DkMYB117, a novel MYB transcription factor, regulates gibberellin catabolism to control plant height in persimmon interstock systems.

Dwarfing cultivation is a transformative strategy in modern fruit tree management, yet the molecular underpinnings of interstock-induced dwarfism remain elusive in persimmon (Diospyros kaki Thunb.). Here, we unveil DkMYB117, a novel R2R3-MYB transcription factor, as a key molecular switch orchestrating scion dwarfing via gibberellin (GA) catabolism. Using the dwarfing interstock "Nantongxiaofangshi," we demonstrate that DkMYB117 expression is specifically suppressed in grafted scions, leading to reduced active GA levels and stunted growth. Mechanistically, DkMYB117 directly binds to the MBSII

A cell-based fluorescent system and statistical framework to detect meiosis-like induction in plants.

Genetic gains made by plant breeders are limited by generational cycling rates and flowering time. Several efforts have been made to reduce the time to switch from vegetative to reproductive stages in plants, but these solutions are usually species-specific and require flowering. The concept of in vitro nurseries is that somatic plant cells can be induced to form haploid cells that have undergone recombination (creating artificial gametes), which can then be used for cell fusion to enable breeding in a Petri dish. The induction of in vitro meiosis, however, is the largest current bottleneck to

GmPGL1, a Thiamine Thiazole Synthase, Is Required for the Biosynthesis of Thiamine in Soybean.

Thiamine is an essential cofactor in several enzymatic reactions for all living organisms. Animals cannot synthesize thiamine and depend on their diet. Enhancing the content of thiamine is one of the most important goals of plant breeding to solve the thiamine deficiency associated with the low-thiamin staple crops. In this study, a Glycine max pale green leaf 1 (Gmpgl1) mutant was isolated from the EMS mutagenized population of soybean cultivar, Williams 82. Map-based cloning of the GmPGL1 locus revealed a single nucleotide deletion at the 292th nucleotide residue of the first exon of Glyma.1…

Mechanism of pod dehiscence in legumes: insights from phenotypic, anatomical, physiological and molecular studies.

Pod shattering describes the sudden splitting of dried mature legume pods to eject seeds via mechanical tension. Though vital for wild legume reproduction, it severely hinders crop harvesting and causes heavy yield losses, making it an unfavorable trait for cultivation. Pod shattering exists in legumes, grasses and brassicas, yet it is most prominent in legumes and a primary focus of crop domestication and breeding. This review analyzes the mechanism of pod shattering in legumes from the perspectives of morphology, anatomy of the pod suture zone, pod valve physiology, and molecular biology, wi

Bioreactor Systems to Mass-Produce the Undervalued Crop, Celosia argentea, With High Nutrient Impact.

Celosia argentea is an undervalued crop that shows potential for production enhancement due to elevated leaf nutrient accumulative ability. By investigating propagation using various in vitro culture systems, thidiazuron (TDZ)-supplemented nutrient media enhanced yield from 10 plants per explant in semi-solid medium, to 27 under continuous immersion in liquid media in recipient for automated temporary immersion (RITA) bioreactors, to 63 under temporary immersion in liquid media in a balloon-type bubble bioreactor (BTBB). TDZ in the BTBB system also increased shoot biomass and subsequent nutrie

A carotenogenic mini-pathway introduced into white corn does not affect development or agronomic performance.

High-carotenoid corn (Carolight®) has been developed as a vehicle to deliver pro-vitamin A in the diet and thus address vitamin A deficiency in at-risk populations in developing countries. Like any other novel crop, the performance of Carolight® must be tested in different environments to ensure that optimal yields and productivity are maintained, particularly in this case to ensure that the engineered metabolic pathway does not attract a yield penalty. Here we compared the performance of Carolight® with its near isogenic white corn inbred parental line under greenhouse and field conditions, a

OsMGT1 Confers Resistance to Magnesium Deficiency By Enhancing the Import of Mg in Rice.

Magnesium (Mg) is an essential nutrient element for plant growth and plays an important role in numerous physiological and biochemical processes. Mg deficiency inhibits plant growth and has become a growing problem for crop productions in agriculture. However, the molecular mechanisms for the resistance to Mg deficiency in plants were not well understood. In this study, we identified a Mg transporter gene OsMGT1 that confers resistance to Mg deficiency in rice (Oryza sativa). The expression of OsMGT1 was highly induced by Mg deficiency in shoots. Investigation of tissue expression patterns rev

A Natural Allele of a Transcription Factor in Rice Confers Broad-Spectrum Blast Resistance.

Rice feeds half the world's population, and rice blast is often a destructive disease that results in significant crop loss. Non-race-specific resistance has been more effective in controlling crop diseases than race-specific resistance because of its broad spectrum and durability. Through a genome-wide association study, we report the identification of a natural allele of a C2H2-type transcription factor in rice that confers non-race-specific resistance to blast. A survey of 3,000 sequenced rice genomes reveals that this allele exists in 10% of rice, suggesting that this favorable trait has b

Envisaging the role of plant Small RNAs: A comprehensive review on antiviral defense mechanisms.

Plant viruses continue to impose severe constraints on global agriculture, often leading to substantial yield and economic losses. Conventional management strategies such as vector control and resistance breeding frequently fail to provide durable and broad-spectrum protection due to rapid evolution of virus, their dependence on host cellular machinery and the lack of effective antiviral compounds. These shortcomings have led researchers to increasingly explore molecular approaches, with RNA interference (RNAi) emerging as a precise and sustainable strategy for managing plant viral diseases. R

AP2/ERF transcription factors in plant stress adaptation: evolutionary expansion, functional diversification and hormone crosstalk.

APETALA2/ethylene response factor (AP2/ERF) transcription factors constitute a plant-specific regulatory family that coordinates plant development, environmental stress responses, and hormone signaling. However, the contribution of AP2/ERF family expansion and functional diversification to environmental adaptation has not been fully integrated across evolutionary and mechanistic scales. This review synthesizes classical findings and representative advances from the past five years in structural classification, duplication-driven expansion, functional diversification, stress adaptation, and hor

MeMLO12 Impairs Cassava Resistance to Xanthomonas axonopodis pv. manihotis by Repressing Reactive Oxygen Species, Callose Deposition, and Defense Hormone Pathways.

Cassava (Manihot esculenta Crantz) is a vital food and energy crop in tropical regions, yet its yield is significantly threatened by cassava bacterial blight, caused by Xanthomonas axonopodis pv. manihotis (Xpm). Mildew resistance locus O (MLO) genes are known negative regulators of plant immunity, but their roles in cassava disease susceptibility remain unclear. In this study, we identified the MeMLO12 gene from cassava and investigated its function in response to Xpm infection. Subcellular localization revealed that MeMLO12 is specifically localized in the nucleus. Functional analysis via tr

Comparing In Vivo and In Vitro Pollen Tube Attraction Assays for Arabidopsis and Maize.

Pollen tube attraction is a crucial step in the reproduction of angiosperms, ensuring that the male gametophytes, which carry two sperm cells, accurately target the compatible female gametophytes, or embryo sacs, in order to achieve double fertilization. Various experimental systems have been developed to study this process in vivo and in vitro, particularly in the model plant Arabidopsis thaliana (Arabidopsis). However, fewer methods have been described for other plant species, including grasses, which generate much thicker reproductive tissues and organs. Here, we present and compare optimiz

B-Box (BBX) proteins and transcriptional regulation: dynamics, signaling and functions in plants.

Transcriptional regulation is the cornerstone of plant developmental plasticity and environmental resilience. Central to these processes are the B-Box (BBX) proteins, a family of zinc-finger transcription factors that have emerged as pivotal signaling hubs. While their roles were initially defined through light signaling and photoperiodic flowering in Arabidopsis, recent advances have repositioned BBX proteins as integrative nodes across a vast array of physiological processes, including seed germination, thermomorphogenesis, shade avoidance and senescence, as well as responses to both abiotic

Molecular mechanisms and agronomic strategies for thermotolerance in chili pepper (Capsicum annuum L.).

Heat stress is a primary environmental constraint on chili pepper (Capsicum annuum L.) productivity and fruit quality across global agricultural systems. Elevated temperatures adversely affect plant growth, flowering, fruit set, pod morphology, physiology, and metabolism. The severity of these impacts varies with the duration and intensity of heat exposure and with the stage of plant development. Reproductive development is especially vulnerable, resulting in compromised fruit formation and a decline in productivity. Heat stress disturbs photosynthesis, cellular structure integrity, and membra

Integrated morpho-physiological and transcriptomic dissection of waterlogging tolerance in Hibiscus hamabo.

Hibiscus hamabo is an important coastal ornamental plant and a pioneer tree species for coastal protection forests. Prolonged waterlogging severely restricts its ecological expansion toward the intertidal‑zone forefront and limits population regeneration; clarifying its waterlogging‑adaptation mechanisms is essential for coastal wetland ecological restoration. In this study, phenotypic, physiological and transcriptomic responses of H. hamabo under 3-, 9-, and 18-day waterlogging treatments were characterized. Prolonged waterlogging markedly suppressed plant growth and biomass accumulation, and

Engineering plant architecture in the ornamental species Eustoma grandiflorum by knockout of strigolactone biosynthesis.

The targeted mutation of the strigolactone biosynthetic gene, CCD8, through genome-editing CRISPR-Cas9 induces dwarfism and enhanced branching in Eustoma grandiflorum. In ornamental plants, modifying shoot architecture is a major breeding objective for diverse applications; however, achieving this goal using conventional techniques remains challenging. Strigolactones are plant hormones that inhibit shoot branching. We aimed to generate a strigolactone-deficient mutant of Eustoma grandiflorum using CRISPR-Cas9 genome editing.
