Trichloroethylene (TRI) administered to mice by gavage for 10 consecutive days at doses of 50-2000 mg/kg body weight elicited dose-dependent increases (up to 700% of control values) of hepatic cyanide insensitive palmitoyl CoA oxidation (a marker of peroxisomal beta-oxidation). No effect was seen on catalase; the other peroxisomal marker examined. Similar experiments with rats demonstrated no effect of TRI on either cyanide insensitive palmitoyl CoA oxidation or catalase. A major metabolite of TRI, trichloroacetic acid (TCA) when administered by gavage for 10 consecutive days at doses of 10-20

The effects of feeding 2,4-dichlorophenoxyacetic acid (2,4-D) and 2,4,5-trichlorophenoxyacetic acid (2,4,5-T) on the level of peroxisomal enzymes in rat liver were studied. The concentration of triglyceride in serum was decreased and the activity of cyanide-insensitive palmitoyl-CoA oxidation, catalase and carnitine acetyltransferase increased. However, the extent of the increase in the activity of these enzymes by treatment with 2,4-D was less pronounced than that by 2,4,5-T treatment. The administration of 2,4-D or 2,4,5-T increased the concentration of polypeptide with a mol. wt of 80,000 i

A boy with ornithine transcarbamylase (OTC) deficiency was relatively symptom free for 9 months and then developed an acute episode with liver failure and metabolic imbalance. Subsequently there was severe cerebral atrophy. Liver ornithine transcarbamylase activity was 3% of the normal mean. Of considerable interest was the finding of an accelerated breakdown of liver peroxisomes during the acute phase.

Long-term administration of some antiepileptic drugs often increases blood lipid levels. In this study, we investigated its molecular mechanism by focusing on the nuclear receptors constitutive active/androstane receptor (CAR) and peroxisome proliferator-activated receptor α (PPARα), which are key transcription factors for enzyme induction and lipid metabolism, respectively, in the liver. Treatment of mice with the CAR activator phenobarbital, an antiepileptic drug, increased plasma triglyceride levels and decreased the hepatic expression of PPARα target genes related to lipid metabolism. A patient with a cerebro-hepato-renal syndrome was investigated. The visceral manifestations were those of the Zellweger syndrome (ZS); however, the child exhibited muscular hypertonia and survived into the 2nd year of life. Ultramicroscopically, hepatocytes were lacking peroxisomes, but, contrary to findings in one patient with ZS [2], contained smooth endoplasmic reticulum. No catalase was found by histochemistry or spectroscopy. Mitochondria showed normal succinate and glutamate respiration, and normal coupling of respiration to the phosphorylation potential. The cytochrome (cyt) content wa…

Alanine/glyoxylate aminotransferase 1 (AGT) is peroxisomal in most normal humans, but in some patients with the hereditary disease primary hyperoxaluria type 1 (PH1), AGT is mislocalized to the mitochondria. In an attempt to identify the sequences in AGT that mediate its targeting to peroxisomes, and to determine the mechanism by which AGT is mistargeted in PH1, we have studied the intracellular compartmentalization of various normal and mutant AGT polypeptides in normal human fibroblasts and cell lines with selective deficiencies of peroxisomal protein import, using immunofluorescence microsc…

Long COVID has emerged as a significant public health challenge with no effective treatments currently available, yet the pathophysiological mechanisms underlying its persistent gastrointestinal (GI) symptoms remain poorly understood. Here, integrating clinical data with transgenic animal models, we discover a critical role for impaired intestinal epithelial repair in the local intestinal etiology of long COVID. Mechanistically, we show that intestinal SARS-CoV-2 reservoirs disrupt very-long-chain fatty acid (VLCFA) metabolism, suppressing activation of peroxisome proliferator-activated recept

Peroxisomes are metabolic organelles that undergo fission to maintain peroxisome number for lipid homeostasis and cellular function. The peroxisomal fission machinery is shared with mitochondria, and although mitochondrial fission regulates neural cell fate decisions during human neurogenesis, it remains unclear whether peroxisomal fission independently influences human neural development. PEX11β selectively regulates peroxisomal fission by promoting membrane deformation, elongation, and downstream fission factor recruitment. To uncouple mitochondrial and peroxisomal fission in early human neu…

The present study assessed the expression of different isoforms of PPAR, an important regulator of cell metabolism and proliferation, in meningiomas and correlated it with different clinicopathological variables and clinical outcomes. A total of 238 meningiomas were studied, including grade 1 (n=133), grade 2 (n=66), grade 3 (n=2), and brain-invasive otherwise benign meningiomas (BIOB) (n=37). The expression of PPAR-γ, phosphorylated PPAR-γ, and PPAR-β was assessed by immunohistochemistry. A semiquantitative grading was used to classify the cases into high and low expression. High nuclear and

Pigmented diseases can significantly impact people's quality of life, with melanogenesis playing a key role. In this study, we first analysed the relationship between peroxisomes, peroxisomal biogenesis factor 16 (PEX16), and melanin synthesis using omics data and clinical samples. Our results showed that peroxisome function and PEX16 expression were negatively associated with melanogenesis. Overexpression of PEX16 in the melanin-producing MNT1 cell line resulted in an increase in peroxisome production and the inhibition of key genes related to melanogenesis, such as microphthalmia-associated …

Activation of peroxisome proliferator-activated receptor alpha (PPARα) may attenuate post-ischemic neuroinflammation. This study examined the neuroprotective potential of pemafibrate, a selective PPARα modulator (SPPARMα), in murine cerebral ischemia. Male CB-17 mice underwent transient middle cerebral artery occlusion (MCAO) for 30 min. Pemafibrate (0.1, 0.3, or 1.0 mg/kg) or vehicle was administered orally once daily for 7 days before MCAO and continued for 2 days thereafter. Pemafibrate improved neurological outcomes (median mNSS: 5.5 with vehicle, 5.0 with pemafibrate 0.1 mg/kg, 4.0 with 0

Mitochondria and peroxisomes are critical for hepatic energy metabolism, lipid homeostasis, and reactive oxygen species (ROS) detoxification. In chronic hepatitis C virus (HCV) infection, continuous injury leads to cirrhosis; however, the spatial arrangement and reversibility of organelle dysfunction remain poorly understood. This study aimed to examine the zonal distribution of mitochondrial and peroxisomal injury in liver biopsies and elucidate the role of circulating cell-free mitochondrial DNA (ccfDNA) in patients with chronic HCV and cirrhosis following antiviral therapy. We employed adva…

Nitric oxide and hydrogen sulfide are signal molecules that can exert regulatory functions in diverse plant processes including fruit ripening. Sulfite oxidase (SOX) is a peroxisomal enzyme that catalyzes the oxidation of sulfite (SO32-) to sulfate (SO42-) with the concomitant generation of H2O2. SOX requires the molybdenum cofactor (Moco) and it has been proposed that SOX functions as a mechanism of protection against sulfite toxicity. Based on the analysis of the pepper genome and fruit transcriptome (RNA-seq), a single gene encoding for a SOX, was identified in chromosome 2. The CaSOX gene

Exposure of perfluorohexane sulfonate (PFHxS) is associated with hepatomegaly and accumulation of lipids that may be mediated by nuclear receptors like peroxisome proliferator-activated receptor-α (PPARα), constitutive androstane receptor (CAR), or pregnane X receptor (PXR). This study tested the hypotheses that: (i) PFHxS causes changes in liver by activating PPARα, CAR, or PXR, and (ii) there is a species difference in PPARα activity by PFHxS. Wild-type, Ppara-null, and PPARA-humanized mice were fed either a control diet, or one containing 2.2 mg PFHxS/kg diet or 25.8 mg PFHxS/kg diet for ei

Osteoarthritis presents a significant clinical challenge due to its high prevalence and the resultant impairment of patients' motor function. Osteoarthritic chondrocytes are characterised by inflammation and metabolic disturbances. Pioglitazone, an agonist of peroxisome proliferator-activated receptor γ (PPAR-γ), has been demonstrated to exert anti-inflammatory effects across various diseases. This study aims to investigate the potential protective effects of Pioglitazone on osteoarthritic chondrocytes. An in vitro chondrocyte inflammation model was established utilising IL-1β. The impact of P

In this editorial, we examine a paper by Koizumi et al, on the role of peroxisome proliferator-activated receptor (PPAR) agonists in alcoholic liver disease (ALD). The study determined whether elafibranor protected the intestinal barrier and reduced liver fibrosis in a mouse model of ALD. The study also underlines the role of PPARs in intestinal barrier function and lipid homeostasis, which are both affected by ALD. Effective therapies are necessary for ALD because it is a critical health issue that affects people worldwide. This editorial analyzes the possibility of PPAR agonists as treatment

Peroxisomal biogenesis disorders (PBD) are autosomal recessive disorders caused by loss-of-function mutations of one of the PEX genes responsible for peroxisomal formation. Impaired peroxisome assembly causes severe multisystemic failure with patient phenotypes ranging from epilepsy, liver disease, feeding issues, biochemical abnormalities, and neurodegeneration. Variants in the same PEX gene can produce wide differences in severity, ranging from individuals with death in the first year of life to adults with milder complications. To study this strong genotype-phenotype correlation, we selecte
