Janus microneedles enabling orderly vitiligo treatment through spatiotemporal sequential promoting melanogenesis to transfer via PAR2 activation.

Vitiligo is characterized by a complex pathogenesis involving multiple factors, necessitating therapeutic strategies that simultaneously achieve immune regulation, melanocyte regeneration, and melanosome transfer modulation. Here, we designed a Janus microneedle (Janus MN) based on a core-shell bilayer structure, inspired by the melanin metabolism pathway and the stratified architecture of the skin. The shell of the MN was loaded with a complex of hypericin-assembled nanoparticles (HY-NPs) and astragalus polysaccharides, which preferentially dissolve in the epidermal hair follicle region and b

Nano-pathogenoids hitchhike activated neutrophils for eliminating of atherosclerosis plaques.

Atherosclerosis (AS) is a chronic inflammatory disease characterized by plaques that are difficult to eliminate. Neutrophil extracellular traps (NETs) are a central factor in the progression of AS, which could trigger a cascade of inflammatory reactions and increase plaque volume. Focusing on inhibiting the release of NETs within plaques is beneficial for erasing plaques. However, the arterial stenosis and poor blood flow caused by plaques prevent most therapeutic agents from entering the AS lesions. Considering that activated neutrophils firstly migrate to the interior of plaques after being

Autosplenectomy in a Patient With Autoimmune Polyglandular Syndrome Type 2 (APS-2).

Autoimmune glandular syndrome type 2 is a complex genetic condition where a triad of endocrinopathies is involved, namely, Addison's disease, type 1 diabetes, and/or autoimmune thyroid disorder. The disease predisposes one to a variety of other autoimmune associations. Here, we report a rare presentation of a patient with autoimmune polyglandular syndrome type 2 (APS-2) presenting with a 7-year history of progressive splenic atrophy causing functional hyposplenism that ultimately progressed to anatomical asplenia (autosplenectomy) as demonstrated in the serial imaging. We postulate that the un

Massive ascites and adnexal masses mimicking malignancy: A case report of Chlamydia trachomatis infection diagnosed by metagenomic next-generation sequencing.

OBJECTIVE: Chlamydia trachomatis (C. trachomatis) is the most commonly reported bacterial sexually transmitted infection among sexually active women. Although often asymptomatic or associated with non-specific clinical manifestations, it can cause inflammatory exudates and encapsulated fluid collections that are similar to adnexal masses on imaging. In rare cases, it may also present with massive ascites, a constellation of findings that may mimic ovarian malignancy. CASE: We report the case of a 32-year-old female with a nearly 3-month history of abdominal pain, adnexal masses and massive as

Hollow Mesoporous Silica Nanoparticles Co-Loaded with Docetaxel and Indocyanine Green for Synergistic Chemo-Photothermal Therapy.

Hollow mesoporous silica nanoparticles (HSNs) were synthesized via the Stöber method using resorcinol-formaldehyde resin as a template and further developed as a multifunctional nanocarrier for synergistic chemo-photothermal therapy. Docetaxel (DTX) and indocyanine green (ICG) were co-loaded into HSNs as the prodrug and photothermal agent. The loading sequence of these agents can critically affect encapsulation efficiency. Preloading DTX followed by ICG incorporation achieved the highest drug loading (38.65%) and preserved the photoactivity of ICG. The resulting ICG&DTX@NH2-HSNs exhibited stro

Development of Microfluidic Culture System for Spatially Regulated Differentiation of Mesenchymal Stem Cells Toward Osteochondral Interface Regeneration.

Regeneration of the osteochondral interface is essential for effective articular cartilage repair and the prevention of graft delamination. In this study, we developed a microfluidic culture device for the spatial regulation of mesenchymal stem cell (MSC) differentiation within a single-phase hydrogel by controlling concentration gradients of differentiation-inducing factors. The system comprised a PDMS culture chamber with dual-flow of culture media to generate concentration gradients of chondrogenic and osteogenic inductive factors. Numerical simulations were performed to characterize the fl

The role of exosomal long non-coding RNAs in head, neck and thyroid cancers.

Exosomal long non-coding RNAs (lncRNAs) have emerged as critical regulators in the pathogenesis, progression, and metastasis of head, neck, and thyroid cancers. These molecules, encapsulated within exosomes and released into biological fluids, mediate intercellular communication and influence various oncogenic processes including epithelial-mesenchymal transition (EMT), angiogenesis, immune evasion, and therapy resistance. Recent studies have demonstrated that specific exosomal lncRNAs are differentially expressed in cancer patients compared to healthy individuals, making them promising candid

Bioengineered hybrid dual-targeting nanoparticles reprogram the tumour microenvironment for deep glioblastoma photodynamic therapy.

Glioblastoma (GBM) poses significant therapeutic challenges due to its hypoxic and immunosuppressive tumour microenvironment (TME), low immunogenicity and physical barriers. While combining photodynamic therapy (PDT) with immunotherapy holds promise, its efficacy is hampered by insufficient immune activation. In this study, we develop a multifunctional photodynamic-enhanced biomimetic intelligent nanoplatform (FBFO@HM@aOPN) responsive to the TME. The nanoplatform consists of a dual-enzyme nanozyme encapsulated in a prokaryotic-eukaryotic hybrid membrane, further modified with a pH-sensitive tu

Lysozyme Fiber-Inspired Versatile Supramolecular Hydrogel Against Drug-Resistant Pathogens and Biofilm Formation.

Multidrug-resistant (MDR) bacterial infections pose a significant threat to human health, particularly when introduced by surgical contamination, resulting in early-stage implant-related biofilm infections and potential implant failure. Inspired by the structure and function of lysozyme fibers, a biomimetic supramolecular hydrogel is developed that mimics the lysozyme fiber architecture and encapsulates D-tyrosine (D-Tyr), integrating bactericidal and anti-biofilm functions within a self-assembled positively charged nanofibrous network. The peptide backbone with quaternary ammonium salt-modifi

Rapid neuralized and vascularized osteogenesis in infected bone defect using biomimetic biomineralized and antibacterial hydrogels.

Infected bone defects represent one of the most prevalent clinical conditions, affecting millions of patients annually. The local infection and necrosis associated with these defects exacerbate the injury, prolong healing times, and result in significant localized pain, presenting a substantial challenge for clinical repair. In this study, we developed a biomimetic mineralized and antibacterial imCOL1MA hydrogel by employing methacrylated COL1, composite native bone inorganic salts (CNBIS), and Magainin II-PLGA microspheres (mMicrospheres), which was further loaded with bone marrow stem cells

Bioresponsive nanoreactor initiates cascade reactions for tumor vascular normalization and lactate depletion to augment immunotherapy.

Immune checkpoint blockade (ICB) therapy has revolutionized cancer treatment. However, abnormal tumor vasculature and excess lactate contribute to tumor immunosuppression and confer resistance to ICB therapy, seriously limiting its clinical application. Here, we have developed a bioresponsive nanoreactor, ALMn, which consists of hollow manganese dioxide nanoparticles with encapsulation of lactate oxidase and L-Arginine, to overcome immunosuppression and sensitize ICB therapy. In the tumor microenvironment, lactate oxidase catalyzes lactate to produce hydrogen peroxide, which subsequently oxidi

Exploring multi-functional biopolymer polyhydroxyalkanoates in diabetes treatment.

Millions of individuals worldwide suffer from a chronic metabolic disorder, diabetes, defined as a reduction in insulin production or sensitivity, which raises blood glucose levels, weakens the immune system, and results in irregularities in the metabolism of carbohydrates, fats, and proteins. Therefore, there is still a high demand for non-invasive ways to administer insulin and other antidiabetics to treat diabetes and suitable therapeutics for wound healing. This generates a need for novel biomaterials that effectively use diabetes-associated therapy. This article emphasized that some speci

Spatial Profiling Identifies Regionally Distinct Microenvironments and Targetable Immunosuppressive Mechanisms in Pediatric Osteosarcoma Pulmonary Metastases.

UNLABELLED: Osteosarcoma is the most common malignant bone tumor in young patients and remains a significant clinical challenge, particularly at the metastatic stage. Studies detailing the immunosuppressive mechanisms within the metastatic osteosarcoma microenvironment are needed to elucidate the cellular communities in the metastatic microenvironment that support metastatic growth and to identify therapeutic approaches to target the cross-talk between cancer cells and their microenvironment. In this study, we performed spatial transcriptional profiling on a cohort of osteosarcoma pulmonary me

Biomineralized Nanocomposite-Integrated Microneedle Patch for Combined Brachytherapy and Photothermal Therapy in Postoperative Melanoma Recurrence and Infectious Wound Healing.

In the surgical management of malignant melanoma, incomplete tumor resection and large-area cutaneous defects are major contributors to high locoregional recurrence and uncontrolled wound infections, resulting in poor prognosis and prolonged recovery times for patients. Herein, a versatile nanocomposite microneedle patch (referred to as GM-Ag2S/Ca32P) is designed to simultaneously eliminate residual tumor post-surgery and promote the healing of infectious wounds. This microneedle patch effectively penetrates subcutaneous tissues, delivering therapeutic payloads to infiltrating tumor cells and

Precisely edited gut microbiota by tungsten-doped Prussian blue nanoparticles for the treatment of inflammatory bowel disease.

Inflammatory bowel disease (IBD) is characterized by recurring gastrointestinal inflammation, accompanied by a significant rise in global prevalence and disease severity. The overaccumulation of reactive oxygen and nitrogen species (RONS) in the intestinal environment disrupts redox homeostasis and drives pathological overgrowth of Escherichia coli, which are central to IBD pathogenesis. Herein, we designed a multifunctional nanozyme (W-PB) to enable sustained and targeted regulation of intestinal homeostasis through dual mechanisms: specific inhibition of E. coli overgrowth during colitis and

Entrapped in the Cocoon: A Rare Case of Sclerosing Encapsulating Peritonitis in a Patient on Dialysis.

Sclerosing encapsulating peritonitis (SEP), also known as abdominal cocoon syndrome, is a rare and potentially fatal condition characterized by the encasement of the small intestine within a dense fibrotic membrane. Fewer than 250 cases have been reported worldwide. We present a case of SEP in a patient on chronic peritoneal dialysis (PD). A 66-year-old female with end-stage renal disease on chronic PD presented with worsening abdominal pain, distension, and hypotension. Laboratory evaluation revealed leukocytosis and an elevated peritoneal fluid white blood cell count. Peritoneal fluid cultur

Systemically delivered lipid nanoparticle-mRNA encoding lysosomal acid β-glucosidase restores the enzyme deficiency in a murine Gaucher disease model.

Gaucher disease (GD) is a rare genetically inherited illness caused by loss of lysosomal acid β-glucosidase (β-GCase) that leads to progressive accumulation of substrates, sphingolipid glucosylceramide (GL1) and glucosylsphingosine (lyso-GL1). The protein-based enzyme replacement therapy (ERT) requires frequent dosing due to short drug half-life causing challenges in long-term patient compliance. JCXH-301 is a lipid nanoparticle (LNP) encapsulated messenger RNA (mRNA) encoding β-GCase. Intravenous administration of JCXH-301 delivered the target mRNA to various tissues in mice with intracellula

Development of a blocking ELISA for evaluating neutralizing antibodies in human and canine serum based on rabies virus glycoprotein epitope I.

Rabies virus (RABV) is extremely hazardous to both humans and animals, causing up to 100 % death. Accurate and easy-to-use serological evaluation of vaccine potency following immunization is crucial for rabies control. In this study, recombinant RABV glycoprotein (rG) was designed and produced in 293FT cells. Subsequently, a monoclonal antibody (S049), against the antigenic epitope I of RABV glycoprotein, was screened. Using the recombinant RABV glycoprotein and S049, a blocking enzyme-linked immunosorbent assay (bELISA) was developed. The rG-encapsulated antigen was optimized to a concentrati

Programmed Nanocloak of Commensal Bacteria-Derived Nanovesicles Amplify Strong Immunoreactivity against Tumor Growth and Metastatic Progression.

Recent discoveries in commensal microbiota demonstrate the great promise of intratumoral bacteria as attractive molecular targets of tumors in improving cancer treatment. However, direct leveraging of in vivo antibacterial strategies such as antibiotics to potentiate cancer therapy often leads to uncertain effectiveness, mainly due to poor selectivity and potential adverse effects. Here, building from the clinical discovery that patients with breast cancer featured rich commensal bacteria, we developed an activatable biointerface by encapsulating commensal bacteria-derived extracellular vesicl
