Virulence traits and plasmid-mediated quinolone resistance among Aggregatibacter actinomycetemcomitans from Iraq: Low rate of highly virulent JP2 genotype.

Aggregatibacter actinomycetemcomitans (A. actinomycetemcomitans) is an important causative agent of periodontitis acting by employment of a series of virulence factors. Our aim was to evaluate the virulence traits and plasmid-mediated quinolone resistance of A. actinomycetemcomitans isolates in Iraq. A total of 1580 samples were collected from dental caries (n = 1190) and periodontitis (n = 390) among which 200 samples were positive. The antibiotic susceptibility pattern and biofilm formation were performed. The antibiotic resistance and virulence determinants were screened using polymerase ch…

Divergence between the Highly Virulent Zoonotic Pathogen Helicobacter heilmannii and Its Closest Relative, the Low-Virulence "Helicobacter ailurogastricus" sp. nov.

Helicobacter heilmannii naturally colonizes the stomachs of dogs and cats and has been associated with gastric disorders in humans. Nine feline Helicobacter strains, classified as H. heilmannii based on ureAB and 16S rRNA gene sequences, were divided into a highly virulent and a low-virulence group. The genomes of these strains were sequenced to investigate their phylogenetic relationships, to define their gene content and diversity, and to determine if the differences in pathogenicity were associated with the presence or absence of potential virulence genes. The capacities of these helicobact…

Comparison of invasion of fibroblasts and macrophages by high- and low-virulence Leptospira strains: colonization of the host-cell nucleus and induction of necrosis by the virulent strain.

The infection cycle of low- and high-virulence strains of Leptospira interrogans was compared in fibroblasts and macrophages. L. interrogans serovar Lai strain Lai was used as a representative high-virulence strain, while L. interrogans serovars Pomona strain Luo was used as a low-virulence strain. L. biflexa serovar Patoc strain Patoc I, a nonparasitic strain of Leptospira, was used as a control. Both the high- and low-virulence strains could adhere to fibroblasts and macrophages using one or both ends of the spirochete, which was followed by phagocytosis of both strains. Both strains adhered…

Non-toxigenic derivatives of the Ames strain of Bacillus anthracis are fully virulent for mice: role of plasmid pX02 and chromosome in strain-dependent virulence.

The toxin-encoding plasmid pX01 and capsule-associated plasmid pX02 are required for full virulence of Bacillus anthracis in some animals. However, the non-toxigenic pX01-cured derivatives of certain anthrax strains are not completely attenuated for mice, and their virulence is strain-dependent. The strain-related differences were partially associated with plasmid pX02 as demonstrated by pX02 transductants of the attenuated vaccine strain UM23-1 cured of pX01. To determine the virulence of non-toxigenic variants of virulent strains, we isolated pX01- derivatives of the Vollum 1B strain and the…

Entamoeba histolytica clones presenting transient virulent phenotypes: subsequent fate of forcibly induced virulent clones by hamster's liver abscess model.

BACKGROUND: Amebiasis, which is caused by Entamoeba histolytica, is prevalent worldwide. Clinical presentation varies from asymptomatic infection to life-threatening extraintestinal disease, and the clinical form can change within one infectious episode. It is also known that periodical animal passages are required for maintaining virulence of E. histolytica in vivo model, such as a hamster liver abscess or murine colitis model. However, it remains uncertain whether or for how long virulence induced by environmental stimuli persists during in vitro passage at the clonal level. RESULTS: We gen…

Multiplex qPCR assay for ultra sensitive detection of JCV DNA with simultaneous identification of genotypes that discriminates non-virulent from virulent variants.

BACKGROUND: JC virus (JCV) is the etiologic agent for progressive multifocal leukoencephalopathy (PML), a demyelinating disease occurring in the brain of patients with underlying immune compromised states. All viable JCV genomes contain a conserved region in the T protein coding nucleotide sequence that when detected by PCR in CSF is a confirmatory diagnostic marker for PML along with clinical and neuroradiological evidence. The non-coding regulatory region (NCRR) is hypervariable, as evidenced by nucleotide sequence of the non-virulent variant, which is predominantly excreted in urine, versus

Infection with a low virulent Mycoplasma hyopneumoniae isolate does not protect piglets against subsequent infection with a highly virulent M. hyopneumoniae isolate.

The study aimed to evaluate the effect of an infection with low virulent isolates of M. hyopneumoniae (LV1 and LV2) on the subsequent infection with a highly virulent isolate (HV). Fifty-five, 3-week-old piglets free of M. hyopneumoniae were randomly allocated to 6 different groups. At 4 weeks of age (D0), groups LV1-HV and LV1 were intratracheally inoculated with LV1, groups LV2-HV and LV2 with LV2, and group HV with sterile culture medium. Four weeks later (D28), the pigs of these different groups were either intratracheally inoculated with the highly virulent isolate (groups LV1-HV, LV2-HV,

Evaluation of DNA polymorphisms amplified by arbitrary primers (RAPD) as genetically associated elements to differentiate virulent and non-virulent Paracoccidioides brasiliensis isolates.

Randomly amplified polymorphic DNA (RAPD) analysis of 35 Paracoccidioides brasiliensis isolates was carried out to evaluate the correlation of RAPD profiles with the virulence degree or the type of the clinical manifestations of human paracoccidioidomycosis. The dendrogram presented two main groups sharing 64% genetic similarity. Group A included two isolates from patients with chronic paracoccidioidomycosis; group B comprised the following isolates showing 65% similarity: two non-virulent, six attenuated, five virulent, eight from patients with chronic paracoccidioidomycosis and two from pati…

PtRTP1, an Effector of Puccinia triticina, Contributes to Virulence in Wheat.

Effector proteins are key virulence factors of obligate parasitic fungi during infection. Wheat leaf rust by Puccinia triticina is a widespread destructive disease drastically limiting wheat production, while the biological role of its effector is still elusive. PtRTP1 is a homolog of Uf-RTP1p, one of the earliest effector proteins verified to localize at the host-pathogen interface. Clarifying PtRTP1 function helps uncover rust pathogenic mechanisms; hence, we cloned PtRTP1 from P. triticina. The gene has a 1453 bp DNA sequence and a 777 bp CDS, encoding a protein with a 23 aa signal peptide,…

Duration of immunity following infection with moderately virulent ASFV.

African swine fever virus (ASFV) poses a significant threat to pork production and wild pig populations worldwide. The study assessed the long-term fate and immunity of animals recovering from a moderately virulent ASFV infection, following the principles of a duration of immunity study for live vaccines. Pigs inoculated with the moderately virulent ASFV strain Estonia14 largely developed mild clinical signs and only transient viremia. Six months after the initial inoculation and once fully recovered, all the animals were challenged with highly virulent ASFV Armenia08. Only one of the previous

An Overview of Virulence, Resistance, and Clinical Significance of Klebsiella Infections.

Gram-negative, encapsulated, non-motile bacilli called Klebsiella species are commonly found in the environment and are a natural component … minal infections, and meningitis. The pathogenicity of Klebsiella species is attributed to multiple virulence factors, including a prominent polysaccharide capsule, adhesins, siderophores, lipopolysaccharide, and the ability to form biofilms. In recent years, the clinical importance of these organisms has grown as a result of the global emergence of multidr …

Virulence and Antimicrobial Resistance in Campylobacter jejuni and Campylobacter coli.

Campylobacter jejuni and Campylobacter coli account for most recognized cases of human campylobacteriosis. For C. coli, gene-carriage surveys often stand in for direct tests of function; this distinction is maintained throughout the review. Virulence is discussed as a sequence of events rather than as a catalogue of factors. Motility and adhesion have the most reproducible support during colonization and epithelial contact, whereas biofilm formation, quorum sensing, and T6SS-associated phenotypes vary substantially across strains and models. We then consider host-cell injury, persistence, tran…

Genomic diversity and virulence of Pasteurella multocida in Norwegian calves.

The bovine respiratory disease (BRD) complex is a major health and welfare challenge in the cattle industry worldwide, with substantial economic and antimicrobial resistance implications. Pasteurella multocida (PM) is a key bacterial agent of BRD; however, knowledge gaps remain regarding its transmission dynamics, genomic diversity and virulence-associated gene repertoire in cattle. In this cross-sectional study, we used population genomics to characterize PM isolates circulating in Norwegian calves and compare them with global bovine PM lineages. By integrating genomics with epidemiological a…

Understanding the role of pyruvate dehydrogenase in Listeria monocytogenes virulence.

Together, these findings demonstrate that PDH is required for the import and metabolism of PTS-dependent carbon sources in the host cytosol and suggest that PDH-dependent redox balance and respiro-fermentative metabolism ultimately contribu …

Influence of the polysaccharide capsule on virulence and fitness of Klebsiella pneumoniae.

INTRODUCTION: The capsular polysaccharide (CPS) of pathogenic bacteria is a critical virulence factor, often evading phagocytosis by host immune cells, while also interfering with the contact of the pathogen with host cells and contributing to biofilm formation. Klebsiella pneumoniae, a Gram-negative human pathogen associated with high antimicrobial resistances, produces 77 CPS serotypes. The CPS masks proteinaceous factors but also protects K. pneumoniae from uptake by host phagocytic cells and activation of the complement system. In addition to nosocomial, urinary tract and bloodstream infec…
