Impact of Multiple Kidney Retransplants on Post-Transplant Outcomes in the United States.

BACKGROUND: Kidney retransplantation offers a valuable treatment option for patients who experience graft failure after their initial transplant. There is an increasing number of patients undergoing multiple retransplants. However, the impact of multiple kidney retransplants on post-transplant outcomes remains unclear. This study aimed to assess the association between the number of kidney retransplants and post-transplant outcomes in kidney retransplant recipients. METHODS: We used the Organ Procurement and Transplantation Network and United Network for Organ Sharing (OPTN/UNOS) database to

Long-term breast cancer risk post childhood cancer radiotherapy: data-driven screening recommendations for survivors.

OBJECTIVES: Modern radiotherapy techniques have significantly reduced radiation exposure to healthy tissues. This study analysed breast tissue radiation doses in a national pediatric radiotherapy database, explored the discrepancy between prescribed and actual doses and analysed shortcomings of current breast screening guidelines. METHODS: We analysed PediaRT database data on radiotherapy dose distributions for 5079 French childhood cancer patients (2013-present), focusing on photon therapy plans with available breast dose data (n = 428). Biological equivalent dose (BED) was calculated, and s

Characterising Glanzmann Thrombasthenia in the United States: Real-World Data From the EPIC Cosmos Dataset.

BACKGROUND: Glanzmann Thrombasthenia (GT) is a rare autosomal recessive bleeding disorder characterised by defective platelet aggregation due to abnormalities in the glycoprotein IIb/IIIa complex. With a lack of large real-world database studies, our study aimed to characterise the demographic, clinical and treatment profiles of GT patients using a large, national database. METHODS: A retrospective cohort study was conducted using the EPIC Cosmos database from 1 January 2013 to 31 December 2024. Patients with at least one encounter coded with the SNOMED code for GT (32942005) were included. T

An Evaluation of Duplicate Adverse Event Reports Characteristics in the Food and Drug Administration Adverse Event Reporting System.

INTRODUCTION: The Food and Drug Administration (FDA) Adverse Event Reporting System (FAERS) receives duplicate reports of adverse events associated with drug and therapeutic biological products. Duplicate reports, defined as multiple reports of the same adverse event(s) related to the administration of the same marketed product(s) to the same individual patient at a particular point in time, may be received in FAERS for many reasons. The presence of duplicate reports can negatively impact public health surveillance efforts by impeding both safety signal identification and signal evaluation. O

Progression trajectories of chronic overlapping pain conditions unveiled across two large clinical data warehouses.

Chronic overlapping pain conditions (COPCs) affect a wide population and incur a substantial disease burden including comorbid mental disorders. Although these comorbidities have been extensively documented, the longitudinal trajectories of their onset, particularly for those with three or more conditions, have been rarely studied. We conducted retrospective cohort studies to identify statistically significant COPCs and mental health trajectories in the All of Us Research Program (AoU v8, 338,170 persons) and the Mayo Data Warehouse (3,957,444 individuals). For each trajectory, cases were matc
