Can overeating induce conditioned taste avoidance in previously food restricted rats?

While feeding is rewarding, the feeling of satiation has been theorized to have a mixed affect. Using a food restriction model of overeating we examined whether bingeing was capable of supporting conditioned taste avoidance (CTA). Adult male Sprague-Dawley rats were maintained on either an ad lib (n=8) or restricted (50% of regular consumption; n=24) food access for 20 days. On Days 9, 14, and 19 all rats were given access to a novel saccharin solution in place of water, and two groups of food restricted rats were given access to either 100% of regular food consumption or ad lib food. Ad lib a…

Duodenal pain and spinal morphine induce conditioned taste aversion in rats.

Conditioned taste aversion (CTA) is a behavioural response essential to the survival of an individual. The combination of taste and odour of most foods provides a strong conditioned stimulus (CS) for an animal to respond in an appropriate way to any harmful unconditioned stimuli (US) that follow. The most widely used conditioned stimuli are drinkable sweet solutions, such as saccharin and sucrose. CTA-like responses are also found for environmental unconditioned stimuli, but these usually take longer training. In the present study, the aversive nature of a duodenal distention with an implanted…

d-fenfluramine's effects on normal ingestion assessed with taste reactivity measures.

The effects of d-fenfluramine on intake and on hedonic responses to taste stimuli in rats were investigated using a modified taste reactivity paradigm. Subjects (n = 15) were first trained to consume a 3% sucrose solution. They were then pretreated with d-fenfluramine (0.3-3.0 mg/kg, i.p.), and tested with access to either 3% sucrose, or a 0.01% quinine HCl solution. In the modified taste reactivity test, chronic oral cannulation was not used; instead, taste reactivity measures were scored during periods of noningestion in a voluntary intake test. d-Fenfluramine reliably reduced both sucrose a…

Chemosensory dysfunction. Clinical evaluation results from a taste and smell clinic.

Findings from 441 patient evaluations performed at the Taste and Smell Clinic of the Connecticut Chemosensory Clinical Research Center are presented. Taste and smell dysfunction was the chief complaint in all patients. The diagnostic approach included tests of taste and smell function; history taking; physical, neurologic, otorhinolaryngologic, and nutritional examinations; and clinical laboratory screening tests. Results indicate that the most common chemosensory dysfunction is probably olfactory: 86% of patients in this series had measurable loss of smell function. The most common cause of o…

Delayed taste aversion learning in preweanling rats exposed to alcohol prenatally.

Parallels between the behavioral profiles of rats exposed to alcohol prenatally and those with hippocampal damage suggest that hippocampal dysfunction may underlie some of the behavioral abnormalities resulting from prenatal alcohol exposure. Because of possible hippocampal involvement in the acquisition of a delayed conditioned taste aversion, this task was assessed in rat pups exposed to alcohol prenatally. Long-Evans rats were maintained on liquid diets containing either 35% or 0% ethanol derived calories from Days 6-20 of gestation. Pair-feeding procedures and an ad lib Lab Chow (LC) group…

Identification and taste characteristics of novel salt taste-enhancing peptides from defatted Antarctic krill (Euphausia superba) powder hydrolysate and their molecular mechanisms of interaction with the TMC4 channel.

This study aimed to prepare and identify novel salt taste-enhancing peptides from defatted Antarctic krill (Euphausia superba) powder hydrolysate (DAKPH), and investigate molecular mechanisms. Six potential peptides were screened via virtual screening from 36,014 peptides identified through UHPLC-MS/MS. Sensory evaluation and electronic tongue analysis verified that 0.4 mg/mL EDLEY, WDDDP, EDYEF, ERDY, and DRPWE significantly enhanced the salt taste perception of 3 mg/mL NaCl (p < 0.05). Molecular docking and molecular dynamics simulation results indicated that these five peptides entered the …

Genetic variations underlying bitter and sweet taste perception are associated with food and beverage intake habits and taste-modifying behaviors among Dutch adults with a prior myocardial infarction.

BACKGROUND: Taste perception influences eating behaviors and varies partly based on genetics. Genome-wide association studies (GWAS) have identified many variants related to bitter, sweet, salt, sour, and umami perception (taste-related variants), but whether these variants inform eating behaviors among patients with cardiovascular disease (CVD) is unknown. This study investigated the associations of taste-related variants with eating behaviors in adults with a prior myocardial infarction (MI) and explored the potential for sex-specific effects. METHODS: Analyzed, were 4,204 Dutch adults with…

Salt-induced effect on the alcohol-stimulative taste in London dry gin.

During the manufacture of London dry gin, large quantities of water are added to dilute the alcohol content before bottling. In this study, proton nuclear magnetic resonance (1H NMR) spectroscopy was employed to determine whether the presence of salts in the dilution water could potentially reduce the alcohol-stimulative taste of gin. Specifically, the effect of MgCl2 was investigated for EtOH-H2O mixtures and three gin samples prepared on the laboratory scale using different types of botanicals, including juniper berries and coriander seeds. The results revealed that MgCl2 strengthened the wa…

Retention of Sweet Taste in Sirenians Reflects Ecological Adaptation in Marine Mammals.

Taste perception is vital for animal survival. Among marine mammals, whales and pinnipeds have lost all sweet and umami taste receptor genes, whereas sirenians (manatees and dugongs) have retained them. The leading hypotheses for taste loss in marine mammals involve feeding behavior, dietary shifts, and high sodium concentrations. To characterize the evolution of sweet taste in sirenians, we analyzed the complete repertoire of sweet taste receptor genes (Tas1r2 and Tas1r3) and their coding sequences in 53 mammal species. Among marine mammals, only sirenians possessed intact sequences for both …

Taste Dysfunction in Oral Candidiasis: Impact of Candida Carriage and Hyphal Presence.

BACKGROUND: Taste dysfunction is a common symptom of oral candidiasis; however, its underlying mechanisms remain unclear. OBJECTIVES: This study aimed to determine whether Candida albicans carriage, particularly its hyphal phenotype, is associated with impaired taste sensitivity. PATIENTS/METHODS: Fifty-seven participants were divided into three groups: noncarriers (n = 20), carriers without hyphae (n = 20), and carriers with hyphae (n = 17). Tongue biofilm samples were collected for Candida culture and smear tests. Salivary flow rates were measured, and taste sensitivity was assessed using …

Circadian clock-gated cell renewal controls time-dependent changes in taste sensitivity.

Circadian regulation of the cell cycle progression generates a diurnal supply of newborn cells to replace those lost in organs and tissues. In this study, we analyzed circadian time-dependent changes in cell types within the mouse tongue epithelium. Using single-cell RNA sequencing, we observed circadian time-dependent changes in the populations of stem/progenitor cells and the differentiated cells in mice tongues. Notably, we observed time-dependent changes in the type II taste cell population, which were abolished by ablation of taste bud stem cells, thereby inhibiting cell proliferation wit…

Four individually identified paired dopamine neurons signal taste punishment in larval Drosophila.

Dopaminergic neurons (DANs) play key roles in processing rewards and punishments across species. They evaluate sensory input, store memories, and update them based on relevance. To understand how individual DANs contribute to these functions, we studied Drosophila larvae, which have only about 120 DANs. Only eight of these project to the mushroom body (MB), a center for olfactory learning. These eight are divided into the pPAM and DL1 clusters, with four DANs each. We confirmed that pPAM neurons in the MB medial lobe encode sugar rewards. In the DL1 cluster, four neurons-DAN-c1, DAN-d1, DAN-f1

Taste Masking Study of Orally Disintegrating Film (ODF) Formulations Containing Memantine Hydrochloride.

Geriatric patients have difficulty to comply to their medication regimen due to complicated medication administration schedule, dysphagia, reduced ability to swallow tablets and dementia. This is particularly more challenging among the Alzheimer Disease's patients. Therefore, a model drug, memantine hydrochloride has been formulated into an orally disintegrating film (ODF) for easier consumption. However, bitter taste of memantine hydrochloride needs to be solved first if the drug is formulated into an ODF. The objective of this study is to taste mask memantine hydrochloride ODF and conduct a …

Strategies for Taste Masking of Orodispersible Dosage Forms: Time, Concentration, and Perception.

Orodispersible dosage forms, characterized as quick dissolving and swallowing without water, have recently gained great attention from the pharmaceutical industry, as these forms can satisfy the needs of children, the elderly, and patients suffering from mental illnesses. However, poor taste by thorough exposure of the drugs' dissolution in the oral cavity hinders the effectiveness of the orodispersible dosage forms. To bridge this gap, we put forward three taste-masking strategies with respect to the intensity of time, concentration, and perception. We further investigated the raw material pr…

Sweet Taste Is Complex: Signaling Cascades and Circuits Involved in Sweet Sensation.

Sweetness is the preferred taste of humans and many animals, likely because sugars are a primary source of energy. In many mammals, sweet compounds are sensed in the tongue by the gustatory organ, the taste buds. Here, a group of taste bud cells expresses a canonical sweet taste receptor, whose activation induces Ca2+ rise, cell depolarization and ATP release to communicate with afferent gustatory nerves. The discovery of the sweet taste receptor, 20 years ago, was a milestone in the understanding of sweet signal transduction and is described here from a historical perspective. Our review brie…
