The membrane permeability transition (MPT) induced by Ca2+ and Pi or Asi was studied in rat kidney mitochondria. Membrane potential, Ca2+ transport and swelling were used to monitor the MPT. BACKGROUND: Polyamines, including putrescine (PU), spermidine (SPD), and spermine (SPM), are ubiquitous bioactive compounds essential for cell proliferation, genomic stability, autophagy, and the regulation of oxidative and inflammatory responses. Growing evidence, particularly for SPD, suggests that polyamine-rich diets may protect against age-related conditions such as cardiovascular disease, metabolic syndrome, and neurodegenerative disorders. As endogenous polyamine synthesis declines with age, dietary intake becomes increasingly important, especially in older adults.

Here we show a current landscape of studies and strategies implemented for the study of polyamine metabolism, as well as molecular aspects that implicate the role of key enzymes, transport proteins, inhibitors, and the study of novel molecules as potential therapeutic targets. This review focused on the synthesis, interconversion and function of these molecules in Trichomonas vaginalis, a common sexually transmitted parasite of humans.

Cryptosporidium parvum infection of T-cell receptor alpha (TCR-alpha)-deficient mice results in a persistent infection. In this study, treatment with a polyamine analogue (SL-11047) prevented C. parvum infection in suckling TCR-alpha-deficient mice and cleared an existing infection in older mice. Treatment with putrescine, while capable of preventing infection, did not clear C. parvum from previously infected mice. These findings provide further evidence that polyamine metabolic pathways are targets for new anticryptosporidial chemotherapeutic agents.
