The Diversity of Yellow-Related Proteins in Sand Flies (Diptera: Psychodidae).

Yellow-related proteins (YRPs) present in sand fly saliva act as affinity binders of bioamines, and help the fly to complete a bloodmeal by scavenging the physiological signals of damaged cells. They are also the main antigens in sand fly saliva and their recombinant form is used as a marker of host exposure to sand flies. Moreover, several salivary proteins and plasmids coding these proteins induce strong immune response in hosts bitten by sand flies and are being used to design protecting vaccines against Leishmania parasites. In this study, thirty two 3D models of different yellow-related p

Ionizable Nitroxides for Studying Local Electrostatic Properties of Lipid Bilayers and Protein Systems by EPR.

Electrostatic interactions are known to play a major role in the myriad of biochemical and biophysical processes. Here, we describe biophysical methods to probe local electrostatic potentials of proteins and lipid bilayer systems that are based on an observation of reversible protonation of nitroxides by electron paramagnetic resonance (EPR). Two types of probes are described: (1) methanethiosulfonate derivatives of protonatable nitroxides for highly specific covalent modification of the cysteine's sulfhydryl groups and (2) spin-labeled phospholipids with a protonatable nitroxide tethered to t

Interplay of electrostatics and lipid packing determines the binding of charged polymer coated nanoparticles to model membranes.

Understanding of nanoparticle-membrane interactions is useful for various applications of nanoparticles like drug delivery and imaging. Here we report on the studies of interaction between hydrophilic charged polymer coated semiconductor quantum dot nanoparticles with model lipid membranes. Atomic force microscopy and X-ray reflectivity measurements suggest that cationic nanoparticles bind and penetrate bilayers of zwitterionic lipids. Penetration and binding depend on the extent of lipid packing and result in the disruption of the lipid bilayer accompanied by enhanced lipid diffusion. On the

Evidence for substrate binding-induced zwitterion formation in the catalytic Cys-His dyad of the SARS-CoV main protease.

The coronavirus main protease (M(pro)) represents an attractive drug target for antiviral therapy of coronavirus (CoV) infections, including severe acute respiratory syndrome (SARS). The SARS-CoV M(pro) and related CoV proteases have several distinct features, such as an uncharged Cys-His catalytic dyad embedded in a chymotrypsin-like protease fold, that clearly separate these enzymes from archetypical cysteine proteases. To further characterize the catalytic system of CoV main proteases and to obtain information about improved inhibitors, we performed comprehensive simulations of the proton-t

Comparing aminoglycoside binding sites in bacterial ribosomal RNA and aminoglycoside modifying enzymes.

Aminoglycoside antibiotics are used against severe bacterial infections. They bind to the bacterial ribosomal RNA and interfere with the translation process. However, bacteria produce aminoglycoside modifying enzymes (AME) to resist aminoglycoside actions. AMEs form a variable group and yet they specifically recognize and efficiently bind aminoglycosides, which are also diverse in terms of total net charge and the number of pseudo-sugar rings. Here, we present the results of 25 molecular dynamics simulations of three AME representatives and aminoglycoside ribosomal RNA binding site, unliganded

Efficient immobilization of glucose oxidase by in situ photo-cross-linking for glucose biosensing.

A glucose biosensor was fabricated based on electrostatic self-assembly in combination with in situ photo-cross-linking of glucose oxidase (GOx) and diazoresin-chitosan (DAR-CS) on Prussian blue deposited multi-walled carbon nanotubes (PB-MWNTs) backbone. It was demonstrated that GOx was initially ionically deposited and subsequently covalently photo-cross-linked onto the PB-MWNTs backbone using photosensitive DAR-CS as the assembly interlayer. The modified electrode exhibited good electrical conductivity and effective electron transfer mediation toward H(2)O(2) reduction due to the employment

Influence of polymer-surfactant aggregates on fluid flow.

This paper describes the influence of interactions of poly(ethylene oxide) (PEO) with cationic cetyltrimethylammonium bromide (CTAB) micelles on drag reduction. Since the interactions between PEO and CTAB micelles alone are weak, salicylate ions were used as CTAB counterions. They facilitate formation of polymer-micelle aggregates by screening the electrostatic repulsions between the charged surfactant headgroups. The influence of polymer-surfactant interactions on drag reduction is of biomedical engineering importance. Drag reducing additives introduced to blood produce beneficial effects on

Protein design for diversity of sequences and conformations using dead-end elimination.

Proteins, especially antibodies, are widely used as therapeutic and diagnostic agents. Computational protein design is a powerful tool for improving the affinity and stability of these molecules. We describe a protein design method which employs the dead-end elimination (DEE) and A* discrete search algorithms with a few improvements aimed at making the procedure more useful for actual projects to design proteins for better affinity or stability. DEE/A* and related algorithms allow vast search spaces of protein sequences and their alternative side chain conformations ("rotamers") to be systemat

Evaluation of an electrostatic toxicity model for predicting Ni(2+) toxicity to barley root elongation in hydroponic cultures and in soils.

Assessing environmental risks of metal contamination in soils is a complex task because the biologically effective concentrations of metals in soils vary widely with soil properties. The factors influencing the toxic effect of nickel (Ni) on root growth of barley (Hordeum vulgare) were re-evaluated using published data from both soil and hydroponic cultures. The electrical potential (ψ(0) (o) ) and ion activities ({I(z) }(0) (o) ) at the outer surfaces of root-cell plasma membranes (PMs) were computed as the basis of the re-evaluation. The reanalyses demonstrated that root growth was related t

The sulfated triphenyl methane derivative acid fuchsin is a potent inhibitor of amyloid formation by human islet amyloid polypeptide and protects against the toxic effects of amyloid formation.

Islet amyloid polypeptide (IAPP), also known as amylin, is responsible for amyloid formation in type 2 diabetes. The formation of islet amyloid is believed to contribute to the pathology of the disease by killing beta-cells, and it may also contribute to islet transplant failure. The design of inhibitors of amyloid formation is an active area of research, but comparatively little attention has been paid to inhibitors of IAPP in contrast to the large body of work on beta-amyloid, and most small-molecule inhibitors of IAPP amyloid are generally effective only when used at a significant molar exc

Electrostatic binding of substituted metal phthalocyanines to enterobacterial cells: its role in photodynamic inactivation.

The effect of ionic substituents in zinc and aluminum phthalocyanine molecules and of membrane surface charge on the interaction of dyes with artificial membranes and enterobacterial cells, as well as on photosensitization efficiency was studied. It has been shown that increasing the number of positively charged substituents enhances the extent of phthalocyanine binding to Escherichia coli cells. This, along with the high quantum yield of singlet oxygen generation, determines efficient photodynamic inactivation of Gram-negative bacteria by zinc and aluminum octacationic phthalocyanines. The ef

Colloidal deposition on remotely controlled charged micropatterned surfaces in a parallel-plate flow chamber.

This article describes a method to influence colloid deposition by varying the zeta potential at microelectrodes with remotely applied electric potentials. Deposition experiments were conducted in a parallel-plate flow chamber for bulk substrates of glass, indium tin oxide (ITO), and ITO-coated glass microelectrodes in 10 and 60 mM potassium chloride solutions. Colloid deposition was found to be a function of solution chemistry and the small locally delivered electric surface potentials. Electric fields and physical surface heterogeneity can be ruled out as cause of the observed deposition. Re

Electron-hole dynamics in CdTe tetrapods.

We present transient absorption studies with femtosecond time resolution on the electron-hole dynamics in CdTe tetrapod nanostructures. Electron-hole pairs are generated by optical excitation in the visible spectral range, and an immediate bleach and induced absorption signal are observed. The relaxation dynamics to the lowest excitonic state is completed in about 6 ps. Experiments with polarized excitation pulses give information about the localization of the excited-state wave functions. The influence of the nanocrystal shape on the optical properties of CdTe nanoparticles is discussed.

Solution structures of a cyanobacterial metallochaperone: insight into an atypical copper-binding motif.

The Atx1 copper metallochaperone from Synechocystis PCC 6803, ScAtx1, interacts with two P(1)-type copper ATPases to supply copper proteins within intracellular compartments, avoiding ATPases for other metals en route. Here we report NMR-derived solution structures for ScAtx1. The monomeric apo form has a betaalphabetabetaalpha fold with backbone motions largely restricted to loop 1 containing Cys-12 and Cys-15. The tumbling rate of Cu(I)ScAtx1 (0.1-0.8 mm) implies dimers. Experimental restraints are satisfied by symmetrical dimers with Cys-12 or His-61, but not Cys-15, invading the copper sit

Specific compositions of amyloid-beta peptides as the determinant of toxic beta-aggregation.

Alzheimer's disease (AD) may be caused by toxic aggregates formed from amyloid-beta (Abeta) peptides. By using Thioflavin T, a dye that specifically binds to beta-sheet structures, we found that highly toxic forms of Abeta-aggregates were formed at the initial stage of fibrillogenesis, which is consistent with recent reports on Abeta oligomers. Formation of such aggregates depends on factors that affect both nucleation and elongation. As reported previously, addition of Abeta42 systematically accelerated the nucleation of Abeta40, most likely because of the extra hydrophobic residues at the C

A novel view of pH titration in biomolecules.

When individual titratable sites in a molecule interact with each other, their pH titration can be considerably more complex than that of an independent site described by the classical Henderson-Hasselbalch equation. We propose a novel framework that decomposes any complex titration behavior into simple standard components. The approach maps the set of N interacting sites in the molecule onto a set of N independent, noninteracting quasi-sites, each characterized by a pK'(a) value. The titration curve of an individual site in the molecule is a weighted sum of Henderson-Hasselbalch curves corres

Affinity extraction of BSA with reversed micellar system composed of unbound Cibacron Blue.

Affinity Cibacron Blue 3GA (CB) dye in aqueous phase was directly transferred to the reversed micelles due to electrostatic interaction between anionic CB and cationic cetyltrimethylammonium bromide (CTAB). The bovine serum albumin (BSA) transfer to the reverse micelles increases significantly in a wide range of pH by the addition of a small amount of CB ( approximately 1.0-7.0% of the total surfactant concentration) to the aqueous phase. For pH < pI, the selectivity can be significantly improved with the presence of affinity CB because no BSA was extracted in the absence of CB. For backward e

Intelligent Acousto-Electrical Metamaterials (IAM) for Sound Source Detection.

Acoustic transducers are essential for object localization and environmental sensing. Conventional transducers rely on piezoelectric crystals, whose acoustic-electric response is fixed by the crystal lattice's inherent asymmetry and orientation. This results in static coupling behavior, necessitating bulky arrays of rigid elements with complex wiring and high computational demands for directional sensing. Here, we report a fundamentally new class of acoustic-electric coupling that emerges from topology-governed charge transport in 3D micro-architected piezoelectric metamaterials. Unlike single

Refinements in the technique of 'awake' electrical nerve stimulation in the management of chronic low ulnar nerve injuries.

The standard technique in the management of chronic low ulnar nerve injuries includes excision of the neuroma and reconstruction using sural nerve grafts in the fully anaesthetised patient. It has been shown that using this standard technique, disappointing results may be observed and that significant improvement in results could be obtained if intra-operative matching of sensory and motor fascicles is performed. This study reports on eight patients with chronic ulnar nerve injuries managed using the technique of electrical fascicular orientation and sural nerve grafting. In all patients, intr

Piezoelectric Nanofibrous Scaffolds for Promoting the Growth of Spiral Ganglion Neurons via Acoustic Stimulation.

The loss of spiral ganglion neurons (SGNs) and hair cells often leads to sensorineural hearing loss (SNHL), whose therapeutic effect depends on the quality and functional integrity of the residual SGNs. Compared with traditional pharmacological therapies, tissue engineering methods have been developed to provide a biomimetic environment for SGN regeneration, but static strategies and non-responsive scaffolds still limit the therapy efficiency. Here, we present a composite piezoelectric nanofibrous scaffold responsive to acoustic stimulation to promote the growth of SGNs. The oriented nanofibro
