The thermal unfolding of F-actin and the changes induced in it by the binding of phosphate analogues were studied by differential scanning calorimetry. It is shown that the conformation of actin is drastically altered by interaction with beryllium fluoride or aluminium fluoride, while the effects of vanadate and phosphate are negligible. The effect of glycine-to-arginine mutations in the alpha 1 (I)-chain on collagen triple-helix structure in lethal perinatal osteogenesis imperfecta was studied by determination of the helix denaturation temperature and by computerized molecular modelling. Efforts to design proteins with greatly reduced sequence diversity have often resulted in proteins with so-called molten globule properties. A novel method was developed to obtain both highly purified bovine serum albumin (BSA) and immunoglobulin G (IgG), at the same time, on a pilot scale. Heat-isopropyl alcohol was used to denature and precipitate the other plasma proteins, except for BSA and IgG; then, CM-Trisacryl was applied to further purify and isolate BSA and IgG. Both oxy and deoxy human hemoglobin A were crosslinked with tris(3,5-dibromosalicyl) tricarballylate. The major species from both reactions contained an inter-subunit crosslink. We have purified Chia enzymes with striking acid stability and protease resistance from chicken and pig stomach tissues using a chitin column and 8 M urea (urea-Chia). Anthrax lethal factor (LF) is a zinc-dependent endopeptidase which, through a process facilitated by protective antigen, translocates to the host cell cytosol in a partially unfolded state.
