Binding of ovarian cancer cells to peritoneal mesothelium in vitro is partly mediated by CD44H.

Epithelial cancer of the ovary spreads by implantation of tumor cells onto the mesothelial lining of the peritoneal cavity. We have developed an in vitro binding assay using confluent monolayers of normal peritoneal mesothelial cells in order to assess the role of known adhesion proteins in this process. Cells from normal ovarian surface epithelium and the ovarian cancer cell lines CAOV-3 and SKOV-3 exhibited significant adhesion to mesothelium in vitro (range 33-56% specific binding). Although these cells expressed several adhesion molecules, including CD44 and integrins such as alpha 4 beta …

Diversity amongst the microglia in growing and regenerating fish CNS: immunohistochemical characterization using FL.1, an anti-macrophage monoclonal antibody.

We have immunohistochemically characterized the forms and distribution of microglia--the macrophages of the CNS--in fish, using a new monocl … ary astrocytes elsewhere in the CNS. An antiserum against the beta 2 subunit of the human leukocyte integrins (Kishimoto et al., Cell 1987a; 50:193-202) was found selectively to recognize the foamy macrophages in Oreochromis. Following lesion to the optic nerve, FL.1-labelling shows that microglia proliferate throughout the visual pathway. In the optic tect …

Modulation of the FAK-Src Signalling Axis by Oncoviruses: Molecular Mechanisms, Functional Consequences, and Therapeutic Opportunities.

Oncoviruses play a role in cellular transformation and cancer progression by manipulating host signalling pathways, such as the FAK-Src axis. Preclinical evidence suggests that viral proteins can modulate this axis through direct interactions, integrin signalling, tumour suppressor inactivation, transcriptional and kinase reprogramming, calcium/ROS signalling, regulation of phosphatases, and epigenetic modifications. Important examples include HPV E2 and E6/E7, EBV LMP1 and LMP2A, HBV HBx, HCV Core and E2, and KSHV proteins. FAK-Src disruption can activate pathways such as PI3K/AKT, Rho GTPase

Versican: A Promising Therapeutic Target for Post-Myocardial Injury Cardiac Repair.

This review re-evaluates the literature from a matrix-centered perspective and determines how strongly it supports versican as a regulator of myocardial renewal rather than merely a marker of tissue remodeling. The evidence is organized around molecular identity, developmental location, temporal behavior after infarction, and causal intervention. During cardiac formation, versican-rich extracellular matrices recur in cushions, valves, septal and outflow structures, where matrix hydration, cell adhesion, and proteolytic processing accompany morphogenesis. In adult injury models, versican abunda

Integrated Multiomics Profiling Reveals Membrane-Associated Signaling Alterations Associated With Disease Activity in Systemic Lupus Erythematosus.

Systemic lupus erythematosus (SLE) is a complex autoimmune disease characterized by pervasive immune dysregulation, yet membrane-associated molecular alterations in immune cells remain incompletely defined. Here, we performed integrated proteomic and phosphoproteomic profiling of peripheral blood mononuclear cells (PBMCs) from a self-established cohort of 130 SLE patients across disease stages and 90 healthy controls, with individual samples pooled into 36 samples for LC-MS/MS analysis, complemented by publicly available transcriptomic data from 1461 SLE patients. Focusing on membrane-associat

Mechanotransduction through LFA-1 controls upstream migration in the KG-1a stem cell line.

Immune cell recruitment into tissues requires cells to migrate on the endothelium while overcoming shear forces. Many extravasating cells, including T lymphocytes and hematopoietic stem cells, migrate upstream against the direction of shear flow on ICAM-1. Upstream migration has been shown to depend on intracellular signaling through Crk family adaptor proteins following engagement of lymphocyte function-associated antigen-1 (LFA-1). Because upstream migration is an important illustration of leukocyte mechanotransduction, we sought to identify key intracellular regulators of upstream migration

The mechanical niche in gastric precancer: extracellular matrix remodeling, mechanotransduction, and stem-cell fate.

BACKGROUND: The Correa cascade foregrounds infection, inflammation, and epithelial change but does not fully explain how the physical tissue environment influences progression from atrophy to metaplasia and dysplasia. METHODS: We integrated human observations, gastric organoid and animal perturbation studies, advanced gastric-cancer experiments, and cross-tissue evidence using the Gastric Precancer Mechanobiology Evidence Stratification framework, which ranks evidence by proximity to causal human precancerous mechanics. RESULTS: Direct human mechanical measurements remain sparse and methodol

Integrative Bioinformatic Identification and Molecular Docking of Quercetin and Sulforaphane-Associated Prognostic Targets in Pancreatic Adenocarcinoma.

Pancreatic adenocarcinoma (PAAD) remains a highly lethal malignancy with limited therapeutic options, motivating the search for robust prognostic markers and tractable therapeutic targets. In this study, we applied an integrative bioinformatic pipeline combining cross-cohort differential expression analysis, high-confidence protein-protein interaction network reconstruction, and topological hub-gene prioritization. Hub candidates were then intersected with curated target repertoires of multi-target chemicals (notably quercetin and sulforaphane [SFN]) to nominate pharmacologically accessible "e

ADAM9: an updated view of its biology and pathology.

A disintegrin and metalloprotease 9 (ADAM9) is a catalytically active member of the ADAM family, a group of transmembrane and secreted proteins that regulate adhesion, proteolysis, migration, and cell signaling. ADAM9 exists in two isoforms, ADAM9-L (membrane bound form) and ADAM9-S (secreted form), which differ in structure and have opposing roles in cell migration. Its multiple domains support ectodomain shedding, integrin binding, intracellular signaling, and interactions with SH3-containing proteins. The activity of ADAM9 is regulated at different levels through transcriptional, post-trans

Bioinspired Anisotropic Collagen-Based Conductive Hydrogels Promote Neuronal Differentiation via Activation of Mechanoelectrical Signaling.

Simulating the anisotropic structure and electromechanical signals of the natural neural extracellular matrix (ECM) is crucial for advancing neural regeneration and constructing in vitro research models. Herein, a three-dimensional collagen-based conductive hydrogel with oriented fibers was constructed by controlling the intrinsic assembly behavior of collagen by combining mechanical pre-deformation with magnetic-driven stretching during gelation. The anisotropic hydrogel exhibited excellent cell compatibility and could guide the orientation and extension of PC-12 cell axons along the fibers.

MZT2A drives epithelial-mesenchymal transition in lung adenocarcinoma via the LGALS3BP/ITGB1/TGF-β/smad2 axis.

The high mortality of lung adenocarcinoma (LUAD) is largely attributed to its metastatic propensity; therefore, elucidating novel mechanisms driving metastasis is crucial for developing diagnostic and therapeutic strategies. This study aimed to elucidate the function and mechanism of MZT2A in the metastasis of LUAD. Analysis of clinical samples and public databases revealed that MZT2A is highly expressed in LUAD tissues and significantly associated with lymph node metastasis, advanced stage, and poor prognosis. In vitro functional assays revealed that MZT2A overexpression significantly enhance

TurboID-Based Proximity Labeling Map Reveals the Protein Interaction Network and Anti-Viral Mechanism Against Influenza A H1N1.

Influenza A virus (IAV) remains a significant public health threat due to its high variability and pathogenicity. Systematic identification of viral-host interfaces is critical for developing targeted therapies. In the present study, TurboID proximity labeling was applied to map interactomes of 10 H1N1 viral proteins in human alveolar epithelial cells A549. Key interactions were validated via co-immunoprecipitation (Co-IP), confocal microscopy, dual-luciferase reporter assays, and functional studies. Results showed that Hemagglutinin (HA) and polymerase acidic (PA) directly bound mitochondrial

Paxillin mediates endothelial cell migration and angiogenesis following spinal cord injury: a transcriptomic and functional analysis.

Spinal cord injury (SCI) is a destructive neurological condition that leads to significant functional deficits in the affected individual. To map the altered pathways within the lesion epicentre and the surrounding rostrocaudal segments, we performed RNA-sequencing on injured spinal cord tissue. Samples were collected from three regions-rostral, epicentre, and caudal-at Days 1, 14, and 28 post-injury to systematically profile the transcriptomic changes and identify pathways associated with angiogenesis following SCI. Gene set enrichment analysis revealed enriched pathways, including hepatocyte

Cytotoxic T cell recognition of α-synuclein drives pathogenic immune responses in multiple system atrophy.

Multiple system atrophy (MSA) is a progressive neurologic disease, known as an α-synucleinopathy. There are currently no effective disease-modifying therapies for MSA. While neuroinflammation is a hallmark of MSA, the contribution of adaptive immune mechanisms remains poorly understood. Here, we profiled peripheral and central T cell responses in patients with MSA, in comparison with Parkinson's disease (PD) and healthy control cohorts, using single-cell transcriptomics, flow cytometry, and antigen-specific functional assays. We demonstrated that peripheral T cells from MSA patients are activa

Oncogenic overexpression of kallikrein-related peptidase 7 is associated with the progression of thyroid cancer.

Mounting evidence suggests that the kallikrein-related peptidase family is dysregulated in thyroid cancer. However, previous analyses of kallikrein-related peptidase 7 (KLK7) expression patterns have yielded inconsistent results. In this study, KLK7 expression was examined in different histological types of thyroid cancer using immunohistochemistry. We found no detectable KLK7 immunoreactivity in the follicular epithelium of normal thyroid tissue or follicular adenomas. Weak KLK7 expression was observed in papillary (mean H-score 47) and follicular carcinomas (mean H-score 29), with moderate e

The dysadherin/carbonic anhydrase 9 axis shapes an acidic tumor microenvironment to promote colorectal cancer progression.

The tumor microenvironment (TME) plays a central role in cancer progression and metastasis. A key feature of the TME is extracellular acidity, which promotes disease progression, immune evasion, and drug resistance. Tumor acidity is increasingly recognized as a critical factor in cancer development and a negative prognostic indicator. Here, we demonstrate that the membrane glycoprotein dysadherin promotes colorectal cancer (CRC) malignancy by modulating TME acidity. Comprehensive bioinformatics and pathological analyses of CRC patient samples revealed that increased tumor acidity is a hallmark

Cellular and Molecular Mechanisms of SARS-CoV-2 Spike Protein-Induced Endothelial Dysfunction.

Severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) infection is initiated by the viral spike proteins, which are key structural components that mediate host cell binding and entry and alter downstream signaling through multiple interactions with endothelial surface receptors. Endothelial dysfunction is a central consequence of COVID-19, contributing to vascular inflammation, barrier disruption, thrombosis, and multi-organ injury affecting the pulmonary, cardiovascular, cerebral, and renal systems. Emerging evidence demonstrates that spike protein-mediated effects, independent of prod

Human Blood IgA+ Memory B Cells Differ From IgG+ B Cells by Expressing Gut-Homing and Regulatory Markers.

IgA is the most produced immunoglobulin by the human body, mainly in secretions, and it exerts an important role in the activation and regulation of the immune system. Despite its importance, the phenotype of circulating IgA+ B cells is poorly described. Here, we deeply explored the phenotype of IgA+ memory B cells compared with IgG+ memory B cells from healthy donors using spectral flow cytometry. A bulk transcriptomic analysis was performed on isolated IgA+, IgG+ and IgM+ memory B cells. IgA2+ memory B cells expressed more gut-homing markers (CCR9, CCR10, integrin α4β7) and CD11b, GPR183, wh
