Emerging chitosan grafted essential oil components: A review on synthesis, characterization, and potential application.

Chitosan is a green, low-cost natural polymer material. The bioactivity of chitosan can be enhanced by coupling with essential oil chemicals, which can broaden the range of applications. Essential oil-chitosan (EOCS), the chitosan modified by essential oil components, which has been widely used as a multi-perspective in numerous filed of food and life in recent years. Herein, the synthesis, characterization, and recent application of EOCS are reviewed. Synthetic methods include 1-ethyl-3-(3-dimethylaminopropyl) carbodiimide (EDC)-mediated modification, free radical-induced modification, enzyme

Genetic Transformation and Complementation.

The disciplines of Borrelia (Borreliella) burgdorferi microbiology and Lyme disease pathogenesis have come to depend on the genetic manipulation of the spirochete. Generating mutants in these recalcitrant bacteria, while not straightforward, is routinely accomplished in numerous laboratories, although there are several crucial caveats to consider. This chapter describes the design of basic molecular genetic experiments as well as the detailed methodologies to prepare and transform competent cells, select for and isolate transformants, and complement or genetically restore mutants.

Development of a Tool for High-Efficiency, Markerless and Iterative Genome Editing in Shouchella clausii.

Shouchella clausii is a spore-forming, Gram-positive bacterium with intrinsic antibiotic resistance and promising potential in biotherapeutics, industrial biotechnology and environmental applications. Its genetic intractability, due to a rigid cell wall and lack of natural competence, has limited its development as a microbial chassis. To facilitate its genetic transformation, a hyperosmotic electroporation protocol was optimised using cell wall weakening agents, achieving efficiencies comparable to other recalcitrant bacilli. A comprehensive and reusable genetic tool was developed centred on

Trimethoprim resistance in urinary bacteria isolated from patients attending general practitioners.

The incidence of trimethoprim resistance amongst enterobacterial strains responsible for significant bacteriuria in patients attending general practitioners in Edinburgh was 11.4%. Two-thirds of these resistant strains were highly resistant to trimethoprim. Trimethoprim resistance was more prevalent in bacteria isolated from men and from elderly patients. Less than half of the highly resistant strians possessed trimethoprim resistance plasmids; however, amongst those that did, one plasmid predominated. This plasmid was identical with the most successful trimethoprim resistance plasmid in hospi

Killing of Haemophilus influenzae cells by integrated ultraviolet-induced lesions from transforming deoxyribonucleic acid.

Highly competent cultures of Haemophilus influenzae are inactivated by exposure to transforming deoxyribonucleic acid (DNA) irradiated with ultraviolet light (UV). As a function of UV dose to the DNA, the killing goes to a maximum and then decreases. The killing of H. influenzae by unirradiated H. parainfluenzae DNA, reported by other workers, is enhanced by low doses of UV, but drops off at high doses. Since there are no such lethal effects in a strain of H. influenzae that takes up DNA normally but does not integrate it, it is concluded that the killing is associated with integrated UV lesio

Cooperation of the multidrug efflux pump and lipopolysaccharides in the intrinsic antibiotic resistance of Salmonella enterica serovar Typhimurium.

OBJECTIVES: In Gram-negative bacteria, drug susceptibility is associated with multidrug efflux systems and an outer membrane (OM) barrier. Previous studies revealed that Salmonella enterica serovar Typhimurium has 10 functional drug efflux pumps. Among them, AcrB is a major factor to maintain the intrinsic drug resistance in this organism. The lipopolysaccharide (LPS) content of OM is also important for resistance to lipophilic drugs; however, the interplay between the multidrug efflux pump and LPS in the intrinsic antibiotic resistance of Salmonella remains to be studied in detail. The aim of

Unveiling nitrogen and sulfur cycling mechanisms of odor reduction in kitchen waste composting driven by exogenous bacterial consortia.

Odor emissions, primarily ammonia (NH3) and hydrogen sulfide (H2S) restrict the application of kitchen waste (KW) composting. Microbial inoculation is a promising strategy, yet mechanisms underlying odor mitigation remain unclear. KW composting with and without an immobilized bacterial consortium (IBC) was compared by monitoring physicochemical conditions, odor emissions, and microbial and functional profiles. IBC extended the thermophilic phase, improved composting efficiency, and reduced cumulative H2S (-44 %) and NH3 (-18 %). IBC reshaped bacterial, fungal and archaeal communities and stren

Study on TGF beta polymorphism in psoriatic patients with plaques infected with Streptococcuspyogenes.

Immune control and skin homeostasis depend on transforming growth factor-beta 1 (TGF-β1), which also may affect pathogenesis of psoriasis. Using MLVA typing, this study looked at the relationship between TGF-β1 509 C/T polymorphism and psoriasis susceptibility in an Iranian population as well as related Streptococcus pyogenes strains. Enrolled were fifty psoriatic patients overall as well as fifty matched healthy controls. Tetra Arms PCR was used for the extraction and analysis of genomic DNA; bacterial strain identification was achieved with MLVA. Though the TT genotype displayed a higher odd

Instant detection of extended-spectrum β-lactamase-producing bacteria from the urine of patients using infrared spectroscopy combined with machine learning.

Antibiotics are considered the most effective treatment against bacterial infections. However, most bacteria have already developed resistance to a broad spectrum of commonly used antibiotics, mainly due to their uncontrolled use. Extended-spectrum beta-lactamase (ESBL)-producing bacteria are an essential class of multidrug-resistant (MDR) bacteria. It is of extreme urgency to develop a method that can detect ESBL-producing bacteria rapidly for the effective treatment of patients with bacterial infectious diseases. Fourier transform infrared (FTIR) microscopy is a sensitive method that can rap

Removal of organic matter and phosphate using ferrihydrite for reduction of microbial regrowth potential.

Dissolved organic matter (DOM) and phosphate in reclaimed water promote microbial growth in distribution systems and deteriorate water quality. In this study, we tested ferrihydrite (Fh) for its potential to remove both DOM and phosphate in order to control bacterial regrowth. Adsorption kinetics on Fh revealed that phosphate has a higher affinity with Fh than has DOM. The removal efficiency of DOM increased at lower pH. Fourier transform infrared spectroscopy (FT-IR) spectra of freeze-dried Fh showed that adsorbed DOM was rich in carboxyl/hydroxyl functional groups, indicating anion exchange

Isolation and characterization of Pseudomonas sp. CBW capable of degrading carbendazim.

With the intensive application of carbendazim in greenhouse production of vegetables and the production of medicinal herbs, there is an increasing need to find a way to remediate carbendazim-contaminated soil. A bacterial stain capable of utilizing carbendazim as the sole source of carbon and energy was isolated from soil. The isolate was designated CBW and identified as a member of Pseudomonas sp. based on its colony morphology, 16S rRNA gene sequencing and Biolog analysis. About 87.1 and 99.1% of carbendazim at concentrations of 1.0 and 10.0 mg l(-1) in mineral salts medium were removed by t

Psychrophilic Bacteria and Enzyme Activity in Antarctic Ice Environments.

Psychrophilic organisms exhibit many adaptation mechanisms that enable them to survive at low temperatures ranging from -20 to 20 °C. Among other characteristics, these organisms produce cold-active enzymes with considerable biotechnological potential. The purpose of this study was to isolate and characterize bacteria from ice core samples collected on Horseshoe, Hovgaard, and Nansen Islands. Microorganisms from the latter two locations have been investigated only to a limited extent to date. Based on the phylogenetic analysis of 16S rRNA nucleotide sequences, eight bacterial isolates, includi

Monoclonal antibodies protect against pandrug-resistant Klebsiella pneumoniae.

The 'silent pandemic' caused by antimicrobial resistance requires innovative therapeutic approaches. Human monoclonal antibodies (mAbs), which are among the most transformative and safe drugs in oncology1 and autoimmunity2, are rarely used for infectious diseases and not yet used for antimicrobial resistance3. Here we applied an antigen-agnostic strategy to isolate extremely potent human mAbs against Klebsiella pneumoniae sequence type 147 (ST147), a hypervirulent and pandrug-resistant lineage that is spreading globally. Isolated mAbs target the KL64 capsule and the O-antigen. However, althoug

Electrophoretic deposition of organic/inorganic composite coatings containing ZnO nanoparticles exhibiting antibacterial properties.

To address one of the serious problems associated with permanent implants, namely bacterial infections, novel organic/inorganic coatings containing zinc oxide nanoparticles (nZnO) are proposed. Coatings were obtained by electrophoretic deposition (EPD) on stainless steel 316L. Different deposition conditions namely: deposition times in the range 60-300s and applied voltage in the range 5-30V as well as developing a layered coating approach were studied. Antibacterial tests against gram-positive Staphylococcus aureus and gram-negative Salmonella enteric bacteria confirmed the activity of nZnO t

Incipient caries lesions on cementum by mono- and co-culture oral bacteria.

OBJECTIVES: There is increasing prevalence of root caries. We hypothesized different biofilms will cause varying demineralization in cementum. This study investigated the extent of demineralization of cementum by oral biofilm formed from three major cariogenic microorganisms: Streptococcus mutans, Lactobacillus acidophilus, and Actinomyces israelii. Sound cementum tooth blocks were incubated with mono-, bi-, and tri-species combinations of the bacteria under investigation. MATERIALS AND METHODS: The matrix (amide I) and phosphate content of the lesions was analyzed by Fourier-transform infrar

Nitrosative stress: metabolic pathway involving the flavohemoglobin.

Nitric oxide (NO) biology has focused on the tightly regulated enzymatic mechanism that transforms L-arginine into a family of molecules, which serve both signaling and defense functions. However, very little is known of the pathways that metabolize these molecules or turn off the signals. The paradigm is well exemplified in bacteria where S-nitrosothiols (SNO)-compounds identified with antimicrobial activities of NO synthase-elicit responses that mediate bacterial resistance by unknown mechanisms. Here we show that Escherichia coli possess both constitutive and inducible elements for SNO meta

Monocytes and histiocytes in cell cultures of cerebrospinal fluid. Morphology of cultured CSF cells.

A method of CSF cell culturing, based on observations of cultured cells isolated from 700 CSF specimens obtained for routine diagnostic procedures by lumbar puncture from patients who had no proven or suspected neoplastic disease, is described which enables the demonstration of proliferating mononuclear elements even when they are present in specimens with low cell count. Spread on surfaces of plastic and glass material, monocytes and histiocytes in CSF cell cultures can appear as polygonal or crescent shaped epitheloid cells, may assume spindle shapes, or transform into multinucleated giant c

Carbon-modified sugars: from natural products to glycomimetics.

Bacteria take prototypical aldohexopyranoses and remodel them into more elaborate sugars that provide them advantages in competitive environments. The attachment of alkyl or acyl units directly onto carbons of the hexose backbone is one means of that transformation of "normal" sugars to the novel ones. Similarly, the incorporation of alkyl, vinyl, and aryl groups directly onto hexose carbons is a structural feature used in the design of glycomimetic compounds. These can collectively be classified as carbon-modified sugars. A survey of synthetic methods used to prepare carbon-modified sugars is

Rapid Thiamethoxam Biodegradation by Paenarthrobacter nicotinovorans GY-1: Mechanistic Insights and Nitro-Reductive Pathway Elucidation.

The neonicotinoid insecticide thiamethoxam (THX) poses ecological risks and requires efficient bioremediation strategies. We isolated a highly efficient THX-degrading strain, Paenarthrobacter nicotinovorans GY-1, from contaminated agricultural soil. Under response surface-optimized conditions, GY-1 achieved an unprecedented THX degradation rate of 2.08 mg·L-1·h-1, the highest reported for a microorganism to our knowledge. Transcriptomics, enzyme assays, and carbon-source profiling showed that strain GY-1 suppresses glycolysis and reprograms central metabolism when THX is used as the sole nitro

Harnessing Microbial Bioremediation and Wastewater Valorization for Production of (Methyl)muconates via Engineered Metabolic Pathways in Aromatic Pollutant Mixtures.

Industrial effluents, particularly (semi)coking wastewaters, pose significant environmental challenges due to their complex aromatic pollutants. This study introduces an innovative approach by engineering Pseudomonas putida BGR4, a robust biodegradative chassis, for the treatment and transformation of simulated aromatic pollutant mixtures of semicoking wastewater into high-value compounds such as (methyl)muconates. We integrated degradation pathways for benzene, toluene, (methyl)phenols, and naphthalene (∼30 kb) into the bacterial chromosome, and through adaptive laboratory evolution (ALE), de
