A novel approach for the production of [125I]amatoxin and anti-amanitin is described. The antigen and the starting material for Bolton Hunter iodination is prepared by periodic acid oxidation of the gamma-delta-dihydroxy-isoleucine side chain of alpha-amanitin followed by reductive amination. The antigen seems to be of low apparent toxicity. Chemical, spectroscopic and immunological evidence suggests that the hapten has a secondary structure similar to alpha-amanitin, but a modified tryptophanyl ring system. New methods for the microdetermination of ascorbic acid are described. The 3 proposed methods based on the fact that N-chlorosuccinimide in acid, alkaline or neutral medium readily oxidizes ascorbic acid. The mechanism of the reaction is discussed. The proposed methods are simple rapid, and sufficiently accurate to determine concentrations as low as 10 micrograms of ascorbic acid. The experimental error does not exceed +/- 2%. The methods were also used for the determination of ascorbic acid in such biological fluids as bloody, urine or edible fruits with an error not exceeding +/- 5%.

This study examines the decomposition of the protonated dipeptide, AsnSer methylated on the serine side chain, Asn(OMe)Ser. α-Glucosidase is considered as a therapeutic target for the treatment of type 2 diabetes mellitus (DM2). In current study, we synthesized pyrrolidine-2,5-dione (succinimide) and thiazolidine-2,4-dione derivatives and evaluated for their ability to inhibit α-Glucosidase. Pyrrolidine-2,5-dione derivatives (11a-o) showed moderate to poor α-glucosidase inhibition. Compound 11o with the IC50 value of 28.3 ± 0.28 µM emerged as a good inhibitor of α-glucosidase. A series of N-(4-amino-2-butynyl)-5-methyl-2-pyrrolidones modified only in the amino group was synthesized. The compounds were agonists, partial agonists, and antagonists on the isolated guinea pig ileum.
