The human GNAS1 gene is imprinted and encodes distinct paternally and biallelically expressed G proteins.

The GNAS1 gene encodes the alpha subunit of the G protein Gs, which couples receptor binding by several hormones to activation of adenylate cyclase. Null mutations of GNAS1 cause pseudohypoparathyroidism (PHP) type Ia, in which hormone resistance occurs in association with a characteristic osteodystrophy. The observation that PHP Ia almost always is inherited maternally has led to the suggestion that GNAS1 may be an imprinted gene. Here, we show that, although Gsalpha expression (directed by the promoter upstream of exon 1) is biallelic, GNAS1 is indeed imprinted in a promoter-specific fashion

Correlations among hormone levels, sperm parameters and paternity in formerly unilaterally cryptorchid men.

PURPOSE: We compared characteristics related to fertility between formerly unilaterally cryptorchid men. MATERIALS AND METHODS: We compared sperm counts and gonadotropin levels before and after gonadotropin-releasing hormone stimulation between formerly unilaterally cryptorchid men and controls who had completed a detailed questionnaire on fertility and other pertinent paternity information. These parameters were also compared between the subsets of formerly cryptorchid men who reported paternity and unsuccessful attempts at paternity. RESULTS: Sperm density and total count, and basal and go

Loss of imprinted genes and paternal SUR1 mutations lead to hyperinsulinism in focal adenomatous hyperplasia.

Two types of histopathological lesions, a focal adenomatous hyperplasia of islet cells of the pancreas in about 30% of operated sporadic cases, and a diffuse form can be observed in congenital hyperinsulinism, or Persistent Hyperinsulinemic Hypoglycemia of Infancy (PHHI). In sporadic focal forms, specific losses of maternal alleles (LOH) of the imprinted chromosomal region 11p15, restricted to the hyperplastic area of the pancreas, were observed. Similar mechanisms are observed in embryonal tumors and in the Beckwith-Wiedemann syndrome which is also associated with neonatal but transient hyper

Sociosexual behaviour and paternity in procarbazine-exposed rats with or without regional testicular circulatory isolation.

Male Sprague-Dawley rats were used in two experiments in which a procarbazine bolus (400 mg kg-1 body mass) was administered with or without testicular circulatory isolation in the form of brief clamping of the spermatic cord and gubernaculum during drug administration. Separate tests of aggressiveness, sexual motivation, copulatory performance and paternity over the subsequent 6 weeks were used to assess functional changes resulting from testicular circulatory isolation. Experiment 1 compared intermale aggression and sexual motivation of animals in groups receiving procarbazine plus testicula

Effects of deletions on mitotic stability of the paternal-sex-ratio (PSR) chromosome from Nasonia.

Paternal-Sex-Ratio (PSR) is a B chromosome that causes all-male offspring in the parasitoid wasp Nasonia vitripennis. It is only transmitted via sperm of carrier males and destroys the other paternal chromosomes during the first mitotic division of the fertilized egg. Because of haplodiploidy, the effect of PSR is to convert diploid (female) eggs into haploid eggs that develop into PSR-bearing males. The PSR chromosome was previously found to contain several families of repetitive DNA, which appear to be present in local blocks. PSR chromosomes with irradiation-induced deletions have decreased

Association of decreased paternal age and late-onset Alzheimer's disease. An example of genetic imprinting?

Several studies have identified advanced maternal age as a risk factor for Alzheimer's disease. This study evaluated maternal and paternal age at birth of 237 patients with Alzheimer's disease, each of whom was matched to five control subjects based on sex, year of birth, survival age, and location of residence. It was found that decreased paternal age substantially increased the susceptibility to the common form of Alzheimer's disease that occurs after the age of 67 years, whereas advanced maternal age had a negligible effect on risk of Alzheimer's disease. The higher incidence of late-onset

A 48,XXY,+21 Down syndrome patient with additional paternal X and maternal 21.

The origin of meiotic nondisjunction of the extra chromosomes X and 21 was studied in a patient with the karyotype 48,XXY,+21 using DNA polymorphisms. The extra chromosome X was the result of paternal first meiotic nondisjunction of X and Y. The extra chromosome 21 was derived from the mother. The meiotic error in the mother most probably occurred in meiosis II. Thus, this is a combination caused by the chance occurrence of two independent events.

Trophoblast from anembryonic pregnancy has both a maternal and a paternal contribution to its genome.

In a series of 14 anembryonic pregnancies, deoxyribonucleic acid from trophoblast was examined with locus-specific minisatellite probes. It was found in each case that trophoblast from anembryonic pregnancy has both a maternal and a paternal contribution to its genome. This means that although anembryonic pregnancy shares characteristics with complete hydatidiform mole and androgenetic development in the mouse in that there is no embryo, it does not arise by the same genetic process. Of three anembryonic pregnancies that were successfully karyotyped, two had normal female 46,XX chromosome cons

Paternal non-disjunction in a 46,XY/47,XXY individual with a fragile 17p12 in the mother.

In a family where the mother carried a fragile site at 17p12, RFLP-analysis with the X-specific probe L1.28 showed that the 46,XY/47,XXY mosaicism detected in her Klinefelter son was due to a non-disjunctional event in paternal meiosis I, followed by a secondary loss of an X-chromosome by a mitotic non-disjunction. Thus, an association between the primary meiotic non-disjunction and the presence of the fragile site could be excluded.

Evidence of a preferential inactivation of the paternally derived X chromosome in a 46,XX true hermaphrodite.

The pattern of inheritance of several X polymorphic markers is studied in the pedigree of a 46,XX true hermaphrodite. The results of the Xga, 12E7, and G6PD segregation analysis favour the hypothesis of a preferential inactivation of the paternally derived X chromosome.

Selective expression of paternal human major histocompatibility antigens on the surface of hydatidiform mole cells.

The typing of human major histocompatibility antigens (HLA) of two cases of invasive hydatidiform mole showed that the fibroblast-like cells as well as trophoblasts from the mole selectively expressed paternal HLA haplotype specificity but not maternal HLA on the surface. This result was completely in agreement with the notion of androgenetic origin of hydatidiform mole from the aspects of HLA specificity. Immunological implication of this finding and development of chorionic tumor were discussed in the light of fetomaternal relationship and host immune surveillance against the tumor.

Advancing paternal perinatal mental health: Integrating qualitative evidence, theory, and measurement science.

AIM: To advance understanding of paternal perinatal mental health through the integration of qualitative evidence, mid-range theory, and conceptual framework development. BACKGROUND: Existing scholarship largely emphasizes prevalence and symptom identification of paternal postpartum depression, with limited integration of fathers lived experiences into theory development and assessment frameworks, and mental health nursing practice. Greater integration of qualitative evidence, theory, and measurement science may strengthen the recognition and assessment of paternal distress. METHODS & RESULT

Advanced paternal age affects miscarriages and live birth outcomes in oocyte donation cycles.

STUDY QUESTION: Does advanced paternal age affect miscarriage and/or live birth outcomes following the first transfer attempt in oocyte donation cycles? SUMMARY ANSWER: Paternal age has a gradual but cumulative effect that becomes apparent above 45 years of age, where it evidently increases miscarriage rates and decreases live birth rates following the first transfer in oocyte donation cycles. WHAT IS ALREADY KNOWN: Donor oocyte cycles are becoming a common strategy for addressing female age-related infertility, offering high success rates (∼60% ongoing pregnancies) attributed to superior oo

Paternal metabolic dysfunction and pregnancy loss following assisted reproduction: A cross-sectional study.

AIMS: Diabetes mellitus often causes anastomotic leakage leading to severe complications after surgery. The purpose of this study was to determine whether collagen synthesis is correlated with the anastomotic strength in diabetic animals in special reference to collagen type differences. METHODS: Thirty Lewis rats weighing approximately 250 g were made diabetic by tail vein injection of streptozotocin (STZ). Laparotomy was performed through a midline incision under ether anesthesia. A colonic segment 5 cm distal to the ileocecal junction was transected then anastomosed with absorbable suture.

Paternal use of metformin and risk of major congenital malformations: A meta-analysis of 4 studies.

Metformin is the first-line drug for the treatment of Type 2 diabetes. In 2022, a Danish registry-based study reported a 40% increased risk of major congenital malformations following paternal exposure during spermatogenesis. This raised widespread concern about the use of metformin in males of reproductive age. Three subsequent studies were unable to replicate this finding. We performed a meta-analysis of four observational studies with data from five countries covering more than 11 000 live-born children whose fathers used metformin during spermatogenesis. The pooled risk ratio from an inver

Procreation, Potency, and the Price of Paternity: The Social and Psychological Impact of a Diagnosis of Male Factor Infertility.

Male factor infertility accounts for roughly half of all infertility cases, yet receives far less clinical, research, and public attention than female infertility. Historical, cultural, and medical factors, particularly the absence of comparable therapeutic advances, contribute to this neglect. A diagnosis can threaten identity and masculinity, leading to depression, anxiety, and sexual dysfunction, while stigma and cultural taboos around semen collection compound distress. Men often feel excluded from care and remain socially invisible. Applying patient-centered frameworks such as the 4 "I" m

Tfcp2l1 participates in paternal vitamin D deficiency-induced fetal lung maldevelopment in mice.

BACKGROUND: Fetal lung development is crucial for neonatal respiratory function, with insufficient maturation contributing to neonatal respiratory distress syndrome. While maternal vitamin D deficiency (VDD) has been linked to adverse offspring outcomes, the impact of paternal VDD on fetal lung programming remains largely unexplored. This study investigates the effects of paternal VDD on fetal lung development and maturation, aiming to provide a theoretical basis for paternal preconception nutritional intervention and highlight the importance of paternal health in offspring respiratory outcome

The involvement of m6A RNA methylation in the transgenerational inheritance of hepatotoxicity induced by paternal cadmium exposure.

Cadmium (Cd), a widespread environmental pollutant, has linked with various adverse health effects. However, the paternally inherited transgenerational effects of cadmium exposure on hepatotoxicity inoffspring and the underlying mechanism remained undefined. To address this issue, male mice (F0) were administrated with 100 mg/L cadmium chloride (CdCl2) in drinking water for 3 months. Then, F0 males were mated with healthy female mice to produce F1 and F2 offspring.Liver function was assessed at age of 6 weeks and 6 months, respectively. Herein, we showed ancestral cadmium exposure led to liver

Preconception paternal exposure to zearalenone impairs male fertility and IVF embryo survival in mice.

Zearalenone (ZEA), a fungal mycotoxin commonly found in various human foods, exhibits a structural similarity to naturally occurring estrogen, raising significant concerns regarding its reproductive toxicity. This study aimed to assess the impact of preconceptional ZEA exposure on male reproductive health and early embryonic development. Healthy B6D2F1 male mice were exposed to corn oil or varying doses of ZEA for two weeks. Our findings revealed that ZEA exposure caused a notable reduction in sperm count and motility, accompanied by significant weight loss in the high-dose group. Additionally

A novel model of central precocious puberty disease: Paternal MKRN3 gene-modified rabbit.

BACKGROUND: Makorin ring finger protein 3 gene (MKRN3) gene mutation is the most common genetic cause of central precocious puberty (CPP) in children. Due to the lack of ideal MKRN3-modified animal model (MKRN3-modified mice enter puberty only 4-5 days earlier than normal mice), the related research is limited. METHODS: Therefore, the MKRN3-modified rabbit was developed using CRISPR (clustered regularly interspaced short palindromic repeats) gene editing technology. The genotype identification and phenotype evaluation of MKRN3-modified rabbits were carried out. RESULTS: The first estrus of M
