Broad and systemic immune-modulating capacity of plant-derived dsRNA.

Double-stranded RNA (dsRNA) is well characterized as an inducer of anti-viral interferon responses. We previously reported that dsRNA extracted from a specific edible plant possesses an immune-modulating capacity to confer, in mice, resistance against respiratory viruses, including the H1N1 strain of the influenza A virus (IAV). We report here that the systemic immune-activating capacity of the plant-derived dsRNA protected mice from infection by a highly virulent H5N1 strain of the IAV. In addition, subcutaneous inoculation of the dsRNA together with the inactivated virion of the H5N1 strain

Sabotage behind enemy lines: Bacterial hijacking of plant cellular functions.

Gram-negative phytopathogenic bacteria employ a Type III Secretion System (T3SS) to deliver effector proteins (T3Es) into plant cells. While the role of T3Es in suppressing plant immunity is well-established, mounting evidence indicates that these proteins also function as sophisticated manipulators of host physiology and metabolism. This review summarizes recent advances in understanding how T3Es modulate cellular processes beyond immune signaling to establish a conducive niche for colonization. In this review, we examine how effectors facilitate pathogen entry and induce water-soaking by man…

Expression and functional analysis of anti-human PD-1 monoclonal antibody in transgenic plants.

Plant-based biopharmaceutical platforms offer a cost-effective and scalable alternative for therapeutic antibody production. In this study, transgenic Nicotiana tabacum (N. tabacum) plants were generated to express pembrolizumab, an anti-human programmed cell death protein-1 (PD-1) monoclonal antibody (mAb), targeting both classical PD-1 on immune cells and the recently identified intrinsic PD-1 (iPD-1) variant in tumor cells. The plant-derived anti-PD-1 mAb (mAbP PD-1) was successfully purified and validated through SDS-PAGE and immunoblotting. Functional analyses using ELISA and immunohistoc

Involvement of Peptidoglycan Receptor Proteins in Mediating the Growth-Promoting Effects of Bacillus pumilus TUAT1 in Arabidopsis thaliana.

Bacillus pumilus TUAT1 acts as plant growth-promoting rhizobacteria for various plants like rice and Arabidopsis. Under stress conditions, B. pumilus TUAT1 forms spores with a thick peptidoglycan (PGN) cell wall. Previous research showed that spores were significantly more effective than vegetative cells in enhancing plant growth. In Arabidopsis, lysin motif proteins, LYM1, LYM3 and CERK1, are required for recognizing bacterial PGNs to mediate immunity. Here, we examined the involvement of PGN receptor proteins in the plant growth promotion (PGP) effects of B. pumilus TUAT1 using Arabidopsis m

Current Understandings on Magnesium Deficiency and Future Outlooks for Sustainable Agriculture.

The productivity of agricultural produce is fairly dependent on the availability of nutrients and efficient use. Magnesium (Mg2+) is an essential macronutrient of living cells and is the second most prevalent free divalent cation in plants. Mg2+ plays a role in several physiological processes that support plant growth and development. However, it has been largely forgotten in fertilization management strategies to increase crop production, which leads to severe reductions in plant growth and yield. In this review, we discuss how the Mg2+ shortage induces several responses in plants at differen

Necessity of a two-stage process for the production of azadirachtin-related limonoids in suspension cultures of Azadirachta indica.

The effect of major nutrients on growth and azadirachtin-related limonoids (AZRL) production in plant cell culture of Azadirachta indica (neem) was studied with the objective to increase the yield of AZRL, one of the major group of pesticidal compounds found in intact neem trees. We report the novel online monitoring of plant cell respiration activities in a new parallel shake flask measuring device. Results obtained using three standard plant cell culture media showed non-growth-associated production characteristics for AZRL. These findings were supported by the oxygen uptake rate data. Furth

Single-cell transcriptomics and time-series metabolite profiling reveal the spatiotemporal regulation of flavonoid biosynthesis genes and phytohormone homeostasis by PAP1 in Arabidopsis.

BACKGROUND: Understanding the spatiotemporal regulation of specialized metabolism in plants is critical for advancing both basic plant biology and biotechnological applications. PRODUCTION OF ANTHOCYANIN PIGMENT 1 (PAP1) is a well-known transcription factor that plays a key regulatory role in the biosynthesis pathway of plant flavonoids. Similar to other secondary metabolites, flavonoid biosynthesis displays cell heterogeneity. However, the cell-specific regulation network of the flavonoid biosynthetic pathway remains unclear. RESULTS: In this study, we utilized single-cell RNA sequencing (sc

Arabidopsis DEFECTIVE KERNEL1 regulates cell wall composition and axial growth in the inflorescence stem.

Axial growth in plant stems requires a fine balance between elongation and stem mechanical reinforcement to ensure mechanical stability. Strength is provided by the plant cell wall, the deposition of which must be coordinated with cell expansion and elongation to ensure that integrity is maintained during growth. Coordination of these processes is critical and yet poorly understood. The plant-specific calpain, DEFECTIVE KERNEL1 (DEK1), plays a key role in growth coordination in leaves, yet its role in regulating stem growth has not been addressed. Using plants overexpressing the active CALPAIN

Overexpression of persimmon DkXTH1 enhanced tolerance to abiotic stress and delayed fruit softening in transgenic plants.

DkXTH1 promoted cell elongation and more strength to maintain structural integrity by involving in cell wall assembly, thus enhanced tolerance to abiotic stress with broader phenotype in transgenic plants. Xyloglucan endotransglucosylase/hydrolase (XTH) is thought to play a key role in cell wall modifications by cleaving and re-joining xyloglucan, and participates in the diverse physiological processes. DkXTH1 was found to peak in immature expanding persimmon fruit, and its higher expression level exhibited along with firmer fruit during storage. In the present study, transgenic Arabidopsis an

Molecular regulation of plant endoreduplication: core mechanisms and their upstream control.

Endoreduplication, or the endocycle, is a widespread developmental program in plants, in which cells repeatedly replicate their nuclear genome without cell division, leading to endopolyploidy. Unlike endomitosis or chemically induced polyploidization, this process arises from an intrinsic reconfiguration of the cell cycle machinery that uncouples G1/S progression from M-phase entry. A useful conceptual framework for this regulation is the "endocycle oscillator", a periodic control logic that supports repeated rounds of DNA replication while bypassing mitosis. Studies in animal systems, particu

Water depth-dependent stem elongation of completely submerged Alternanthera philoxeroides is mediated by intra-internodal growth variations.

Complete submergence, especially deep submergence, poses a serious threat to the growth and survival of plants. One study previously showed that Alternanthera philoxeroides (a herbaceous perennial plant) submerged at depth of 2 m presented fast stem elongation and reduced stem elongation as water depth increased. In the present study, we aimed to figure out from the morphological and anatomical perspective how the differential growth response of the plant to water depth was achieved. We investigated the elongation of different stem parts and the relationship of stem elongation to cell size and

Flagellin peptide flg22 gains access to long-distance trafficking in Arabidopsis via its receptor, FLS2.

Diverse pathogen-derived molecules, such as bacterial flagellin and its conserved peptide flg22, are recognized in plants via plasma membrane receptors and induce both local and systemic immune responses. The fate of such ligands was unknown: whether and by what mechanism(s) they enter plant cells and whether they are transported to distal tissues. We used biologically active fluorophore and radiolabeled peptides to establish that flg22 moves to distal organs with the closest vascular connections. Remarkably, entry into the plant cell via endocytosis together with the FLS2 receptor is needed f…

A novel approach to enhance resistance to vascular disease by expressing cell-death-inducing fungal elicitors in the xylem tissue.

Constitutive expression of hypersensitive cell-death-inducing effectors secreted by pathogens has proved to be difficult in plants. Herein, xylem-specific promoters (PVND7 or PVND2) were utilized to express three cell-death-inducing proteins (FocSIX1A, FocSIX6 or TvSm1) of fungal origin in banana plants. The regenerated plants were not only healthy but also showed improved resistance to fungal (Foc vcg0124 and Foc vcg0125) infection. These banana plants developed systemic resistance-like symptoms, including elevated jasmonic acid content, callose depositions, increased xylem wall thickness and

London Rocket (Sisymbrium irio L.) as Healthy Green: Bioactive Compounds and Bioactivity of Plants Grown in Wild and Controlled Environments.

London rocket (Sisymbrium irio) is a wild green consumed globally, yet its phytochemical composition remains underexplored. In this study, we analyzed the leaves of wild S. irio plants and those grown in controlled environments (GCE) with varying electrical conductivities (EC) and light spectra. Plants were assessed for growth, phenolic content, vitamin C, antioxidant activity, glucosinolates, and antiproliferative effects against HT-29 human colorectal cancer cells. The optimal biomass yield occurred at the EC levels of 3.0-3.5 dS m-1 under Valoya® LED light. Wild plants showed higher antioxi

[Compartmentation of salicylate-induced proteins].

A key factor of immunity, salicylic acid (exogenous or accumulating under the action of biotrophic or semibiotrophic pathogens on plants), causes the formation of not only protective antipathogenic proteins but also many proteins, which enhance the resistance of host plant cells. Salicylate-induced proteins, which are encoded by nuclear genes and formed with cytoplasmic ribosomes, function in the cytosol or are transported into nuclei, vacuoles, plastids, mitochondria, and outside the plasmalemma. This review is focused on salicylate-induced proteins, which are not only delivered into differen…

Glycan-directed SARS-CoV-2 inhibition by leek extract and lectins with insights into the mode-of-action of Concanavalin A.

Four years after its outbreak, severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) remains a global challenge for human health. At its surface, SARS-CoV-2 features numerous extensively glycosylated spike proteins. This glycan coat supports virion docking and entry into host cells and at the same time renders the virus less susceptible to neutralizing antibodies. Given the high genetic plasticity of SARS-CoV-2 and the rapid emergence of immune escape variants, targeting the glycan shield by carbohydrate-binding agents emerges as a promising strategy. However, the potential of carbohydr
