Retrograde transport of dopamine beta-hydroxylase antibodies in sympathetic neurons: effects of drugs modifying noradrenergic transmission.

Antibodies to dopamine beta-hydroxylase (anti-D beta H) were taken up by noradrenergic nerve terminals in the iris following attachment to D beta H, and were transported back to, and accumulated in, the superior cervical ganglion (SCG). Concurrent, or prior destruction of noradrenergic terminals with 6-hydroxydopamine, injected intraocularly, blocked the retrograde transport of anti-D beta H. However, recovery was rapid, reaching 50% of control values within 1 day. Such transport was characterized by a shorter time period before accumulation could be detected in the SCG and by a slower rate of

Empty clathrin cages in neurons.

Clathrin has established roles in endocytosis and short-range vesicle trafficking, where clathrin cages envelop membrane-pits on the plasma membrane or vesicles exchanging between Golgi/endosomes, and this canonical role of clathrin is thought to be conserved in all cell types including neurons. However, the polarized architecture of neurons creates unique demands on clathrin recruitment and function, requiring clathrin to operate in presynapses that are far from sites of protein synthesis in the soma and act within the limited membrane area of periactive-zones in a nonconstitutive manner-resp

Control of autophagosome axonal retrograde flux by presynaptic activity unveiled using botulinum neurotoxin type a.

Botulinum neurotoxin type A (BoNT/A) is a highly potent neurotoxin that elicits flaccid paralysis by enzymatic cleavage of the exocytic machinery component SNAP25 in motor nerve terminals. However, recent evidence suggests that the neurotoxic activity of BoNT/A is not restricted to the periphery, but also reaches the CNS after retrograde axonal transport. Because BoNT/A is internalized in recycling synaptic vesicles, it is unclear which compartment facilitates this transport. Using live-cell confocal and single-molecule imaging of rat hippocampal neurons cultured in microfluidic devices, we sh

Spectrum and functions of ion channels and transporters in osteoclasts.

Osteoclasts are essential for bone resorption and interact with osteoblasts during bone remodeling. Ion channels and transporters located in the ruffled border or intracellular vesicles coordinate the transport of various ions and substrates, which is fundamental to the primary functions of osteoclasts. Numerous channels and transporters are implicated in bone metabolic disorders and genetic diseases. Among these, the voltage-gated chloride channel 7 (ClC-7) and vacuolar proton ATPases (V-ATPase) represent the most well-characterized examples in osteoclasts. Using the classification system of

The ESCRT-III machinery participates in the production of extracellular vesicles and protein export during Plasmodium falciparum infection.

Infection with Plasmodium falciparum enhances extracellular vesicle (EV) production in parasitized red blood cells (pRBCs), an important mechanism for parasite-to-parasite communication during the asexual intraerythrocytic life cycle. The endosomal sorting complex required for transport (ESCRT), and in particular the ESCRT-III sub-complex, participates in the formation of EVs in higher eukaryotes. However, RBCs have lost the majority of their organelles through the maturation process, including an important reduction in their vesicular network. Therefore, the mechanism of EV production in P. f

Calmodulin-dependent regulation of the catalytic function of the human serotonin transporter in placental choriocarcinoma cells.

We investigated the involvement of calmodulin-dependent cellular processes in the regulation of the human serotonin transporter in placental choriocarcinoma cells. Treatment of JAR and BeWo cells with the selective calmodulin antagonist 1,3-dihydro-1-[1-((4-methyl-4H, 6H-pyrrolo[1,2-a][4,1]-benzoxapin-4-yl)-methyl)-4-piperindinyl+ ++]- 2H-benzimidazol-2-one (CGS93-43B (CGS)) for 1 h decreased the imipramine-sensitive serotonin transport activity markedly. The inhibitory effect was specific and was reproducible with other calmodulin antagonists. The basal serotonin transport activity as well as

Extracellular vesicle therapy for retinal diseases.

Extracellular vesicles (EV), which include exosomes and microvesicles, are secreted from virtually every cell. EV contain mRNA, miRNA, lipids and proteins and can deliver this expansive cargo into nearby cells as well as over long distances via the blood stream. Great interest has been given to them for their role in cell to cell communication, disease progression, or as biomarkers, and more recent studies have interrogated their potential as a therapeutic that may replace paracrine-acting cell therapies. The retina is a conveniently accessible component of the central nervous system and the p

Clodronate: The Influence on ATP Purinergic Signaling.

ATP is involved in numerous physiological functions, such as neurotransmission, modulation, and secretion, as well as in cell proliferation, … pheral nociceptors, and the central nervous system. ATP produced inside cells is accumulated within transport vesicles, where it penetrates via a VNUT channel and is then released extracellularly, playing an active role in acute and chronic inflammatory processes, neurotransmission of pain, and liver disease regulation. Clodronate influences these processes due to i …

Protection effect of GDNF and neurturin on photosensitized crayfish neurons and glial cells.

Neurons and glial cells can protect each other from stress and following death by mutual exchange with neurotrophins. In order to examine involvement of different neurotrophic factors in neuroglial interactions in a photosensitized crayfish stretch receptor, a simple model object consisting of only two sensory neurons enveloped by glial cells, we studied the influence of glial cell line-derived neurotrophic factor (GDNF), neurturin, and ciliary neurotrophic factor (CNTF) on its photodynamic injury. Photodynamic treatment, which causes strong oxidative stress, induced firing abolition and necro

Iron transport pathways in the human malaria parasite Plasmodium falciparum revealed by RNA-sequencing.

Host iron deficiency is protective against severe malaria as the human malaria parasite Plasmodium falciparum depends on bioavailable iron from its host to proliferate. The essential pathways of iron acquisition, storage, export, and detoxification in the parasite differ from those in humans, as orthologs of the mammalian transferrin receptor, ferritin, or ferroportin, and a functional heme oxygenase are absent in P. falciparum. Thus, the proteins involved in these processes may be excellent targets for therapeutic development, yet remain largely unknown. Here, we show that parasites cultured

The H(+)-motive and Na(+)-motive respiratory chains in Bacillus FTU subcellular vesicles.

Respiration-dependent pumping of Na+ and H+ into the inside-out subcellular vesicles of alkalotolerant and halotolerant Bacillus FTU grown at alkaline pH was studied. The vesicles were shown to be competent in Na+ and H+ transport coupled to ascorbate oxidation via N,N,N',N'-tetramethyl-p-phenylenediamine or diaminodurene. The uphill Na+ uptake is strongly stimulated by either protonophores or valinomycin, whereas H+ uptake is stimulated by valinomycin and completely inhibited by protonophores. Engineering Principles of Extracellular Vesicles for Central Nervous System Therapies.

Extracellular Vesicles (EVs) are active mediators of intercellular signaling in the Central Nervous System (CNS), operating alongside synaptic transmission as an extra form of wireless communication between neurons and other brain cell types. Their biocompatibility positions them as promising carriers for neurobiological therapeutics; however, translation hinges on clarifying when and how EVs traverse the Blood-Brain Barrier (BBB) and how their design can be aligned to pathophysiology. We synthesize evidence for BBB transport from in vitro models and in vivo visualization and functional studie

Scaffold protein JLP is critical for CD40 signaling in B lymphocytes.

CD40 expression on the surface of B lymphocytes is essential for their biological function and fate decision. The engagement of CD40 with its cognate ligand, CD154, leads to a sequence of cellular events in B lymphocytes, including CD40 cytoplasmic translocation, a temporal and spatial organization of effector molecules, and a cascade of CD40-induced signal transduction. The JLP scaffold protein was expressed in murine B lymphocytes. Using B lymphocytes from jlp-deficient mice, we observed that JLP deficiency resulted in defective CD40 internalization upon CD154/CD40 engagement. Examination of

Chlamydia pneumoniae CPj0783 interaction with Huntingtin-protein14.

Chlamydia pneumoniae is a Gram-negative, obligate intracellular pathogen that causes community-acquired respiratory infections. After C. pneumoniae invades host cells, it disturbs the vesicle transport system to escape host lysosomal or autophagosomal degradation. By using a yeast mis-sorting assay, we found 10 C. pneumoniae candidate genes involved in aberrant vesicular trafficking in host cells. One of the candidate genes, CPj0783, was recognized by antibodies from C. pneumoniae-infected patients. The expression of CPj0783 was detected at mid to late-cycle time points and increased during th

Identification and functional analysis of the vision-specific BBS3 (ARL6) long isoform.

Bardet-Biedl Syndrome (BBS) is a heterogeneous syndromic form of retinal degeneration. We have identified a novel transcript of a known BBS gene, BBS3 (ARL6), which includes an additional exon. This transcript, BBS3L, is evolutionally conserved and is expressed predominantly in the eye, suggesting a specialized role in vision. Using antisense oligonucleotide knockdown in zebrafish, we previously demonstrated that bbs3 knockdown results in the cardinal features of BBS in zebrafish, including defects to the ciliated Kupffer's Vesicle and delayed retrograde melanosome transport. Unlike bbs3, knoc

Expression and immunohistochemical localization of aquaporin-1 in male reproductive organs of the mouse.

Aquaporin-1 (AQP-1), a six-transmembrane domain protein, is found to be responsible for water transport. The purpose of this study was to investigate the expression of AQP-1 mRNA and protein in the testis, epididymis, vas deferens, ventral prostate, and seminal vesicle from mature mice by using reverse transcription polymerase chain reaction (RT-PCR) and Western blotting, and the cellular localization of AQP-1 by immunohistochemistry. RT-PCR revealed that AQP-1 mRNA was expressed in all organs we examined. Western blotting displayed a 29-kDa band and a 35- to 45-kDa band corresponding to non-g

Lactate metabolism and lactylation in uveal melanoma: from metabolic adaptation to immune regulation.

Uveal melanoma (UM) is the most common primary intraocular malignancy in adults. The liver is its most common site of metastasis, and in some patients, distant metastases may become clinically apparent years after treatment of the primary tumor. Metastatic UM generally shows limited responsiveness to immune checkpoint blockade. Although genetic alterations in GNAQ, GNA11, BAP1, SF3B1, and EIF1AX are important for the molecular classification and prognostic stratification of UM, these factors do not fully explain late metastatic recurrence, persistence of low-proliferative tumor cell states, im

Merkel cells, corpuscular nerve endings and free nerve endings in the mouse palatine mucosa express three subtypes of vesicular glutamate transporters.

The hard palate of rodents is a mucous membrane covered by a keratinized epithelium that typically contains Merkel cell (MC)-neurite complexes. MCs have engendered considerable research activity because of their involvement in mechanoreception and possibly also Merkel cell carcinomas. MCs derive from the neural crest, differentiate under control of peripheral nerve factors, are enriched in large dense core vesicles, and secrete neuropeptides and other neuroactive molecules. Upon stimulation, MC-neurite complexes produce slowly adapting type I responses. Here we emphasize that the murine hard p

A novel RAB33B mutation in Smith-McCort dysplasia.

Smith-McCort dysplasia (SMC) is a rare autosomal recessive spondylo-epi-metaphyseal dysplasia with skeletal features identical to those of Dyggve-Melchior-Clausen syndrome (DMC) but with normal intelligence and no microcephaly. Although both syndromes were shown to result from mutations in the DYM gene, which encodes the Golgi protein DYMECLIN, a few SMC patients remained negative in DYM mutation screening. Recently, autozygosity mapping and exome sequencing in a large SMC family have allowed the identification of a missense mutation in RAB33B, another Golgi protein involved in retrograde tran

Different miRNA Profiles in Plasma Derived Small and Large Extracellular Vesicles from Patients with Neurodegenerative Diseases.

Identifying biomarkers is essential for early diagnosis of neurodegenerative diseases (NDs). Large (LEVs) and small extracellular vesicles (SEVs) are extracellular vesicles (EVs) of different sizes and biological functions transported in blood and they may be valid biomarkers for NDs. The aim of our study was to investigate common and different miRNA signatures in plasma derived LEVs and SEVs of Alzheimer's disease (AD), Parkinson's disease (PD), Amyotrophic Lateral Sclerosis (ALS) and Fronto-Temporal Dementia (FTD) patients. LEVs and SEVs were isolated from plasma of patients and healthy volu
