Novel functions of the primary cilium in bone disease and cancer.

The primary cilium, a sensory organelle that emanates from the cell surface of most mammalian cell types during growth arrest, has attracted the attention of many researchers over the past decade. Recently, a large number of new findings have assigned novel functions and roles to the primary cilium in signal transduction and related diseases, which has greatly augmented the importance of the cilium in human health and development. Here, we review emerging evidence supporting the primary cilium as a sensory organelle in signal transduction in microgravity, electromagnetic field sensing, chemose

Efficacy of manual versus free-weight training to improve maximal strength and performance for microgravity conditions.

We tested a simple and compact device designed for manual resistance training in conditions of microgravity (Self-Powered Rope Trainer Duo (SPoRT Duo)) to increase muscle performance. Twenty-four participants (20.8 ± 2.1 years) were randomly assigned to a manual resistance group (n = 12) and a free-weight group (n = 12). Participants performed eight exercises (three sets; 8-12 efforts) either with free weights or the SPoRT Duo twice a week for 6 weeks. Maximal isometric force of trunk flexion, back extension and chest press increased (P at least 0.01, d at least 0.52) both in the manual resist

Effects of sex and gender on adaptation to space: neurosensory systems.

Sex and gender differences have long been a research topic of interest, yet few studies have explored the specific differences in neurological responses between men and women during and after spaceflight. Knowledge in this field is limited due to the significant disproportion of sexes enrolled in the astronaut corps. Research indicates that general neurological and sensory differences exist between the sexes, such as those in laterality of amygdala activity, sensitivity and discrimination in vision processing, and neuronal cell death (apoptosis) pathways. In spaceflight, sex differences may in

A computational model for cortical endosteal surface remodeling induced by mechanical disuse.

In mechanical disuse conditions associated with immobilization and microgravity in spaceflight, cortical endosteal surface moved outward with periosteal surface moving slightly or unchanged, resulting in reduction of cortical thickness. Reduced thickness of the shaft cortex of long bone can be considered as an independent predictor of fractures. Accordingly, it is important to study the remodeling process at cortical endosteal surface. This paper presents a computer simulation of cortical endosteal remodeling induced by mechanical disuse at the Basic Multicellular Units level with cortical thi

Escherichia coli biofilms formed under low-shear modeled microgravity in a ground-based system.

Bacterial biofilms cause chronic diseases that are difficult to control. Since biofilm formation in space is well documented and planktonic cells become more resistant and virulent under modeled microgravity, it is important to determine the effect of this gravity condition on biofilms. Inclusion of glass microcarrier beads of appropriate dimensions and density with medium and inoculum, in vessels specially designed to permit ground-based investigations into aspects of low-shear modeled microgravity (LSMMG), facilitated these studies. Mathematical modeling of microcarrier behavior based on exp

[Correlation of the cosmonauts' physiological reactions to +Gx loads during deorbit with the hemodynamic shifts in the period of short-term microgravity].

Physiological reactions of four cosmonauts to +Gx during the descent after short-term (less than 30 d) space flights were correlated with the hemodynamic shifts developed previously during the LBNP test and occlusive plethysmography in microgravity. The cosmonauts who had exhibited a sharp decrease in cerebral circulation during the in-flight LBNP tests showed signs of deteriorated cerebral blood flow (vertigo, weakness, hyperhydration) at the attempt to assume the vertical posture and to climb out of the landing module. Also, the cosmonauts found to have the arterial and venous tone significa

[Development and application of self-powered short arm human centrifuge].

OBJECTIVE: To present the development and application of a self-powered short arm human centrifuge. METHOD: Self-powered short arm human centrifuge consisted of human power, adjustable resistance trig, short arm and supporting construction parts. It was driven by human power. The short arm turned around the supporting part so artificial gravity is produced. The physical load can be adjusted by the adjustable resistance part. G-level depends on the rotative velocity of the central shaft. Nine healthy male volunteers received self-powered short arm human centrifuge training for 5 min per day wi

Effects of altered gravity on the swimming behaviour of fish.

Humans taking part in parabolic aircraft flights (PAFs) may suffer from space motion sickness-phenomena (SMS, a kinetosis). It has been argued that SMS during PAFs might not be based on microgravity alone but rather on changing accelerations from 0 g to 2 g. We test here the hypothesis that PAF-induced kinetosis is based on asymmetric statoliths (i.e., differently weighed statoliths on the right and the left side of the head), with asymmetric inputs to the brain being disclosed at microgravity. Since fish frequently reveal kinetotic behaviour during PAFs (especially so-called spinning movement

[Microgravity and autonomic nervous system].

How microgravity influences autonomic function is still under investigation. Microgravity induces neuro-vestibular alterations and body fluid shift, and these two changes cause "space motion sickness(SMS)" and cardiovascular deconditioning. "Space motion sickness" is a autonomic syndrome that exhibits nausea, vomiting, headache, anorexia, pallor etc., whose incidence in Space Shuttle mission reaches 67.1%. There are several hypotheses for SMS mechanism: 1) sensory conflict, 2) fluid shift, 3) otolith asymmetry, 4) space orientation readaptation, 5) otolith tilt-translation reinterpretation, an

Histology and histochemistry of intervertebral discs of rats participated in spaceflight.

The qualitative and quantitative histological and histochemical changes in the structure and macromolecular composition of lumbar intervertebral discs of rat during a 12.5-day space flight (Cosmos 1887 biosatellite) were determined using light and polarization microscopy. Semiquantitative histochemical, topo-optical reactions were measured and evaluated by retardation values of birefringence. (a) Lateral expansion and accumulation of the notochordal cells in the nucleus pulposus was observed in contrast with the vivarium control, where the chondroid cells dominated. Quantified EEG in different G situations.

The electrical activity of the brain (EEG) has been recorded during parabolic flights in trained astronauts and non trained volunteers as well. The Fast Fourier analysis of the EEG activity evidenced more asymmetry between the two brain hemispheres in the subjects who suffered from motion sickness than in the others. However, such a FFT classification does not lead to a discrimination between deterministic and stochastic events. Therefore, a first attempt was made to calculate the dimensionality of "chaotic attractors" in the EEG patterns as a function of the different g-epochs of one parabola

Subgravity states: key to understanding the role of terrestrial gravity in human behaviour.

The early orbital flights, although undertaken with considerable confidence, involved some uncertainty because of the impossibility of simulating under terrestrial conditions all of the conditions encountered in space. However, space-flight achievements by both the American astronauts and Soviet cosmonauts have firmly established that man, if appropriately selected, trained, and protected by suitable life-support systems, can perform efficiently for long periods of time in the hostile environment of space. We know that the side effects of vestibular origin pose important problems in space expl

Gastrodia elata Blume (Tianma) extract mitigates simulated microgravity-induced oxidative stress and enhance learning and memory in Caenorhabditis elegans.

Microgravity during long-duration spaceflights promotes oxidative damage, impairs mitochondrial efficiency, and induces neurodegeneration, highlighting the need for safe countermeasures. Gastrodia elata (Tianma) is a traditional medicinal herb with known antioxidant and neuroprotective properties that has not yet been examined under microgravity-analog conditions. The main aim of this study was to evaluate whether Tianma mitigates oxidative, mitochondrial, and neurobehavioral deficits induced by simulated microgravity (SM) in Caenorhabditis elegans (C. elegans). Simulated microgravity models sex differences in knee osteoarthritis through a CD36-regulated mechano-metabolic circuit.

BACKGROUND: Sex differences in knee osteoarthritis (KOA) are well documented, but the molecular drivers within meniscal tissue remain unclear. We studied whether simulated microgravity (SMG) models sex‑dependent meniscal degeneration and whether the lipid scavenger receptor CD36 mediates these effects. METHODS: Human menisci from total knee arthroplasty (TKA) donors were analyzed (histology n = 5 females/5 males; flow cytometry and RT‑qPCR n = 7 females/7 males). In mice, we assessed spontaneous anterior‑horn mineralization by micro‑CT at 4, 12, and 24 weeks in wild‑type (WT) and CD36‑deficien

Microgravity-driven Rab27B activation amplifies mesenchymal stem cell-derived extracellular vesicle production and functions.

BACKGROUND: Mesenchymal stem cell-derived extracellular vesicles (MSC-EVs) are emerging as promising treatments for immunomodulation and tissue regeneration. However, the scalable production of functionally enhanced EVs remains a critical challenge. This study introduces microgravity culture using a three-dimensional (3D) rotating cell culture system as a novel strategy to optimize MSC-EVs yield and bioactivity. METHODS: We first investigated the effects of microgravity on the proliferation and stemness of human umbilical cord-derived MSCs (UCMSCs). The yield of microgravity-derived EVs (µg-E

Simulated microgravity-induced dysregulation of cerebrospinal fluid immune homeostasis by disrupting the blood-cerebrospinal fluid barrier.

BACKGROUND: The blood-cerebrospinal fluid barrier (BCSFB) comprises the choroid plexus epithelia. It is important for brain development, maintenance, function, and especially for maintaining immune homeostasis in the cerebrospinal fluid (CSF). Although previous studies have shown that the peripheral immune function of the body is impaired upon exposure to microgravity, no studies have reported changes in immune cells and cytokines in the CSF that reflect neuroimmune status. The purpose of this study is to investigate the alterations in cerebrospinal fluid (CSF) immune homeostasis induced by mi

Inflammasome-Inhibiting Nanoligomers Are Neuroprotective against Space-Induced Pathology in Healthy and Diseased Three-Dimensional Human Motor and Prefrontal Cortex Brain Organoids.

The microgravity and space environment has been linked to deficits in neuromuscular and cognitive capabilities, hypothesized to occur due to accelerated aging and neurodegeneration in space. While the specific mechanisms are still being investigated, spaceflight-associated neuropathology is an important health risk to astronauts and space tourists and is being actively investigated for the development of appropriate countermeasures. However, such space-induced neuropathology offers an opportunity for accelerated screening of therapeutic targets and lead molecules for treating neurodegenerative

Sensory conflict compared in microgravity, artificial gravity, motion sickness, and vestibular disorders.

UNLABELLED: Perceptual disturbances and motion sickness are often attributed to sensory conflict. We investigated several conditions: head movements in microgravity, periodic motions in 1-g, and locomotion with vestibular disorders. In every case, linear vectors such as linear and gravitational acceleration are crucial factors, as previously found for head movements in artificial gravity, and thus the importance of measuring linear vectors emerges as a common theme. By modeling the sensory conflict between the vestibular and somatosensory systems, we computed a measure of linear conflict known

The effect of microgravity on the autonomic nervous system of rhesus monkeys.

Gastric myoelectric activity and respiratory sinus arrhythmia of two monkeys flown on Bion 11 were investigated. During flight, vagal effects on cardiac and gastric function of the two animals varied differently: in one animal they decreased at an early flight stage and returned to the control level by the end of flight, whereas in the second monkey the trend was the opposite. The decrease was accompanied by space motion sickness symptoms.
