epithelial surfaces contact cell culture media. Replacing apical media with water, to generate asymmetrical conditions (i.e. the situation encountered by the freshwater gill), rapidly increases transepithelial resistance (TER). Proteomic analysis revealed that this is associated with enhanced expression of pre-apolipoprotei

Integron-containing IncU R plasmids pRAS1 and pAr-32 from the fish pathogen Aeromonas salmonicida.

A 45-kb R plasmid, pRAS1, that confers resistance to tetracyclines, trimethoprim, and sulfonamides was isolated in 1989 from an atypical strain of the fish pathogen Aeromonas salmonicida. This plasmid could be transferred by conjugation to Escherichia coli with a high degree of efficiency (frequency, 0.48). The following year pRAS1 was isolated from A. salmonicida subsp. salmonicida in the same area. Incompatibility group U plasmid pRAS1 contained a drug resistance-determining region of 12 kb consisting of a class 1 integron similar to In4 of Tn1696 but with a dfrA16 gene cassette inserted. Cl

Hierarchies of ATP-consuming processes: direct compared with indirect measurements, and comparative aspects.

The original aim of the present study was to deal with two problems that had emerged from a study on hierarchies of ATP-consuming processes in cells [Buttgereit and Brand (1995) Biochem. J. 312, 163-167]. Firstly, we wanted to find out whether the results of that study had been influenced by the method used for the determination of process activity and, secondly, we wondered whether and to what extent the structure of the hierarchy established for cell suspensions under energy-limiting conditions might depend on the type of cell or on the lifestyle, ecology and phylogenetic status of the speci
