Incidental detection of microfilariae in aspirates from Ewing's sarcoma of bone.

Microfilariae have been reported as coincidental findings with various neoplasms. We present a case in which microfilariae were encountered in fine-needle aspiration (FNA) smears from a 10-yr-old boy who presented with a lytic lesion in the diaphysis of the right tibial bone. Stained aspirates from the swelling were composed of a dispersed population of small round cells with scanty to vacuolated cytoplasm. Two sheathed microfilariae with several nuclei and cephalic and caudal clearing were identified. A diagnosis of Ewing's sarcoma with presence of microfilariae of Wuchereria bancrofti was ma…

Quinone analogue irrecoverably paralyses the filarial parasites in vitro.

2,3-Dimethoxy-5-methyl-1,4-benzoquinone (Q0), an analogue of ubiquinone, irreversibly paralyses the adult and microfilariae of the cattle filarial parasite Setaria digitata. The same concentration of Q0 that paralyses the microfilariae of S. digitata also paralyses the microfilariae of the human filarial parasite Wuchereria bancrofti within the same duration. Thus the experiments done in the model S. digitata system can well be extended to the human filarial system. A drug at the level of the quinone-centered energy generating system, perhaps an analogue of quinone like Q0, can inactivate the …

Chromosome aberrations in leukocytes of filaria-infected indigenous inhabitants in the Philippines.

In the course of genetic population surveys of a tribal group of the Philippine Negritos living on Mindanao island, microfilariae (Wuchereria bancrofti) were incidentally found on the microscopic slides for a cytogenetic investigation. Structural chromosome aberrations in the cultured leukocytes from 19 filaria-infected and 22 uninfected subjects were studied. The frequencies of chromosome type aberrations such as dicentrics and chromosome breaks in the infected subjects were significantly higher than those in the uninfected subjects (P less than 0.05). The frequencies of chromatid type aberra…

Antibiotics for the treatment of onchocerciasis and other filarial infections.

More effective drugs are needed for the treatment of human filarial diseases and the elimination of these infections as a public health problem. The drugs must either kill or sterilize adult worms. The relevant filariae, Onchocerca volvulus, Wuchereria bancofti and Brugia species, harbor rickettsial endoboacteria of the genus Wolbachia as symbionts. Animal experiments have shown that the elimination of these endobacteria causes inhibition of embryogenesis, and with Onchocerca ochengi a macrofilaricidal effect. Trials with human onchocerciasis patients using doxycydine demonstrated a long-term …

Recovery of a species of Brugia, probably B. ceylonensis, from the conjunctiva of a patient in Sri Lanka.

A species of Brugia, probably B. ceylonensis, was recovered from the conjunctiva of a patient in Sri Lanka for the first time. This infection represents only the second record of Brugia in the human conjunctiva, and is clearly zoonotic, acquired from a dog. Brugia ceylonensis has a distinct head bulb like that of Wuchereria bancrofti and B. malayi. However, the parasite recovered was not W. bancrofti, as specific IFAT and DNA probes gave negative results, and B. malayi is believed to have been eradicated from Sri Lanka several years ago. The presence of a distinct head bulb excludes the possib…

Lymphatic filariasis in a hyperendemic region: a ten-year, follow-up panel survey.

The present study is a long-term panel survey of a population living in a previously identified Wuchereria bancrofti-endemic area of Benin. Unexpectedly, a marked decrease in the prevalence of microfilaremia (from 9.4% to 0.48%; P < 0.001) occurred over a 10-year period in the absence of chemotherapy or vector control measures. The percentage of patients with chronic pathology remained stable during the study period. The decrease in the prevalence of parasitemia could not be explained by environmental or sociologic changes in the region, or by differences between the two study populations. The…

On the infectivity of early third-stage Brugia larvae isolated from the abdomen of Aedes aegypti.

Third-stage larvae (L3) of Wuchereria bancrofti, Brugia malayi and B. pahangi are known to be recovered not only from the head and thorax but also from the abdomen of dissected vector mosquitoes. For epidemiological reasons, was of interest to determine whether L3 larvae from the abdomen of the vector would be infectious for the final host. Early abdominal L3 larvae of B. pahangi isolated as early as on day 7 post-infection (p.i.) from Aedes aegypti were injected s.c. into five male Meriones unguiculatus. Four of the five jirds were microfilaria-positive after 67 days.

Biting periodicity index of Culex quinquefasciatus & its relationship with microfilaria periodicity in Pondicherry.

The biting periodicity index of the bancroftian filariasis vector, C. quinquefasciatus, was calculated on the basis of a large longitudinal data base and compared with the periodicity index of microfilariae of Wuchereria bancrofti. The biting periodicity index of the vector population was 29.78 and found to be close to the periodicity index of the microfilariae. The peak biting activity of the vector and peak appearance of microfilariae in the peripheral blood occurred at about 01.00 h, which accounts for the optimum infection of the vector population. The early night biting density of the vec…

Chemotherapy with spaced doses of diethylcarbamazine preceded by levamisole on Bancroftian filariasis.

The administration of levamisole (2.5 mg/kg) in a single oral dose, the day before beginning weekly spaced dose of diethylcarbamazine (DEC), resulted in low blood levels of microfilariae of Wuchereria bancrofti six weeks after commencing treatment. This rapid decrease in microfilaraemia associated with the addition of levamisole overcomes a major disadvantage of using DEC alone-namely the necessity to commence spaced dose treatment several months before the transmission season. A further advantage of combined levamisole-DEC therapy is that levamisole is effective in ascariasis and hookworm-con…

Wb E34 monoclonal antibody: further characterization and diagnostic use in bancroftian filariasis.

The Wb E34 monoclonal antibody raised against Wuchereria bancrofti microfilarial excretory secretory (mf ES) antigen was reported to be useful in detecting the filarial antigen in W. bancrofti and Brugia malayi infected sera. Further studies in this laboratory showed that this monoclonal antibody reacts with a stage-specific antigen of W. bancrofti filarial parasite. Wb E34 identified three antigenic components with molecular weights, 55, 57.5, and 63 kilodaltons (Kd) in western blot analysis. The target antigen of Wb E34 was found to be located in the cytoplasm of microfilariae by indirect im…

Mosquito vectors of lymphatic and zoonotic filariasis in Malaysia: a systematic review and proportionate meta-analysis of infection rates.

Lymphatic and zoonotic filariasis in Malaysia are transmitted by multiple mosquito species, but the infection rates in these vectors have not been quantitatively reviewed. This systematic review and meta-analysis of proportions compiled species-level infection data for Malaysian mosquitoes infected with Brugia malayi, Brugia pahangi, Wuchereria bancrofti and Dirofilaria immitis. Searches of PubMed, Scopus and Web of Science identified 297 records; 12 studies met the predefined inclusion criteria, yielding 42 mosquito-parasite records involving Mansonia, Anopheles, Armigeres, Aedes and Culex. I…

Safety and tolerability of moxidectin and ivermectin combination treatments for lymphatic filariasis in Côte d'Ivoire: A randomized controlled superiority study.

BACKGROUND: Moxidectin is a macrocyclic lactone registered for the treatment of human onchocerciasis. The drug has a good safety profile, large volume of distribution and a long elimination half-life. This paper reports tolerability data from the first use of moxidectin in persons with Wuchereria bancrofti infection. METHODS: In this randomized, open-label, masked-observer superiority trial, adults with Wuchereria bancrofti microfilaremia in Côte d'Ivoire were randomized to 1 of 4 treatment arms: ivermectin + albendazole (IA), moxidectin + albendazole (MoxA), ivermectin + diethylcarbamazine (…

Distinct Immune Profiles of Exhausted Effector and Memory CD8+ T Cells in Individuals With Filarial Lymphedema.

CD8+ T cells are crucial for the clearance of viral infections, and current research begins to highlight their importance in parasitic diseases too. In-depth research about characteristics of CD8+ T-cell subsets and exhaustion remains uncertain, especially during filariasis, a chronic helminth infection. Lymphatic filariasis, elicited by Wuchereria bancrofti, remains a serious health problem in endemic areas in Ghana, especially in those suffering from morbidity due to lymphedema (LE). In this observational study, the characteristics and profiles of CD8+ T cells were compared between asymptoma…

[Rational for morbidity management in bancroftian filariasis endemic areas].

Selection of the most appropriate therapy for the patient with bancroftian filariasis requires a knowledge of the diverse clinical characteristics of filarial disease and their pathogenesis. As a result of new diagnostic tests and clinical advances, our understanding of bancroftian filariasis has changed rapidly, as have our ideas about treatment. In the past, it was believed that elephantiasis was caused by an immunologic reaction of the host to the filarial parasite. From this perspective, elephantiasis was seen as the endpoint of an unalterable relationship between the host and the parasite

The Wellcome Trust Lecture. Infection and disease in lymphatic filariasis: an immunological perspective.

The basic tenet of the immunological perspective of filarial disease is that differential immune responsiveness among individuals exposed to infection results in the different clinical manifestations that develop. The mechanisms involved in this differential responsiveness appear to reflect different T-cell cytokine response patterns. Asymptomatic patients with the clinically silent presentation of 'asymptomatic microfilaraemia', who have been previously described as being 'immunosuppressed' with respect to their generating pro-inflammatory (Th1-type) immune responses to parasite antigen, are

Diagnosis of microfilaria in gastric brush cytology. A case report.

BACKGROUND: Filariasis due to Wuchereria bancrofti is endemic to southern Asia. While the laboratory diagnosis has been conventionally made by demonstrating microfilariae in peripheral blood smears, these have also been occasionally diagnosed on aspiration cytology of various organs. CASE: A 54-year-old male presented with a burning sensation in the epigastrium of five months' duration. Endoscopic brush biopsy revealed numerous sheathed microfilariae of W bancrofti. The patient had had no symptoms suggestive of filarial infection in the past. Cytology revealed numerous microfilariae among lym…
