Complementary enhancement of osmotic tolerance in Ligilactobacillus salivarius AR612 by exogenous purine nucleobases and gene engineering.

High-density fermentation and high-sugar environments place substantial osmotic stress on the probiotic strain Ligilactobacillus salivarius, causing its viability to be insufficient for food industrial applications, biological preservation, and probiotic formulation development. We investigated the efficacy of exogenous nucleobases and genetic engineering for enhancing the hyperosmotic tolerance of L. salivarius AR612. A thorough investigation using physiological assessments, molecular cloning, and metabolomics demonstrated that supplementation with purine nucleobases, particularly adenine, en

XYLB promotes malignant behaviors of triple-negative breast cancer via SUSD3 upregulation.

Triple-negative breast cancer (TNBC) remains a clinically challenging breast cancer subtype because of its molecular heterogeneity, early recurrence, metastatic propensity, and limited targetable drivers. Metabolic remodeling is increasingly recognized as a determinant of TNBC progression, but the contribution of pentose phosphate pathway-related enzymes to TNBC remains incompletely defined. Here, we investigated the biological role of xylulokinase (XYLB), an enzyme that converts D-xylulose to xylulose-5-phosphate (Xu5P), and its downstream mechanisms in TNBC. We found that XYLB was enriched i

PPAR-δ is a potential modulator of central carbon metabolism in human adipocytes.

Adipogenesis involves adipocyte differentiation and synthesis and storage of fats. PPAR-γ is the master regulator of adipogenesis and regulates genes for adipocyte differentiation, lipogenesis, adipocyte survival and adipokine secretion. Central carbon metabolism (CCM) comprises of three key pathways glycolysis, tricarboxylic acid (TCA) cycle and pentose phosphate pathway (PPP). CCM utilizes carbon sources to provide energy and building blocks for lipogenesis. Several targets of transcription factor PPAR-γ have been identified in CCM pathways. However, the exact mechanism of PPAR-γ modulation

Immunometabolic control of macrophage plasticity in wound healing: mechanistic insights and therapeutic opportunities.

Macrophages play a pivotal regulatory role in inflammation, tissue repair, and fibrosis through their dynamic changes in phenotype and function. The tissue microenvironment following injury induces alterations in key metabolic enzymes, signaling pathways, and metabolites within macrophages, thereby driving shifts in their phenotype and function. Early in acute injury, macrophages primarily rely on glycolysis and the pentose phosphate pathway, transitioning to a pro-inflammatory phenotype. Persistent activation of pro-inflammatory macrophages can lead to tissue damage. As the metabolic microenv

Tumor Microenvironment-Responsive Nanoreactor Induces Disulfidptosis in Pancreatic Cancer via Metabolic Interference and Redox Catalysis.

Pancreatic cancer exhibits extensive metabolic reprogramming that supports rapid progression and therapeutic resistance, making metabolic vulnerabilities attractive targets for intervention. Here, a tumor microenvironment-responsive nanoreactor (Pht@HMnO2-HA) is designed to induce disulfidptosis in pancreatic cancer through coordinated metabolic interference and redox catalysis. Following CD44-mediated tumor targeting and cellular internalization, the nanoreactor's responsive self-optimization within the tumor microenvironment enables localized release of phloretin, suppressing glucose uptake

Integrated Photoacoustic-Metabolomic Platform for Multimodal Assessment of Sepsis-Induced Brain Dysfunction.

Sepsis-induced brain dysfunction (SIBD), a critical determinant of mortality and long-term neurological sequelae in sepsis patients. The mechanistic understanding of SIBD has been limited by conventional single-modality approaches, which fail to capture the complex oxygenation-metabolism interplay. Here, we present an integrated photoacoustic-metabolomic platform that combines high-resolution photoacoustic imaging with targeted metabolomics to comprehensively assess cerebral oxygenation and metabolic alterations during sepsis. Our imaging system provides high spatiotemporal resolution, enablin

Cellular Metabolomics Reveals Hepatoprotective Mechanisms of Platycoside Metabolites Against Acetaminophen-Induced Liver Injury.

Elucidating mechanisms of natural product metabolites is key to safer, more effective drugs against drug-induced liver injury (DILI). This study used cellular metabolomics to investigate hepatoprotective effects of three structurally similar platycoside metabolites-platycodigenin (PDN), 3-O-β-D-glucopyranosyl platycodigenin (OPDN), and polygalacic acid (POLA)-against APAP-induced liver injury. Using AML-12 hepatocytes challenged with APAP, intracellular metabolites were profiled by UHPLC-LTQ-Orbitrap mass spectrometry (ESI+ and ESI-), followed by PCA/OPLS-DA to identify differential metabolite

NOS-derived NO modulates photosynthetic integrity, carbon metabolism and cellular homeostasis in the cyanobacterium Aphanizomenon flos-aquae under methyl viologen stress.

Photosynthetic organisms often face environmental challenges that disrupt cellular homeostasis. Of the various protective mechanisms, nitric oxide (NO) is a crucial signaling molecule with diverse regulatory functions. In mammalian cells, NO is enzymatically synthesized by nitric oxide synthase (NOS) and regulates several physiological processes such as neuronal differentiation, vascular relaxation, synaptic signaling, and immune regulation. Although recent studies have reported the presence of canonical NOS homologs in photosynthetic microorganisms, their function is yet to be elucidated. Hen

The Role of Glucose Metabolism in Xenopus Primitive Myeloid Cell Development and Function.

Primitive Myeloid Cells (PMCs) arise from the anterior ventral blood islands during Xenopus laevis embryogenesis and migrate to colonize embryonic tissues, contributing to wound healing, innate immunity, and development. Because cell movement and growth are metabolically demanding, we investigated whether metabolic pathways regulate PMC behaviour integrating public single-cell RNA-seq data with whole-mount in situ hybridization (WMISH) to profile metabolic gene expression in PMCs, followed by functional assays using pathway-specific inhibitors: 2-deoxy-D-glucose 2-DG (global glucose metabolism

Engineering of central metabolism for high-yield 1,3-propanediol production using co-substrates by Klebsiella pneumoniae.

1,3-Propanediol is a versatile C3 diol serving as a key building block used in various industries. Klebsiella pneumoniae represents an attractive microbial chassis for 1,3-PDO production due to its endogenous 1,3-PDO and Vitamin B₁₂ biosynthetic pathways. However, low carbon flux and by-product formation remain bottlenecks to high-efficiency 1,3-PDO production. Here, a novel co-substrate (glucose-glycerol) strategy was reported to address these limitations. First, the glycerol oxidation and 3-hydroxypropionic acid biosynthetic pathways were eliminated, driving glycerol to serve exclusively as

Disrupting Leishmania redox homeostasis: Mechanistic insights into a platinum-based antileishmanial complex.

Leishmaniasis remains a major neglected tropical disease, and the development of new chemotherapeutic agents with improved selectivity and novel mechanisms is urgently required. Since the discovery of cisplatin, platinum-based compounds have been extensively developed for cancer therapy; however, their potential in the treatment of parasitic infections has received limited attention. Here, we report a mechanistic investigation of the antileishmanial activity of the platinum(II) complex [PtCl(phpy)(PTA)], incorporating the oxidation-resistant phosphine ligand 1,3,5-triaza-7-phosphaadamantane. Peptides from plants and microorganisms: a defense strategy for biotic and abiotic stresses.

Antimicrobial peptides (AMPs) from plants and microorganisms have emerged as promising tools due to their multifunctional roles in plant defense. These small, bioactive molecules, such as thionins, systemins, defensins, cyclotides, hevein-like peptides, and cyclic dipeptides, exhibit broad-spectrum activity against fungal pathogens, bacteria, and insect pests. Recent studies have further elucidated their supportive roles in conferring tolerance to abiotic stresses, including salinity, drought, and heavy metals exposure, thus expanding their potential applications. Previous studies demonstrated

How denitrifiers defense ciprofloxacin: Insights from intracellular and extracellular stress response.

Overuse of antibiotics has led to their existence in nitrogen-containing water. The impacts of antibiotics on bio-denitrification and the metabolic response of denitrifiers to antibiotics are unclear. We systematically analyzed the effect of ciprofloxacin (CIP) on bio-denitrification and found that 5 mg/L CIP greatly inhibited denitrification with a model denitrifier (Paracoccus denitrificans). Nitrate reduction decreased by 32.89 % and nitrous oxide emission increased by 75.53 %. The balance analysis of carbon and nitrogen metabolism during denitrification showed that CIP exposure blocked ele

What exactly does the PfK13 C580Y mutation in Plasmodium falciparum influence?

BACKGROUND: The emergence and spread of artemisinin resistance threaten global malaria control and elimination goals, and encourage research on the mechanisms of drug resistance in malaria parasites. Mutations in Plasmodium falciparum Kelch 13 (PfK13) protein are associated with artemisinin resistance, but the unique or common mechanism which results in this resistance is unclear. METHODS: We analyzed the effects of the PfK13 mutation on the transcriptome and proteome of P. falciparum at different developmental stages. Additionally, the number of merozoites, hemozoin amount, and growth of P.

Differential roles of highly expressed PFKFB4 in colon adenocarcinoma patients.

Colon adenocarcinoma (COAD) is a common malignant tumor, and the role of the protein PFKFB4 in glycolysis and pentose phosphate pathways is crucial. Researchers investigated the clinical significance of PFKFB4 in COAD by studying its expression in 79 tissue samples using immunohistochemistry. We found that PFKFB4 expression was significantly higher in COAD patients, particularly in the sigmoid colon. Interestingly, high PFKFB4 expression was associated with both improved overall survival (OS) and worse progression-free survival (PPS) in COAD patients. Further analysis revealed that genes assoc

Photocatalysis of carbamazepine via activating bisulfite by ultraviolet: Performance, transformation mechanism, and residual toxicity assessment of intermediates products.

Carbamazepine (CBZ) as an extensively distributed emerging pollutant has menaced ecological security. The degradation performance of CBZ by UV driven bisulfite process was investigated in this work. The kinetics results indicated that CBZ was high-efficiently degraded by UV/bisulfite following a pseudo first-order kinetic model (Kobs = 0.0925 min-1). SO4•- and •OH were verified as the reactive oxidants by EPR test and the radicals scavenging experiment using MeOH and TBA. SO4•- played a dominant role for CBZ degradation. The Density functional theory (DFT) and LC-qTOF-MS/MS clarified that hydr

Metabolomic evolution of the postpartum dairy cow uterus.

High rates of early pregnancy loss are a critical issue in dairy herds, particularly in seasonal, grazing systems. Components of the uterine luminal fluid (ULF), on which the early embryo depends for sustenance and growth, partly determine early pregnancy losses. Here, changes in ULF from early to mid-postpartum in crossbred dairy cows were explored, linking them with divergent embryo development. For this, the uteri of 87 cows at Day 7 of pregnancy at first and third estrus postpartum were flushed to collect ULF. Eighteen metabolites (chiefly organic acids and sugars) significantly varied in

An Optimized Dihydrodibenzothiazepine Lead Compound (SBI-0797750) as a Potent and Selective Inhibitor of Plasmodium falciparum and P. vivax Glucose 6-Phosphate Dehydrogenase 6-Phosphogluconolactonase.

In Plasmodium, the first two and rate-limiting enzymes of the pentose phosphate pathway, glucose 6-phosphate dehydrogenase (G6PD) and the 6-phosphogluconolactonase, are bifunctionally fused to a unique enzyme named GluPho, differing structurally and mechanistically from the respective human orthologs. Consistent with the enzyme's essentiality for malaria parasite proliferation and propagation, human G6PD deficiency has immense impact on protection against severe malaria, making PfGluPho an attractive antimalarial drug target. Herein we report on the optimized lead compound N-(((2R,4S)-1-cyclob

Jejunal mucosa proteomics unravel metabolic adaptive processes to mild chronic heat stress in dairy cows.

Climate change affects the duration and intensity of heat waves during summer months and jeopardizes animal health and welfare. High ambient temperatures cause heat stress in dairy cows resulting in a reduction of milk yield, feed intake, and alterations in gut barrier function. The objectives of this study were to investigate the mucosal amino acid, glucose and lactate metabolism, as well as the proteomic response of the small intestine in heat stressed (HS) Holstein dairy cows. Cows of the HS group (n = 5) were exposed for 4 days to 28 °C (THI = 76) in a climate chamber. Percentage decrease

Toxic effects of copper on the jejunum and colon of pigs: mechanisms related to gut barrier dysfunction and inflammation influenced by the gut microbiota.

Copper (Cu) is an essential trace mineral, but its excessive intake can lead to potentially toxic effects on host physiology. The mammalian intestine harbors various microorganisms that are associated with intestinal barrier function and inflammation. In this study, the influences of Cu on barrier function, microbiota, and its metabolites were examined in the jejunum and colon of pigs. Here, we identified that the physical and chemical barrier functions were impaired both in the jejunum and colon, as evidenced by the decreased expression of tight junction proteins (ZO-1, Occludin, Claudin-1, a
