Melatonin increases serotonin N-acetyltransferase activity and decreases dopamine synthesis in light-exposed chick retina: in vivo evidence supporting melatonin-dopamine interaction in retina.

The administration of melatonin, either peripherally (0.01-10 mg/kg) or intraocularly (0.001-10 mumol/eye), to light-exposed chicks dose-dependently increased serotonin N-acetyltransferase (NAT) activity in retina but not in pineal gland. The effect of melatonin was slightly but significantly reduced by luzindole (2-benzyl-N-acetyltryptamine), and not affected by two other purported melatonin antagonists, N-acetyltryptamine and N-(2,4-dinitrophenyl)-5-methoxytryptamine (ML-23). The elevation of the enzyme activity induced by melatonin was substantially stronger than that evoked by 5-hydroxytry…

Physiological Postnatal Neurogenesis, Development, and Maturation in the Regenerable Retina.

The retina enlarges postnatally by ∼440% in axolotls and ∼150% in humans, predicting a reduction in neuron densities by 81.5% and 60%, respectively, if neurogenesis is absent. The concurrent retinal remodeling is poorly characterized. We studied retinas of 2-15 cm axolotls. Results showed that hatchling (∼2 cm) retinas, particularly the peripheral (Peri) had 1-2 more soma rows in the ganglion cell (GCL) layer and outer (ONL) and inner nuclear (INL) layers. Cell density reduced in GCL (GCs, -49%) in juveniles (4-12 cm) but increased in INL (74%) and ONL (55.5%) in young juveniles (4-6 cm), demo…

Distribution of doublecortin in the developing retina of birds: A comparative analysis.

The avian retina presents an elegant and highly accessible model for investigating the spatial and temporal dynamics of neurogenesis within the central nervous system. In recent years, the microtubule cytoskeleton has emerged not merely as a structural scaffold, but as a dynamic regulator of the transition of a morphologically homogeneous pool of multipotent progenitor cells to postmitotic neurons. Doublecortin (DCX) is a microtubule-associated protein broadly recognized for its fundamental roles in neuronal migration, differentiation, and structural plasticity within the developing nervous sy…

The existence of puckering in the retina beyond the macula: a prospective analysis.

PURPOSE: To examine the existence of idiopathic epiretinal membranes (ERM) in the peripheral retina in patients with or without macular pucker. METHODS: The presence of the peripheral idiopathic epiretinal membranes was sought using 200-degree widefield OCT (OPTOS Silverstone Swept source-OCT (SS-OCT) in two age, sex, and ocular comorbidities-matched groups consisting of 38 patients (24 with macular ERM and 14 without macular ERM). The significance of the correlation between the presence of macular and peripheral ERM was determined using Fisher's exact test and unpaired t-test. RESULTS: Seve…

Adaptive immune crosstalk and complement dysregulation in the aging retina.

The eye, long viewed as immune-privileged, is now recognized as a dynamic immunological environment where innate and adaptive mechanisms intersect to maintain retinal homeostasis. In aging and age-related retinal diseases such as age-related macular degeneration (AMD), this balance deteriorates as chronic, low-grade inflammation replaces immune quiescence. Adaptive immune involvement in AMD is reflected by increased antiretinal autoantibodies and systemic immune changes, including accelerated aging of CD8+ T-cells. Complement activation further amplifies adaptive responses; C5a stimulates T-ce…

Defocus coding and transcriptomic remodeling in the mouse myopic retina.

The theory of visually guided ocular growth is well supported in explaining myopia, but how the retina senses focus versus defocus and converts the signaling into growth-modulating genetic signals remains unresolved. Using whole-cell recordings and single-cell RNA-seq in the mouse retina, we show that lateral inhibitory networks-horizontal cells in the outer retina, but not AII amacrine cells in the inner retina, respond to optical defocus. Dopaminergic amacrine cells (DACs) are maximally excited by focused images and increasingly inhibited by high blur, consistent with dopamine's anti-myopiag…

Geographic Access to Pediatric Retina Specialists in the United States.

BACKGROUND AND OBJECTIVE: With rising incidence of pediatric retina disease, this study aimed to identify geographic and socioeconomic factors that predict residential access to pediatric retinal specialists. PATIENTS AND METHODS: This cross-sectional, retrospective study identified pediatric retina specialists in the United States and de-identified census tract-level data from public datasets. An origin-destination cost matrix was used to calculate travel time to the closest pediatric vitreoretinal specialist. RESULTS: Factors associated with greater travel burden to the nearest pediatric r…

Discovering flexible codes for prediction across timescales in the retina.

The retina must encode visual information in a way that supports fast, predictive behavior despite significant processing delays. How this encoding adapts in an ever-changing world, when the temporal statistics of visual input shift, remains an open question. Here we record from populations of retinal ganglion cells in the axolotl as they respond to a stochastic moving bar stimulus across five different temporal correlation scales. Using the information bottleneck (IB) framework, and treating the prediction horizon as a free parameter inferred from the data, we ask what timescale of future mot…

Effect of Endogenous FGF21 Deficiency on the Inflammatory Microenvironment of the Retina.

BACKGROUND: While the systemic metabolic role of fibroblast growth factor 21 (FGF21) is well-established, its function in retinal homeostasis and its link to retinal diseases like age-related macular degeneration (AMD) and diabetic retinopathy (DR) remains poorly understood. This study investigated the impact of endogenous FGF21 deficiency on the retinal immune microenvironment. METHODS: Retinal structure was assessed in FGF21 KO and wild-type mice using spectral-domain optical coherence tomography. Transcriptomic profiles of the retina/choroid were analyzed by RNA-seq. Differentially express…

mRNA delivery to the retina restores REP1 function in choroideremia.

Inherited retinal diseases (IRDs) represent a major cause of blindness, yet current viral vector-based gene therapies are limited by high cost, restricted cargo capacity, and safety concerns related to immunogenicity. We explore here retinal delivery of non-viral, in-vitro-transcribed mRNA using lipid nanoparticles (LNPs) as a safe, cost-effective gene augmentation strategy for choroideremia (CHM). CHM is an X-linked IRD caused by loss-of-function mutations in CHM that encodes Rab escort protein-1 (REP1), which is essential for Rab GTPases prenylation and intracellular trafficking. We demonstr…

The human retina in Alzheimer's disease: Pathology, mechanisms, and biomarkers.

Alzheimer's disease (AD) is characterized by progressive neurodegeneration and synaptic dysfunction that begins decades before clinical symptoms emerge. While AD research has traditionally focused on the brain, increasing evidence suggests that the retina undergoes pathological remodeling that shares features with cerebral changes. Advances in retinal imaging, including optical coherence tomography (OCT), OCT angiography, and hyperspectral approaches, have identified structural, vascular, and functional abnormalities in individuals with mild cognitive impairment (MCI) and early-stage AD. This …

Adaptations of the Vertebrate Retina to Low-Light Conditions: A Review.

Vision is a critical sensory modality in vertebrates, enabling diverse adaptations to environmental challenges. This review explores the remarkable adaptations of vertebrate eyes to low-light environments, such as nocturnal habitats, deep-sea ecosystems and subterranean niches. Key structural and functional modifications include enhanced eye size, rod-dominated retinas, reflective tapetal layers and photopigment spectral tuning. The role of photoreceptor specialisation and retinomotor movements in optimising photon capture and minimising light scatter is examined, alongside evolutionary trade-…

Characterization of age-related dolichol increases in the mouse retina.

We characterized the age-related increases in dolichol levels in the mouse retina using liquid chromatography-mass spectrometry (LC-MS). The four major dolichol species, dolichol-17 (Dol-17), Dol-18, Dol-19, and Dol-20-all increased drastically with age. The largest increase was in Dol-18 levels, which rose by a factor of 100 from postnatal day 5 (PD 5) to PD 600. These increases occurred in two distinct phases: a linear increase in Phase I (PD 5 to PD 30), and a nonlinear increase in Phase II (PD 30 to PD 600). There was also an age-dependent shift in the dolichol chain length profile toward …

Expression of Versican in the Retina and Its Implication in Retinal Disease.

Chondroitin sulfate proteoglycans are characterized by their sulfated glycosaminoglycan chains covalently attached to the core protein. Versican, encoded by the CSPG2 gene, is a chondroitin sulfate proteoglycan highly expressed in the human and mouse retina. Due to alternative splicing, it exists in four different isoforms: V0, V1, V2 and V3. Interactions with a variety of proteins lead to its main functions, such as influencing cellular adhesion, migration, proliferation and regulating inflammatory processes. Versican expression is high during retinal development and decreases in the mature t…
