Three-dimensional in vivo imaging of the mouse intraocular vasculature during development and disease.

PURPOSE: Aberrant growth of blood vessels in the eye is a major cause of vision loss, occurring as a complication of diabetic retinopathy, age-related macular degeneration, and retinal vascular occlusions, among others. Whereas in humans, in vivo angiography is routinely used to image such diseases, many animal models of ocular vascular disease and development rely on dissected tissues that may not accurately represent in vivo conditions and require enucleation of the eye, the death of the animal, or both. METHODS: A method of three-dimensional imaging of blood vessels was used in the living

Unusual case of enlarged choroidal vessels in neurofibromatosis type 1 observed with near-infrared reflectance and spectral domain optical coherence tomography.

The cardinal diagnostic signs of neurofibromatosis type 1 (NF1) are Lisch nodules of the iris and optic pathway gliomas. Retinal mi-crovascular alterations have been described but with uncertain signi-ficance. Choroidal nodules, detectable with near-infrared reflectance (NIR) imaging, are present in most cases and have been proposed as a new diagnostic criterion. Recently, a study reported the presence of unusual dilated choroidal vessels, visible through NIR examination. We report a case of a 65-year-old patient with NF1. Best-corrected visual acuity was 20/20 with a refractive error of +2.75

Improved interpretation of flow maps obtained by scanning laser Doppler flowmetry using a rat model of retinal artery occlusion.

PURPOSE: To improve the interpretation of Heidelberg Retina Flowmeter (HRF; Heidelberg Engineering GmBH, Dosselheim, Germany) flow maps by examining the influence of specific vascular structures and focus depth in the presence and absence of retinal blood flow. METHODS: HRF flow maps were recorded from the inferior retina of anesthetized Brown Norway rats over a wide range of focus levels, before and after laser occlusion of the retinal circulation. Analysis of the resultant flow maps showed that the sample window was positioned on a retinal artery, arteriole, or vein, or in a retinal capilla

Effect of Pupil Dilation with Tropicamide on Retinal Vascular Caliber.

Purpose: The retinal blood vessels reflect changes in the brain's micro-circulation and these changes have been shown to correlate with the … e magnification. Conclusion: These results suggest that tropicamide does not change the width of the retinal vessels, however width parameters as measured by standard approaches may be reduced due to image magnification changes resulting from cycloplegia. In this study, improved optic nerve head segmentation for image scale conversion removed the magnification err …

An automated retinal imaging method for the early diagnosis of diabetic retinopathy.

BACKGROUND: Diabetic retinopathy is a microvascular complication of long-term diabetes and is the major cause for eyesight loss due to changes in blood vessels of the retina. Major vision loss due to diabetic retinopathy is highly preventable with regular screening and timely intervention at the earlier stages. Retinal blood vessel segmentation methods help to identify the successive stages of such sight threatening diseases like diabetes. OBJECTIVE: To develop and test a novel retinal imaging method which segments the blood vessels automatically from retinal images, which helps the ophthalmo

The effect of acetazolamide on the retinal circulation.

The effect of the carbonic anhydrase inhibitor, acetazolamide, on the retinal circulation was studied in 10 healthy volunteers. Acetazolamide was administered intravenously at a dose of 500 mg. Retinal blood flow was determined from red cell velocity using laser Doppler velocimetry and vessel diameter measurement using computerised digital image analysis of monochromatic fundus photographs. There was a significant increase in retinal blood flow at 30 minutes and 60 minutes after acetazolamide injection (p = 0.002). Retinal vessels showed vasodilatation reaching significant levels 60 minutes af…

Hypothesis: vasoconstriction contributes to amaurosis fugax.

Platelets play a critical role in the pathophysiology of amaurosis fugax. Emboli to retinal vessels apparently produce amaurosis but, in addition, we propose that augmented vasoconstrictor responses and vasospasm may contribute to amaurosis. In this study we tested the hypothesis that constrictor responses of retinal vessels to serotonin, which is released when platelets aggregate, are potentiated in experimental atherosclerosis. Blood flow to the retina was measured in normal and atherosclerotic cynomolgus monkeys. In normal monkeys, infusion of serotonin did not alter flow to the retina. In …

Cellular mechanisms in retinal vascular development.

Since the pioneering work of Ashton and others, the primate retina has been thought to vascularize by a vasculogenic linkage of endothelial precursor cells. Recent investigations using specific histologic and morphologic criteria question the contribution of vasculogenesis to retinal development. Instead, in primates and mice cells previously designated as retinal angioblasts have been identified as astrocytes that form a vascular-like plexus preceding vessel invasion. Further, in primates and mice retinal vascularization proceeds via angiogenic sprouting from pre-existing vessels in all regio

The gut microbiota regulates diabetic retinopathy in adult rats.

INTRODUCTION: Diabetic retinopathy (DR) is the most common complication of diabetes. Neuronal apoptosis, activated microglia, and microvascular changes are early features of DR. The gut microbiota is critical for the maturation and activation of microglia in the brain, and DR patients exhibit gut dysbiosis. However, the effect of the gut microbiota on retinal microglia under normal or diabetic conditions is still unclear. METHODS: Type 2 diabetes (T2D) was established in male adult Brown Norway (BN) rats, and they were treated with gavage of broad-spectrum antibiotic (ABX) suspension. Retinal
