Yeast regulatory protein LEU3: a structure-function analysis.

Eleven mutations resulting in partially deleted or truncated LEU3 protein were generated by linker insertion or other modifications at restriction sites, deletion of restriction fragments, or oligonucleotide-directed mutagenesis. Functional studies of these mutants showed the following: (i) A specific DNA binding region is contained within the 173 N-terminal residues, but other regions of the protein are required for optimal binding. (ii) Activation of LEU2 expression depends on the C-terminal 113 residues of the LEU3 protein. (iii) Deletion of part or all of a central section of LEU3 eliminat

Yeast connection among 100 patients with chronic fatigue.

PURPOSE: Patients with the "yeast connection" are characterized by fatigue and multiple systemic symptoms. The purpose of our study was to compare patients with chronic fatigue who believed they had the yeast connection to patients with chronic fatigue without the yeast connection. PATIENTS AND METHODS: One hundred consecutive patients with a chief complaint of chronic fatigue were evaluated in a specialty clinic setting at the University of Connecticut Health Center. A complete history was obtained from each patient, and a 168-item review of systems and a complete physical examination were p

Genetic effects of chlorinated ethylenes in the yeast Saccharomyces cerevisiae.

The chlorinated ethylenes 1,1-dichloroethylene (vinylidene chloride), trans-1,2-dichloroethylene, trichloroethylene, and tetrachloroethylene (perchloroethylene) were assayed for their ability to induce mitotic gene conversion and point mutation as well as mitotic aneuploidy in diploid strains of the yeast Saccharomyces cerevisiae. From strain D7 late logarithmic-phase cells grown in 20% glucose liquid medium, containing a high level of cytochrome P-450, as well as stationary-phase cells combined with an exogenous metabolic activating system (S9) were used, in order to activate the chlorinated

Inducible DNA-repair systems in yeast: competition for lesions.

DNA lesions may be recognized and repaired by more than one DNA-repair process. If two repair systems with different error frequencies have overlapping lesion specificity and one or both is inducible, the resulting variable competition for the lesions can change the biological consequences of these lesions. This concept was demonstrated by observing mutation in yeast cells (Saccharomyces cerevisiae) exposed to combinations of mutagens under conditions which influenced the induction of error-free recombinational repair or error-prone repair. Total mutation frequency was reduced in a manner prop

Resolution of dicentric chromosomes by Ty-mediated recombination in yeast.

We have integrated a plasmid containing a yeast centromere, CEN5, into the HIS4 region of chromosome III by transformation. Of the three transformant colonies examined, none contained a dicentric chromosome, but all contained a rearranged chromosome III. In one transformant, rearrangement occurred by homologous recombination between two Ty elements; one on the left arm and the other on the right arm of chromosome III. This event produced a ring chromosome (ring chromosome III) of about 60 kb consisting of CEN3 and all other sequences between the two Ty elements. In addition, a linear chromosom

The oral yeast flora of 10-year-old schoolchildren.

The yeast flora of the hard palate mucosa and pooled saliva of 122 schoolchildren (average age 9 years 10 months, 63 males, 59 females) has been studied in detail. Of the population sampled, 71.3% carried oral yeasts. The carriage was approximately 3 times greater in females than in males. The yeast isolates were Candida albicans 71% Saccharomyces spp. 19.7% and Candida tropicalis 8.6%. No isolates of Torulopsis spp. or Rhodotorula spp. were recovered. 71.5% of the Candida albicans isolates were serotype A and 26.3% were serotype B.

Effect of sodium chloride on bakers' yeast growing in gelatin.

In recent years, industrial fermentation researchers have shifted their attention from liquid to solid and semisolid culture conditions. We converted liquid cultures to the semisolid mode by adding high levels of gelatin. Previous studies on liquid cultures have revealed the inhibitory activity of mineral salts, such as NaCl, on the fermentation of sugars by yeasts. We made a kinetic study of the effects of 1 to 5% (wt/vol) NaCl on the alcoholic fermentations of glucose by Saccharomyces cerevisiae in a growth medium containing 16% gelatin. Our results showed that the effect of high salt conten…

Factors Affecting Death of Yeast by Sulfur Dioxide 1.

The interrelated factors that influence the effectiveness of SO2 as a preservative against yeast were studied and the correct quantitative relationships of SO2 molecular species determined. Widely differing pK values for SO2 were found in the literature, compared with experimental data, and one set of values was selected. Undissociated H2SO3 is the only effective form of SO2 against yeast and can be calculated from measurement of free SO2 and pH, and the correct dissociation constants. Duration of contact, pH, concentration of SO2 and yeast, and binding of SO2 all influence the preservative ac

Studies on the delta 5-desaturation in ergosterol biosynthesis in yeast.

Studies were carried out on the delta 5-desaturation reaction in ergosterol biosynthesis with a particulate fraction of cell-free extract of yeast. A reduced pyridine nucleotide coenzyme and molecular oxygen were required for the reaction. It was shown that the enzyme activity is located in a fraction corresponding to microsomes. The reaction was inhibited by KCN, but not by CO. Menadione and potassium ferricyanide inhibited the NADPH- and NADH-dependent reactions, respectively, and cytochrome c inhibited both of them. These results suggested an involvement in delta 5-desaturation of a mixed f

The 5' ends of yeast killer factor RNAs are pppGp.

The 5' nucleotides of the double-stranded RNAs of yeast killer factor have been isolated by digestion with pancreatic, T1 and T2 RNase followed by two-dimensional electrophoresis. They were identified by bacterial alkaline phosphatase and snake venom phosphodiesterase digestions. Both the larger double-stranded RNA (L, of 2.5 x 10(6) daltons) and the smaller double-stranded RNA (M, of 1.4 x 10(6) daltons) have the 5' end groups pppGp. These 5' ends are dissimilar to those of the double-stranded RNAs of animal viruses but may be characteristic of the 5' ends of the double-stranded RNAs of funga

Development of mitochondrial membranes in anaerobically grown yeast cells.

Biochemical analyses of mitochondrial marker substances, especially cardiolipin and oligomycin-sensitive ATPase [EC 3.6.1.3], as well as electron microscopic observations were carried out to eludicate the process of mitochondrial development in annaerobic yeast cells. Cardiolipin was found to be localized in the mitochondria in anaerobic cells. Its cellular content was a little higher in the stationary phase than in the exponential phase in glucose-grown cells and increased further in galactose-grown cells. The lipid content of the mitochondrial preparation obtained from glucose-grown stationa

White spots and gaps of microbial single cell proteins for food applications: yeasts, bacteria and filamentous fungi.

This review comprehensively evaluates microbial single cell proteins (SCPs) including yeasts, bacteria and filamentous fungi as sustainable and high-quality protein sources for future food systems. Adopting a multidisciplinary perspective, it identifies key research gaps and white spots along the entire value chain, from production to consumer acceptance. Yeast-based SCPs are the most advanced, with established protein ingredients or biomasses and broad functional potential, while filamentous fungi show strong relevance for meat analogues due to their inherent structuring properties. In contra…

Antifungal Potential of Peptide-Enriched Fractions Isolated from Capsicum frutescens Seeds Against Yeasts of the Genus Candida.

The increasing emergence of drug-resistant fungal pathogens has heightened the search for new antimicrobial compounds with previously unexplored modes of action. Among candidate molecules for control of fungal diseases, plant antimicrobial peptides (AMPs) have attracted considerable interest because they are natural molecules with broad-spectrum inhibitory activities against microorganisms. In this study, peptide-enriched fractions obtained from Capsicum frutescens seeds were evaluated for antimicrobial activity and mechanisms of action against Candida species. Hemotoxicity was evaluated in vi

Molecular evolution of the members of the Snq2/Pdr18 subfamily of Pdr transporters in the Hemiascomycete yeasts.

The transporters of the ATP-Binding Cassette (ABC) Superfamily involved in the Multidrug Resistance (MDR) phenomena are also known as ABC-Pleiotropic Drug Resistance (PDR) proteins. The homologs of the Saccharomyces cerevisiae SNQ2 and PDR18 genes were identified in 171 yeast genomes, representing 68 different hemiascomycetous species. All early-divergent yeast species analyzed in this work lack Snq2/Pdr18 homologs, suggesting that the origin of these ABC-PDR genes in hemiascomycete yeasts resulted from a horizontal transfer event. The evolutionary pathway of the Snq2/Pdr18 protein subfamily i…

Geo-temporal distribution and biotechnological valorization of indigenous soil yeasts in Mediterranean agroecosystems for sustainable farming solutions.

This paper focuses on the environmental distribution (geographical and temporal) of indigenous yeast between four Mediterranean agroecosystems (barley, faba bean, olive, and vine) and assesses their efficacy as biofertilisers. The analysis indicates high seasonal variations and spatial heterogeneity of yeast communities and density extended between 3 × 102 and 4.5 × 104 CFU/g of soil that depend on five determining factors, which control this distribution, namely, crop type, phenological cycles, rhizospheric effects, vertical stratification, and seasonal variation. These populations are also g

Molecular identification of yeasts involved in the alcoholic fermentation of Tchoukoutou and Atan in Benin using sequencing.

In Benin, the consumption of local beverages is increasing, particularly "Tchoukoutou" and "Atan". However, they face numerous challenges that hinder their recognition as fermented drinks with potential medical and cultural benefits. This study aims to determine the biodiversity of yeasts responsible for the fermentation of "Tchoukoutou" and "Atan" produced in Benin to promote their production and commercialization. To achieve this, beverage samples were randomly collected from six (06) municipalities in Benin and subjected to physicochemical and microbiological analysis. Yeast identification …
