Moving pieces in a taxonomic puzzle: venom 2D-LC/MS and data clustering analyses to infer phylogenetic relationships in some scorpions from the Buthidae family (Scorpiones).

The Buthidae is the most clinically important scorpion family, with over 500 species distributed worldwide. Taxonomical positions and phylogenetic relationships concerning the representative genera and species of this family have been mostly inferred based upon comparisons between morphological characters. Yet, some authors have performed such inferences by comparing some structural properties of a few selected molecules found in the venoms from these scorpions. Here, we propose a novel methodology pipeline designed to address these issues. We have analyzed the whole venoms from some species t…

Different pharmacological properties between scorpion toxin BmKcug2 and its degraded analogs highlight the diversity of K+ channel blockers from thermally processed scorpions.

Novel degraded potassium channel-modulatory peptides were recently found in thermally processed scorpions, but their pharmacological properties remain unclear. Here, we identified a full-length scorpion toxin (i.e., BmKcug2) and its four truncated analogs (i.e., BmKcug2-P1, BmKcug2-P2, BmKcug2-P3 and BmKcug2-P4) with three conserved disulfide bonds in processed scorpion medicinal material by mass spectrometry. The pharmacological experiments revealed that the recombinant BmKcug2 and BmKcug2-P1 could selectively inhibit the human Kv1.2 and human Kv1.3 potassium channels, while the other three a…

A variable residue in the pore of Kv1 channels is critical for the high affinity of blockers from sea anemones and scorpions.

Animal toxins are associated with well defined selectivity profiles; however the molecular basis for this property is not understood. To address this issue we refined our previous three-dimensional models of the complex between the sea anemone toxin BgK and the S5-S6 region of Kv1.1 (Gilquin, B., Racape, J., Wrisch, A., Visan, V., Lecoq, A., Grissmer, S., Ménez, A., and Gasparini, S. Characterization of BmKbpp, a multifunctional peptide from the Chinese scorpion Mesobuthus martensii Karsch: gaining insight into a new mechanism for the functional diversification of scorpion venom peptides.

BmKbpp is a novel cationic and α-helical peptide from the Chinese scorpion Mesobuthus martensii Karsch, of which function or biological activity has not been characterized so far. Here we showed that BmKbpp possesses strong antimicrobial activity against both Gram-positive and Gram-negative bacteria with a MIC range from 2.3 μM to 68.2 μM for the majority of tested bacteria. BmKbpp also inhibits the growth of tested fungi with an IC50 range from 0.2 μM to 3.1 μM. Because BmKbpp potently inhibits the growth of some antibiotics-resistant pathogens, and shows very weak hemolytic activity, it has

Efficacy and safety of scorpion antivenom plus prazosin compared with prazosin alone for venomous scorpion (Mesobuthus tamulus) sting: randomised open label clinical trial.

OBJECTIVE: Envenomation by Mesobuthus tamulus scorpion sting can result in serious cardiovascular effects. Scorpion antivenom is a specific treatment for scorpion sting. Evidence for the benefit of scorpion antivenom and its efficacy compared with that of commonly used vasodilators, such as prazosin, is scarce. We assessed the efficacy of prazosin combined with scorpion antivenom, compared with prazosin alone, in individuals with autonomic storm caused by scorpion sting. DESIGN: Prospective, open label randomised controlled trial. SETTING: General hospital inpatients (Bawaskar Hospital and R

The amino acid sequence of toxin IV from the Androctonus australis scorpion: differing effects of natural mutations in scorpion alpha-toxins on their antigenic and toxic properties.

The complete amino acid sequence (64 residues) of the AaH IV toxin from the scorpion Androctonus australis Hector was determined by automated Edman degradation and was compared with the sequences of other Androctonus toxins. AaH IV was also tested by radioimmunoassay for binding to antisera raised against other toxins of the same species. The results indicated that AaH IV shares some of the antigenic properties of AaH I and AaH III toxins, but does not cross-react with anti-AaH II antibodies. The structural basis for the observed antigenic relationships can be found in the high degree of homol

Gas gill respiration enhances underwater copulation in a water scorpion.

To overcome physiological constraints during submergence, many air-breathing aquatic animals utilize an air store. If this store functions as a gas gill for dissolved oxygen uptake, this will extend underwater copulation when atmospheric oxygen is inaccessible. We tested this hypothesis using water scorpion Nepa hoffmanni (Hemiptera: Nepidae). Our field surveys showed that, although N. hoffmanni typically inhabit shallow water, males cannot access atmospheric oxygen via their respiratory siphon during underwater copulation. Laboratory experiments revealed active oxygen use from the air store b

Scorpion Venom Neurotoxins: Molecular Diversity, Mechanisms, and Drug Scaffolds.

Venom is a key evolutionary innovation of venomous organisms in the long-term process of survival adaptation. As one of the oldest arthropods, scorpions produce venom rich in bioactive peptides that also constitute a valuable pharmacological resource. Omics-driven discovery and structural biology have expanded the peptide catalog and clarified structure-function principles across disulfide-bridged (DBPs) and non-disulfide-bridged peptides (NDBPs). Within this arsenal, ion-channel targeting neurotoxins predominantly modulate Nav, Kv, Calcium, Chloride, and TRP channels to achieve predation, def…

Socioenvironmental, socioeconomic, and public management indicators as predictors of scorpionism in Brazil.

Scorpionism is an increasing public health problem in tropical countries, particularly in Brazil, where social inequality, environmental degradation, and unplanned urbanization favor the proliferation of scorpions and human-scorpion encounters. In this study, we investigated the association between scorpion sting notifications and hospitalizations and a set of socioeconomic, socioenvironmental, environmental, and public management indicators related to sustainability across all Brazilian municipalities. We used secondary data from national health information systems and indicators from the Sus…

Untargeted metabolomics reveal the corrective effects of scorpion on epileptic mice.

Scorpion is a commonly used drug in traditional Chinese medicine for treating epilepsy, although the exact mechanisms are not yet fully understood. This study aimed to compare the treatment effects of Scorpion water extract (SWE) and Scorpion ethanol extract (SEE) on mice with pentetrazole-induced epilepsy and investigate the possible mechanisms through metabolomics methods. A pentetrazole-induced epileptic mice model was used to assess the corrective effects of SWE and SEE. Untargeted metabolomics, utilizing UPLC-Q-TOF/MS, was employed to analyze the metabolic profiles of mice and identify me

AI models uncover factors influencing scorpionism in Northern Brazil.

Envenomation by scorpion stings is a serious public health problem in tropical regions of the world. In Brazil's Northern region, there has been a significant increase in cases over the last decade, accompanied by a rise in the fatality rate. Climate change and intensive land use are altering the distribution of species that pose health risks and may be associated with the increased incidence of accidents. We integrated species distribution models (SDMs) of three medically important species (Tityus obscurus, T. metuendus, and T. silvestris), bioclimatic data, and land use to predict scorpionis

Scorpion Venom Peptides in Ophthalmology: Insights from the Babylonian Talmud.

Historical medical texts can reveal overlooked therapeutic approaches relevant to modern ophthalmology. This perspective revisits a remedy from the Babylonian Talmud prescribing a mixture of scorpion and kohl for an eye condition called buruqti (also transliterated as beroketi), traditionally translated as cataract. Through philologic, zoologic, and pathologic analysis, we argue that buruqti likely refers to a corneal or conjunctival lesion rather than a true lens opacity. The description within the remedy of the scorpion's "seven-beaded tail" is interpreted symbolically within the cultural an

Advancements in diagnostic techniques for scorpion venom identification: A comprehensive review.

Scorpion envenomation's ignored public health problem in tropical and subtropical countries is alarming. Particularly dangerous for small children and the elderly, it can cause severe problems and even death. Recent studies have proposed the creation of rapid, easy, species-specific, and sensitive detection kits as an alternative to the methods currently used to identify scorpions. Unfortunately, there is currently no commercially available technology for detecting scorpion envenomation in clinical settings, especially in remote tropical health centres. This study delineates the most dangerous…

Study of the Acute Toxicity of Scorpion Leiurus macroctenus Venom in Rats.

BACKGROUND: The expansion of the territory of human habitation leads to inevitable interference in the natural range of distribution of one or another species of animals, some of which may be dangerous for human life. Scorpions-the Arachnida class and order Scorpiones-can be considered as such typical representatives. Scorpions of the Buthidae family pose a particular danger to humans. However, LD50 has not yet been defined for many species of this family, in particular, new representatives of the genus Leiurus. Leiurus macroctenus is a newly described species of scorpion distributed in Oman, …
