In halothane-anaesthetized cats implanted with several push-pull cannulae the release of dopamine from nerve terminals and dendrites of the two nigrostriatal dopaminergic pathways is shown to be asymmetrically or symmetrically regulated bilaterally when dopaminergic or GABAergic drugs are infused into one side of the nigra. Nigrothalamic GABAergic neurons may intervene in the asymmetrical regulation, as sagittal section of the thalamic massa intermedia blocks the contralateral effects induced by dopaminergic drugs. The role of thalamic nuclei in the bilateral regulation of dopamine and GABA re

Engineered cell-based therapy is an emerging approach for treating intractable diseases, in which the remote control of cellular behavior necessitates robust physical input-sensing-response systems. Nanomaterials represent promising tools to enable wireless remote control of cells. To achieve this, the precise anchoring of nanomaterials to target cells is crucial for nanomaterial-engineered cells to prevent undesired effects such as endocytosis and diffusion. Herein, we describe a general cell engineering strategy using nanomaterials to achieve remote-controlled and sustained insulin release f

Electroconvulsive therapy (ECT) is an established therapeutic method for the treatment of severe mental disorders refractory to pharmaco- and psychotherapy. ECT is a first-line treatment option in delusional disorders, severe depression with acute suicidal tendency or life-threatening catatonia. Usually, ECT is performed as a treatment series. Under short-term anaesthesia and muscle relaxation, tonic-clonic seizures are induced using an external stimulation electrode. Convulsion can be exerted by uni- or bipolar stimulation using an electric charge up to 1000 millicoulomb (mC) with an amperage

Transcranial Direct brain stimulation (tDCS) is commonly used in order to modulate cortical networks activity during physiological processes through the application of weak electrical fields with scalp electrodes. Cathodal stimulation has been shown to decrease brain excitability in the context of epilepsy, with variable success. However, the cellular mechanisms responsible for the acute and the long-lasting effect of tDCS remain elusive. Using a novel approach of computational modeling that combines detailed but functionally integrated neurons we built a physiologically-based thalamocortical

Since 1988, intermittent electric stimulation of the cervical portion of the left vagus nerve is proposed as additive treatment of epilepsy. The effects of vagus nerve stimulation (VNS) on the modulation of cerebral activity, shown in animals and confirmed by the data of functional imagery in human beings, can be apprehended by the anatomic and functional characteristics of this nerve, whereas the anti-epileptic mode of action remains unknown. Following five controlled multicentric studies, including 440 patients presenting refractory epilepsy, 20,000 patients worldwide have been treated by VN…

We studied the effects of a new bombesin/gastrin-releasing peptide (GRP) receptor antagonist, Leu13-psi-(CH2NH)-Leu14-bombesin, on the secretion of gastrin and somatostatin and on the motor activity of isolated perfused porcine antrum in response to infusions of GRP at 10(-10) or 10(-9) mol/l and in response to electric stimulation of the vagus nerves. GRP significantly increased the secretion of gastrin and somatostatin and increased the frequency of antral contractions threefold. At 0.5 x 10(-6) mol/l the antagonist completely abolished the effects on motality and gastrin secretion and stron…

1. Single units that responded to yaw rotation were recorded extracellularly in the caudal inferior olive (IO) of barbiturate-anesthetized cats. Of 276 neurons, 55 responded reliably to yaw, and extensive quantitative data were recorded from 25. 2. No yaw-sensitive IO neuron responded to somatosensory or auditory stimuli but two responded, though unreliably, to flash. 3. Yaw-sensitive IO cells fired at low (1-4 spikes/s), irregular rates during one direction of rotation. Though cells responded reliably during yaw, firing rates varied considerably from cycle to cycle. Despite adequate surgery a number of patients have a return of back pain and sciatica following operation, the so-called failed back. The results of a prospective study of 101 patients entering an intensive rehabilitation programme for the failed back is described. The programme consists of a team approach to the patient and his problems, using a variety of techniques to produce pain relief. Non-organic pain represents a significant problem in the failed back; 58% of patients were completely or substantially relieved of their pain. Transcutaneous nerve stimulation was found to be the single mo

Electrical stimulation of the hypothalamic dorsomedial and wentromedial nuclei induced an activation of the coagulating system in rats. Plasmin in the vascular bed increased the electrical activity of medial and mamillary hypothalamic neurons, the activity coinciding in time with the maximal level of the blood hypercoagulating response. The activation does nor occur after plasminogen administration. The activation of the blood coagulating system is due to an excessive amount of plasmin in the blood and can be suppressed with sympatholytic agents. The plasmin-induced electrical activity is supp

Cortical activity can be modulated by endogenous and exogenous electric fields (EFs). Recent experimental and computational data suggested that endogenous EF-mediated effects are compatible with electric dipoles, which contribute to the synchronization of neighboring cortical columns. Consistently, exogenous EFs created by means of transcranial direct-current stimulation (tDCS) have shown that the orientation of current flow determines the effect of the intervention. Here, we investigated the impact of an exogenous EF's orientation on cortical modulation. We hypothesized that the columnar orga

A new therapeutic approach, known as neuromodulation therapy-which encompasses a variety of interventional techniques meant to alter the nervous system in order to achieve therapeutic effects-has emerged in recent years as a result of advancements in neuroscience. Currently used methods for neuromodulation include direct and indirect approaches, as well as invasive and non-invasive interventions. For instance, the two primary methods of stimulating the vagus nerve (VN) are invasive VN stimulation (iVNS) and transcutaneous VN stimulation (tVNS). Since the latter is non-invasive, basic, clinical

BACKGROUND: Implanted vagus nerve stimulation (VNS), when synchronized with post-stroke motor rehabilitation improves conventional motor rehabilitation training. A non-invasive VNS method known as transcutaneous auricular vagus nerves stimulation (taVNS) has emerged, which may mimic the effects of implanted VNS.
OBJECTIVE: To determine whether taVNS paired with motor rehabilitation improves post-stroke motor function, and whether synchronization with movement and amount of stimulation is critical to outcomes.
METHODS: We developed a closed-loop taVNS system for motor rehabilitation called mo
