Clinical impact of predictive assays for acute and late radiation morbidity.

BACKGROUND: Clinically reliable predictive assays for normal tissue radiation sensitivity would help to avoid severe radiation induced morbidity and result in individualized dose prescriptions. Profound differences of individual fibroblast and lymphocyte radiation sensitivity in vitro have been documented in patients with certain genetic syndromes but also in patients without known genetic disorders. The following review evaluates whether fibroblast or lymphocyte radiation sensitivity measured in vitro correlates with the degree of acute and late radiation induced morbidity. RESULTS: Acute ra

Survival of human normal thyroid cells after X-ray irradiation.

Normal thyroid cells from 25 individuals treated surgically for malignant or benign thyroid tumour were cultured in vitro and radiation induced cytotoxicity was studied. The mean lethal dose (Do), quasi-threshold (Dq), and extrapolation number (n) of survival curves of actively dividing thyroid cells assayed by colony formation were estimated to be 92.9 +/- 2.8 cGy (rad), 58.1 +/- 6.9 cGy and 2.0 +/- 0.1, respectively (average for 25 individuals +/- standard error). These results suggest that proliferating human thyroid cells are more sensitive to X-rays than most other nonhaematologic mammali

Synergistic effect of broad-spectrum sunscreens and antihistamines in the control of idiopathic solar urticaria.

BACKGROUND: It can be difficult to provide patients with idiopathic solar urticaria adequate protection from sunlight. In a nonrandomized controlled trial, we used a standardized phototest procedure to determine the effects of using sunscreen and antihistamine to control idiopathic solar urticaria. OBSERVATIONS: Three patients with idiopathic solar urticaria underwent phototesting with UV-B and UV-A radiation. The minimal urticarial dose (MUD) was determined 15 minutes after irradiation. The patients were subsequently tested with 5 times the MUD, and the reaction was graded every minute for 1

Radiosensitivity of the developing haemopoietic system in mammals and its adult consequences: animal studies.

The haemopoietic system in the developing mammal is very sensitive to the damaging effects of ionizing radiation. Epidemiological studies have established a strong association between obstetric exposure to diagnostic radiation and an increase in the incidence of childhood leukaemia and between low dose gamma irradiation during the early fetal period and mental retardation in children. It has been suggested that insufficient oxygen supply to the developing brain due to radiation induced damage to fetal haemopoietic tissue has a role in inducing the severe mental retardation observed in the Japa

Secondary Effects of Radiation Therapy to the Hand for Benign Conditions.

Background: Emerging literature introduces radiation therapy for benign hand conditions. However, hand surgeons are wary recommending radiation therapy for nonmalignant conditions. In our practice, we have used radiation therapy for patients who present with infiltrative or recurrent giant cell tumor of the tendon sheath (GCTTS) since 1998. The purpose of this study is to examine the secondary effects of radiation to the hand through the critical lens of a hand surgeon. Methods: A case series of patients who received radiation therapy for GCTTS were reviewed. The Radiation Oncology/Toxicity Gr

Predicting toxicity for head and neck cancer patients undergoing radiation therapy: an independent and external validation of MDASI-HN based nomogram.

AIM: We conducted a study to validate the MDASI-HN based nomogram, which is used to predict the acute toxicities in head and neck cancer patients undergoing radiation therapy with or without chemotherapy. BACKGROUND: Tolerance to radiation varies from patient to patient and also depends on various other factors like tumor volume, dose of radiation, chemotherapy. Predicting the toxicities allow us to identify potential candidates who are likely to have a higher toxicity and, in addition, evaluates the nomogram when done on an independent group of patients. MATERIALS AND METHODS: Sixty biopsy

Health impacts of large releases of radionuclides. Biological effects of prenatal irradiation.

After large releases of radionuclides, exposure of the embryo or fetus can take place by external irradiation or uptake of radionuclides. The embryo and fetus are radiosensitive throughout prenatal development. The quality and extent of radiation effects depend on the developmental stage. During the preimplantation period (one to 10 days postconception, p.c.), a radiation exposure of at least 0.2 Gy can cause the death of the embryo. Malformations are only observed in rare cases when genetic predispositions exist. Macroscopic, anatomical malformations are induced only after irradiation during

Multi-Species Tardigrade RNA Isolation Protocol.

Tardigrades are microscopic animals that are known for their exceptional tolerance to environmental stress. These animals can survive desiccation, extreme pressure, low temperatures, and high levels of ionizing radiation. However, the molecular mechanisms underlying these extremotolerant capabilities remain incompletely understood. The molecular approaches used to investigate these mechanisms, which include RNA sequencing, RNA interference (RNAi), and heterologous expression of tardigrade genes in systems such as bacteria, require the isolation of high-quality messenger RNA (mRNA) from limited

Coil embolization of a neonatal pulmonary arteriovenous malformation.

Pulmonary arteriovenous malformation (PAVM), as a part of Osler-Weber-Rendu Syndrome, in the neonate, is a rare hereditary vascular malformation. Large intrapulmonary right-to-left shunting, causing hypoxaemia and cyanosis, can be a life-threatening condition. Repeated transcatheter coil embolization procedures proved to be a favorable strategy to improve systemic arterial oxygen saturation, with a good outcome in a newborn child. While the radiation dose was high, the use of this amount of radiation was felt to be justified and its effects considered tolerable in the treatment of this patient

Senile cataract. The case against light.

A photochemical theory of the etiology of senile cataract is developed on the assumption that observed prevalences are representable by cumulants of tolerances, the latter being distributed normally and depending in a quantitatively defined way on the amount of short-wavelength radiation absorbed during life. The theoretical risk-curves are consistent with the observed ones. The variation of the predicted and observed values has a slope not differing significantly from unity, but the intercept differs from zero. It is concluded tentatively that the photochemical hypothesis accounts for the obs

Preoperative radiation treatment for rectal cancer: comparison of target coverage and small bowel NTCP in conventional vs. 3D-conformal planning.

A prospective study was undertaken to evaluate the improvement in rectal cancer radiation treatment achieved with the implementation of target delineation for conformal radiotherapy, replacing conventional technique using standard radiological anatomy for target volume definition. In 10 patients receiving preoperative pelvic irradiation for rectal cancer, a 3-field technique was designed by a 3-dimensional (3D) planning system. Two plans were simulated for each patient, one with the fields designed in the conventional way based on radiological anatomy, and the other with the fields designed on

Organ preservation with interstitial radiation for base of tongue cancer.

Management options for squamous cell carcinoma of the base of tongue include surgical resection (often with adjuvant radiation), definitive external radiation and external combined with interstitial radiation. The reported series is a single institution experience with interstitial radiation for base of tongue cancer. Twenty patients were treated definitively with interstitial radiation as a boost to external radiation, and four patients were treated palliatively with interstitial radiation alone for recurrent base of tongue cancers or disease arising in a previously irradiated base of tongue.

A radiation therapist perspective on the use of a small gauge radiopaque catheter for urethral definition and dose escalation in prostate SBRT.

INTRODUCTION/BACKGROUND: An increase in targeting accuracy for Stereotactic Body Radiation Therapy (SBRT) to the prostate allows for a reduction in Planning Target Volume (PTV). This can reduce treatment-related side effects. In this study, we used a small gauge radiopaque catheter as a fiducial marker for prostate SBRT treatment delivery. Preparation and early engagement of radiation therapists is critical to the success of a catheter-guided prostate SBRT program. METHODS: This study included forty-two patients from two cancer centers in Alberta, Canada that underwent prostate SBRT treatment

Combination of ionizing radiation and 2-thio-6-azauridine induces cell death in radioresistant triple negative breast cancer cells by downregulating CD151 expression.

BACKGROUND: Triple-negative breast cancer (TNBC) represents the most aggressive subtype of breast cancer and is frequently resistant to therapy, ultimately resulting in treatment failure. Clinical trials have demonstrated the potential of sensitizing radiation therapy (RT)-resistant TNBC through the combination of chemotherapy and RT. This study sought to explore the potential of CD151 as a therapy response marker in the co-treatment strategy involving ionizing radiation (IR) and the repurposed antiviral drug 2-Thio-6-azauridine (TAU) for sensitizing RT-resistant TNBC (TNBC/RR). METHODS: The

Proteomic analysis of irradiated lung tissue of mice using gel-based proteomic approach.

PURPOSE: Exposure to radiation causes severe alterations of protein expression level inside the cell, thus it may influence the biological events and stress response. In the present investigation, we have demonstrated the effect of radiation on mice lung tissues. MATERIALS AND METHODS: Two-dimensional gel electrophoresis (2-DE) coupled with MALDI-TOF/TOF was used to check the expression changes in lung proteome profile of strain 'A' female mice after exposure to lethal doses of gamma irradiation at different time periods (24 and 48 h). Identified proteins were analysed for their altered expre
