The role of superoxide anion generation in antibody-dependent polymorphonuclear leukocyte-mediated cytotoxicity.

Human polymorphonuclear leukocytes (PMNs) can act as killer cells in an antibody-dependent cellular cytotoxicity (ADCC) system. Cytolysis is rapid and detectable at low effector/target cell ratios. PMNs from a patient affected by chronic granulomatous disease (CGD) exhibit an absolute defect in ADCC. In order to investigate the role of the toxic oxygen derivatives (H2O2 and superoxide anions) lacking in CGD, experiments in presence of specific inhibitors were carried out. Superoxide dismutase, an O-2 scavenger, displayed a significant inhibition activity in a dose dependent way. On the other h

Evaluation of negative staining technique for determination of CN--insensitive superoxide dismutase activity.

The different forms of superoxide dismutase (superoxide-superoxide oxidoreductase, EC 1.15.11) have been studied, in tissues of rat, mouse and chicken, by the ectrophoresis-nitro blue tetrazolium technique proposed by Beauchamp. Similar enzyme patterns were evident in every tissue. A fast migrating CN--sensitive form of dismutase activity was present in isolated liver mitochondria of each species. Chicken and mouse liver mitochondria, as well as whole homogenate of every tissue of these two species, showed two additional slow-migrating bands of CN--insensitive activity. In contrast, such bands

Production and some properties of catalase and superoxide dismutase from the anaerobe Bacteroides distasonis.

The catalase level of Bacteroides distasonis (ATCC 8503, type strain) varied with the amount of hemin supplied to the medium when the cells were grown in either a prereduced medium containing 0.5% peptone, 0.5% yeast extract, and 1% glucose or in a prereduced, defined heme-deficient medium. The effect of hemin on catalase production could not be duplicated by ferrous sulfate or ferrous ammonium citrate. Catalase activity reached peak values in late log phase, whereas superoxide dismutase specific activity remained constant throughout the culture growth cycle. The catalase was a nondialyzable,

A pulse-radiolysis study of the manganese-containing superoxide dismutase from Bacillus stearothermophilus.

In the preceding paper the mechanism of catalysis of the manganese-containing superoxide dismutase from Bacillus stearothermophilus was shown to involve a 'fast cycle' and a 'slow cycle' [McAdam, Fox, Lavelle & Fielden, 1977 (Biochem. J. 165, 71-79)]. Further properties of the enzyme was considered in the present paper. Pulse-radiolysis studies, under conditions of low substrate concentration to (i.e. when the fast cycle predominates), showed that enzyme activity decreases as pH increases (6.5-10.2). Activity was unaffected by the addition of H2O2 or NaN3 but slightly decreased by KCN. Both H2

Immunocytochemical study of the distribution of the free radical scavenging enzymes Cu/Zn superoxide dismutase (SOD1); MN superoxide dismutase (MN SOD) and catalase in the normal human spinal cord and in motor neuron disease.

Interest in free radical-mediated mechanisms of motor neuron injury has arisen following the discovery of point mutations in the Cu/Zn superoxide dismutase (SOD1) gene in a proportion of cases of familial motor neuron disease (MND). Evidence is emerging which indicates that oxidative stress may contribute to the pathophysiology of sporadic MND. Free radical scavenging enzymes form a major component of the anti-oxidant defense system. The aim of this study was to compare the distribution and density of immunoreactivity to Cu/Zn SOD. Mn SOD and catalase in the spinal cord of sporadic MND cases (

SKQ1 promotes tumor cell apoptosis by directly interacting with NDUFV2 and inducing superoxide production.

SKQ1, a mitochondria-targeted antioxidant composed of triphenylphosphonium (TPP+) and plastoquinone, has been reported to exhibit antitumor potential; however, its underlying mechanism of action and specific molecular targets remain largely unclear. Here, we investigated the antitumor efficacy of SKQ1 in vitro using human hepatocellular carcinoma cells (MHCC97-H) and murine colorectal cancer cells (MC38), and in vivo using a human-derived hepatocellular carcinoma xenograft model and a murine syngeneic colorectal cancer model. We found that SKQ1 induced apoptosis and inhibited the proliferation

Superoxide-Anion-Dominated Labeling of Graphene Defects in an Interfacial Nanoreactor of Electron-Donated Substrate.

Harnessing the chemical reactivity of defects delivers a fundamental tool for precise nanoscale characterization of graphene defects. Herein, we develop a confined photochemical strategy based on the photosensitization of dye molecules targeted at the chemically active defects of graphene to label the defects. The graphene/copper interface enriches the electrons of graphene defects to construct a nanoreactor within defect-positioning interfaces, which modulates the preferential adsorption and the photosensitization reaction pathway of photoexcited methylene blue (MB). The nanoscale reactor pre

Superoxide Dismutase-Centered Modulation by Curcumin in Cardiovascular Diseases: Mechanistic Insights and Translational Implications.

Cardiovascular diseases (CVD) remain the leading global cause of morbidity and mortality, driven in part by dysregulated redox homeostasis and chronic inflammation. Superoxide dismutase (SOD), a key enzymatic defence against reactive oxygen species (ROS), plays a central role in maintaining cardiovascular integrity through regulation of oxidative stress across cytosolic (SOD1), mitochondrial (SOD2) and extracellular (SOD3) compartments. Impairment of SOD function contributes directly to endothelial dysfunction, myocardial injury and vascular remodelling. Curcumin (Cur), a pleiotropic polypheno

Correlation between Polymorphisms of the Superoxide Dismutase Gene and Idiopathic Nephrotic Syndrome in Chinese Children.

BACKGROUND: Genetic mutations are closely linked to childhood idiopathic nephrotic syndrome (INS). This study aimed to explore the associations between superoxide dismutase (SOD) gene polymorphisms and susceptibility to INS, as well as response to steroid therapy, in Chinese children. METHODS: Five single nucleotide polymorphisms (SNPs; rs4816407, rs1041740, rs4880, rs2758346, and rs3798215) in the SOD1 and SOD2 genes were genotyped using multiplex PCR combined with next-generation sequencing in 183 pediatric patients with INS and 100 healthy controls. RESULTS: The genotypic distributions of

Intranasal delivery of cerium oxide nanozyme and superoxide dismutase in exosomes for Parkinson's disease therapy.

The excessive reactive oxygen species (ROS) play an important role in the occurrence and progression of Parkinson's disease (PD). To investigate whether the antioxidant enzymes can scavenge the ROS level in vivo and treat PD efficiently, superoxide dismutase (SOD) was encapsulated into HEK293-derived exosomes to prepare SOD-loaded exosomes (SOD@EXO), and catalase (CAT)-like cerium oxide nanozyme (CeO2) was mixed with SOD@EXO to construct the formulation, namely SOD@EXO + CeO2. The formulation with cup-shaped morphology showed high SOD and CAT activities, which could scavenge the ROS level in a

Molecular Characterization of Superoxide Dismutase and Its Putative Relationship With Drug Resistance in Eimeria Tenella.

Avian coccidiosis, an intestinal parasitic disease caused by Eimeria spp., remains one of the most economically significant diseases in poultry. Although anticoccidial drugs have been effective in controlling coccidiosis, the emergence of drug resistance in Eimeria poses a persistent challenge to disease management. Our previous study revealed differential expression of superoxide dismutase (EtSOD) between drug-sensitive (DS) and drug-resistant Eimeria tenella strains. In this study, qPCR and western blot analyses demonstrated that EtSOD expression was significantly upregulated in diclazuril-r

The ArcAB two-component system is associated with the susceptibility of Aggregatibacter actinomycetemcomitans to superoxide and hydrogen peroxide.

Aggregatibacter actinomycetemcomitans is a Gram-negative facultative anaerobe and is associated with periodontal disease. This bacterium is exposed to environmental stresses, such as osmotic pressure, temperature shifts, pH shifts, and antimicrobial substances, including reactive oxygen species (ROS), in the human oral cavity. The bacterial two-component system ArcAB modulates gene expression in response to environmental changes, primarily by sensing oxygen pressure in several pathogens belonging to the γ-proteobacteria. It is also known to provide adaptation to ROS stress; however, its functi

Electrocatalytic methane conversion via in-situ generated superoxide radicals in an aprotic ionic liquid.

The electrochemical activation and partial oxidation of methane are highly attractive to enable the direct conversion in a sustainable and decentralized way. Herein, we report an electrochemical system in a non-diaphragm electrochemical bath to convert CH4 to CH3OH and CH3CH2OH at room temperature, in which V3O7·H2O as the anodic catalyst to activate CH4 and an aprotic ionic liquid [BMIM]BF4 as supporting electrolyte to control superoxide radicals (O2-) as the main active oxygen species generated on cathode. As a result, methanol and ethanol were identified as the liquid products, and the supe

The Pathophysiology of Alcohol-Associated Liver Disease: Focusing on Superoxide Dismutase 1 as a Therapeutic Target.

Alcohol-associated liver disease (ALD) is a major health problem of global importance, caused by chronic alcohol consumption, leading to the accumulation of reactive oxygen species (ROS) and subsequent oxidative stress-a central mechanism in liver injury. Superoxide dismutase 1 (SOD1), a Cu-Zn containing antioxidant enzyme, plays a crucial role in attenuating ALD-induced oxidative stress triggered by ethanol metabolism. However, alcohol exposure, whether chronic, acute or binge, differentially affects SOD1 levels, either diminishing its expression or temporarily compensating for alcohol-induce

Assembly of Genetically Engineered Ionizable Protein Nanocage-based Nanozymes for Intracellular Superoxide Scavenging.

Nanozymes play a pivotal role in mitigating excessive oxidative stress, however, determining their specific enzyme-mimicking activities for intracellular free radical scavenging is challenging due to endo-lysosomal entrapment. In this study, we employ a genetic engineering strategy to generate ionizable ferritin nanocages (iFTn), enabling their escape from endo-lysosomes and entry into the cytoplasm. Specifically, ionizable repeated Histidine-Histidine-Glutamic acid (9H2E) sequences are genetically incorporated into the outer surface of human heavy chain FTn, followed by the assembly of variou

A superoxide anion responsive and self-reporting fluorescent H2S donor for the treatment of diabetic wound.

Superoxide anion (O2•-) not only serves as a critical precursor for numerous damaging reactive oxygen species (ROS), but also is implicated in a variety of diseases, including cancer, cardiovascular disorders, and diabetes. Consequently, reducing the levels of superoxide anions and alleviating oxidative stress are of paramount importance. Conversely, hydrogen sulfide (H2S), recognized as a significant biological signaling molecule, plays vital roles in protecting mammalian cells from oxidative damage and promoting tissue regeneration. In this study, we reported a novel superoxide anion-respons

Mapping transcriptomic signatures downstream of mitochondrial superoxide dismutase SOD2 in OVCA433 multicellular aggregates.

During metastatic spread, tumor cells adapt to evade anchorage-independent cell death by upregulating mitochondrial antioxidant systems. We previously showed that ovarian cancer cells upregulate mitochondrial manganese superoxide dismutase (SOD2) following detachment and multicellular aggregate (MCA) formation. SOD2 scavenges mitochondrial superoxide and manipulates cellular hydrogen peroxide levels, both functions necessary for metastasis. Here, we investigated SOD2's metastatic function by assessing its transcriptomic effects in OVCA433 ovarian cancer cells cultured in low-attachment conditi

1,2,4,5-Tetrazine-tethered probes for fluorogenically imaging superoxide in live cells with ultrahigh specificity.

Superoxide (O2·-) is the primary reactive oxygen species in mammal cells. Detecting superoxide is crucial for understanding redox signaling but remains challenging. Herein, we introduce a class of activity-based sensing probes. The probes utilize 1,2,4,5-tetrazine as a superoxide-responsive trigger, which can be modularly tethered to various fluorophores to tune probe sensitivity and emission color. These probes afford ultra-specific and ultra-fluorogenic responses towards superoxide, and enable multiplexed imaging of various cellular superoxide levels in an organelle-resolved way. Notably, th
