[Viruses and unconventional transmissible agents: update on transmission via blood ].

The blood borne viruses must be separated into major and minor agents. Major viruses transmissible by blood transfusion are human immunodeficiency virus (HIV) and hepatitis B and C viruses (respectively HBV, HCV). The prevalence of virological markers in French blood donors has been continuously decreasing since the implementation of serological screening methods as soon as they were available. In 1998, the prevalences (per 10,000 donations) were 0.17 for antibody to HIV, 0.08 for antibody to human T-cell leukemia virus (HTLV), 2.23 for hepatitis B surface antigen (HBs Ag) and 2.52 for antibod

Primary tubercular granulomatous hepatitis presenting as fluctuating jaundice.

Granulomatous hepatitis is an uncommon presentation of tuberculosis (TB). It is even more peculiar to have TB confined to the liver alone with no pulmonary or a disseminated form. In either form, there is the usual presentation of nonprogressive cholestatic jaundice, but no documented case with fluctuating jaundice in the literature was found. In order to highlight this rare presentation aiding the right diagnosis, we present one such case of a 46-year-old woman with no known comorbidities, who complained of fluctuating and painless type of jaundice, associated with fatiguability, pruritus, an

Revealing Nanomedicine Design Principles for Efficient siRNA Delivery to the Central Nervous System.

A series of polymer nanocapsules were designed and synthesized for non-viral siRNA delivery to the central nervous system via intracerebroventricular injection. Various factors can contribute to superior diffusion profiles. In this study, we find that size, charge, and morphology play important roles. Further, we report an analysis that demonstrates the subtle effects that small chemical changes can have on whole-particle properties and the corresponding in vivo readouts. We use computational modeling to reveal previously unconsidered design criteria and hypothesize that cargo-templating consi

Panobinostat suppresses cGAS-STING pathway activation and ameliorates DSS-induced colitis in mice.

Abnormal or excessive activation of the cyclic GMP-AMP synthase (cGAS)-stimulator of interferon genes (STING) pathway in natural immunity nucleic acid sensing causing the overproduction of cytokines, triggering inflammatory tissue damage, immune pathology, or autoimmune diseases closely associated with the release of type I interferons. Inhibiting aberrant cGAS activation represents a promising therapeutic strategy for clinical development. Through screening a library of epigenetic drugs, we identified that the histone deacetylase inhibitor panobinostat significantly suppresses the cGAS-STING

Ultra-Sensitive Detection of Chloramphenicol by CdS@NiMoS Nanorods-Based Photoelectrochemical Aptasensor.

A novel nanomaterial photoelectrochemical aptamer sensor based on CdS@NiMoS heterojunction nanocomposites was constructed for highly sensitive detection of chloramphenicol (CAP) in antibiotic residues. Through optimization of the material synthesis process, the optimal doping ratio of MoS2 to Ni3+ (70% MoS2 and 10% Ni3+) was identified, which significantly enhanced the photogenerated carrier separation efficiency. In thin-film preparation, comparative analysis of four film-forming methods led to the determination of an optimal process with stability. To achieve highly specific CAP detection, t

Differential diagnosis of perianal herpes: sexual abuse or autoinoculation?

A 13-year-old boy was referred to the forensic department for suspicion of unreported sexual abuse due to the presence of perianal dermatologic lesions. The forensic examination revealed perianal herpes simplex virus type 1 (HSV-1) lesions using nucleic acid amplification by polymerase chain reaction (PCR) outside of any traumatic or other dermatological lesions. Repeated serological tests for sexually transmitted infections (STIs) were negative. However during the psychological interview, the child reported penile touching by a 15-year-old boy at least one week earlier. Both the examination o

TB dactylitis in a toddler.

A well-thriving male toddler presented with a painless swelling on the dorsal surface of his right hand, incidentally noticed by the mother. There was no history of local trauma or any systemic symptoms. On examination, there were no signs of inflammation over the swelling. An X-ray was taken, and an expansile lytic lesion of the right fifth metacarpal bone was seen. He underwent debridement, and the tissue was sent for histopathological examination (HPE) and cartridge-based nucleic acid amplification test (CBNAAT) for Mycobacterium tuberculosis (M. tb). HPE revealed ill-defined granulomas and

Methylation in cornea and corneal diseases: a systematic review.

Corneal diseases are among the primary causes of blindness and vision loss worldwide. However, the pathogenesis of corneal diseases remains elusive, and diagnostic and therapeutic tools are limited. Thus, identifying new targets for the diagnosis and treatment of corneal diseases has gained great interest. Methylation, a type of epigenetic modification, modulates various cellular processes at both nucleic acid and protein levels. Growing evidence shows that methylation is a key regulator in the pathogenesis of corneal diseases, including inflammation, fibrosis, and neovascularization, making i

Impact of glycosylation on viral vaccines.

Glycosylation is the most prominent modification important for vaccines and its specific pattern depends on several factors that need to be considered when developing a new biopharmaceutical. Tailor-made glycosylation can be exploited to develop more effective and safer vaccines; for this reason, a deep understanding of both glycoengineering strategies and glycans structures and functions is required. In this review we discuss the recent advances concerning glycoprotein expression systems and the explanation of glycans immunomodulation mechanisms. Furthermore, we highlight how glycans tune the

Point-of-care testing of methamphetamine and cocaine utilizing wearable sensors.

The imperative for the point-of-care testing of methamphetamine and cocaine in drug abuse prevention necessitates innovative solutions. To address this need, we have introduced a multi-channel wearable sensor harnessing CRISPR/Cas12a system. A CRISPR/Cas12a based system, integrated with aptamers specific to methamphetamine and cocaine, has been engineered. These aptamers function as signal-mediated intermediaries, converting methamphetamine and cocaine into nucleic acid signals, subsequently generating single-stranded DNA to activate the Cas12 protein. Additionally, we have integrated a microf

The prototypical interferonopathy: Aicardi-Goutières syndrome from bedside to bench.

Aicardi-Goutières syndrome (AGS) is a progressive genetic encephalopathy caused by pathogenic mutations in genes controlling cellular anti-viral responses and nucleic acid metabolism. The mutations initiate autoinflammatory processes in the brain and systemically that are triggered by chronic overproduction of type I interferon (IFN), including IFN-alpha. Emerging disease-directed therapies aim to dampen autoinflammation and block cellular responses to IFN production, creating an urgent and unmet need to understand better which cells, compartments, and mechanisms underlying disease pathogenesi

Nutcracker Syndrome and its Exceptional Occurrence with Pulmonary Tuberculosis: A Case Report.

Nutcracker syndrome (NCS), also known as left renal vein (LRV) entrapment syndrome, is a condition resulting due to compression of LRV between the aorta and superior mesenteric artery (SMA), with dilatation of the distal portion of LRV. We present a case of an elderly female presenting with left lumbar pain for 1 year. Initial investigations revealed microscopic hematuria and mild ascites. Further investigations revealed compression of LRV between the aorta and SMA, with aorto-SMA angle reduced up to 12°, suggesting a diagnosis of NCS. Along with this, cystic bronchiectasis with multiple centr

Peripancreatic tuberculosis.

Peripancreatic tuberculosis (PTB) is a very rare variant of tuberculosis and its clinical and radiological findings are similar to those of pancreatic malignancy. Diagnosis of PTB is usually incidental and is made after surgical resection. We are presenting a male patient who had complaints of prolonged fever, significant weight loss and yellowish discolouration of eyes and dark-coloured urine. Investigations revealed that there was a pancreatic mass causing obstructive jaundice. However, the aetiology of the mass, whether tubercular or malignant, was not clear. Hence, the patient was planned

Recent development of nanotechnology-empowered antigen assay methods for the control of infectious diseases.

The global spread of air-borne diseases, such as Covid-19 caused by the new coronavirus (SARS-CoV-2), has significantly impacted public health and economic development worldwide. Accurate and rapid detection of pathogens is the key to controlling the spread of infection and reducing severe illness and death. Compared to nucleic acid testing, rapid antigen testing for pathogen proteins shows unique advantages such as convenience, speed, and cost-effectiveness, but its sensitivity is limited. Here, we review the latest progress in the development of immunological assay methods for infectious dis

Poultry Vaccine Technology Platforms.

The different technology platforms used to make poultry vaccines are reviewed. Vaccines based on classical technologies are either live attenuated or inactivated vaccines. Genetic engineering is applied to design by deletion, mutation, insertion, or chimerization, genetically modified target microorganisms that are used either as live or inactivated vaccines. Other vaccine platforms are based on one or a few genes of the target pathogen agent coding for proteins that can induce a protective immune response ("protective genes"). These genes can be expressed in vitro to produce subunit vaccines.

Disseminated Gonococcal Infection With Dermatitis-Arthritis Syndrome.

Gonorrhea is a sexually transmitted infection caused by gram-negative diplococci, Neisseria gonorrhoeae. Disseminated gonorrhea is diagnosed infrequently, partly due to low suspicion at the time of presentation, and at times, due to overlapping symptoms associated with non-infectious conditions like systemic lupus erythematosus (SLE). In this article, we present a 42-year-old sexually active female with knee pain and swelling, fever, and rash. Knee aspirate showed the presence of monosodium urate crystals, and the synovial culture grew gram-negative diplococci, requiring multiple joint washout
