The occurrence of the snail Lymnaea columella on grazing areas in New South Wales and studies on its susceptibility to Fasciola hepatica.

Field surveys were carried out in the Lismore and Casino area for the presence of fresh water snails potentially responsible for the transmission of trematodes in ruminants. Although the North American snail, Lymnaea columella has previously only been reported from metropolitan areas, large populations of the snail were found east of Lismore. Natural infection with F. hepatica was detected in some of the snails and laboratory studies showed that this snail species was highly susceptible to infection with miracidia of F. hepatica and produced viable metacercariae. Three species of planorbid sna…

Permissive infection of murine osteoblastic MC3T3-E1 cells by goatpox virus: establishing a novel cell model for mechanistic studies of host-pathogen interactions.

Goatpox is a highly contagious viral disease that poses a significant threat to the health of goats and other small ruminants. Here, we report for the first time that the murine osteoblastic cell line MC3T3-E1 supports productive GTPV infection, as demonstrated by Western blotting, indirect immunofluorescence assay, and flow cytometry. RNA sequencing revealed thousands of differentially expressed genes (DEGs). Signaling pathways and functions of the DEGs were analyzed using Gene Ontology and the Kyoto Encyclopedia of Genes and Genomes. Real-time quantitative PCR further confirmed the expressio…

Molecular diagnosis of natural fasciolosis by DNA detection in sheep faeces.

Fasciolosis is an important parasitic zoonosis considered the most important helminth infection of ruminants in tropical countries. The aim of this study was to develop a PCR assay for the sensitive and specific detection of F. hepatica in formalin preserved sheep faeces. A 405-bp fragment of the cytochrome c oxidase subunit 1 gene of F. hepatica was amplified from stool samples of infected sheep. The PCR assay showed a detection limit of 20 pg of F. hepatica DNA. No cross-reactions were observed with samples containing coccidian oocysts or gastrointestinal nematodes eggs. Our PCR technique sh…

Mass spectrometry data from proteomics-based screening of immunoreactive proteins of fully virulent Brucella strains using sera from naturally infected animals.

Here, we provide the dataset associated with our research article on comprehensive screening of Brucella immunoreactive proteins using sera of naturally infected hosts published in Biochemical and Biophysical Research Communications Wareth et al., 2015 [1]. Whole-cell protein extracts were prepared from Brucella abortus and Brucella melitensis, separated using sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDS-PAGE) and subsequently western blotting was carried out using sera from bovines (cows and buffaloes) and small ruminants (goats and sheep). The mass spectrometry proteomics d…

Countermeasure development for Rift Valley fever: deletion, modification or targeting of major virulence factor NSs.

Rift Valley fever (RVF) is a mosquito-borne zoonotic disease characterized by a high rate of abortion in ruminants, and febrile illness, hemorrhagic fever, retinitis and encephalitis in humans. RVF is caused by the RVF virus (RVFV), belonging to the genus Phlebovirus of the family Bunyaviridae. RVFV encodes a major virulence factor, NSs, which is dispensable for viral replication, yet required for evasion of host innate immune responses. RVFV NSs inhibits host gene upregulation at the transcriptional level, while promoting viral translation in the cytoplasm. In this article, we summarize the v…

Hypereosinophilia and liver mass in an immigrant.

Human infection with the sheep liver fluke Fasciola hepatica is a global zoonosis that usually parallels the prevalence of infection in sheep and other ruminants. The disease is endemic in South and Central America, Puerto Rico, the Caribbean region, many parts of Africa, Asia, the Middle East, Australia, and China. There have been a number of focal outbreaks reported from Europe, including southern France and the Mediterranean region. Since acute fascioliasis has rarely been reported in the United States, physicians in this country frequently overlook the diagnosis. Therefore, we report a cas…

New variant Creutzfeldt-Jakob disease.

Since the report of new variant Creutzfeldt-Jakob Disease (nvCJD) in humans last year, the search was on for direct evidence to link the condition to Bovine Spongiform Encephalopathy (BSE). The first case nvCJD was noted 10 years after the recognition of BSE in UK cattle. A direct link is now established. There are, however, some 'protective' mechanisms, the most important of which are the inefficiency of the gastric route of introducing the infected material, the species barrier and genetic 'predisposition'. None of these is full proof and time will tell whether feeding ruminants with their o…

Health evaluation of black-faced impala (Aepyceros melampus petersi) using blood chemistry and serology.

As part of ongoing ecological studies of black-faced impala (Aepyceros melampus petersi) in northern Namibia, blood samples were collected and analyzed from 26 animals captured for translocation in 1992. All animals appeared to be in good condition and no abnormality was noted during physical examination. Serum chemistry and mineral levels were measured and correlated with the results of bacterial and viral serology and were within the normal ranges for domestic ruminants. Antibody titers for infectious bovine rhinotracheitis and bovine viral diarrhea were detected. Serological tests for bluet…

Helminths of sheep and goats in desert areas of south-west Mauritania (Trarza).

Faecal samples were obtained from sheep and goats before, during and after the rainy season at three locations in south-west Mauritania. Several animals were also necropsied at the same time. Haemonchus contortus was the most prevalent worm. Infection by digestive-tract strongyles and Strongyloides papillosus was always very light (prevalence less than 20%). Sheep were more heavily infected than goats but animals under 1 year of age were not infected by digestive-tract strongyles. It is likely that young small ruminants became infected during the rainy season and that the parasites so acquired…

Reconstituted versus dry alfalfa hay in starter feed diets of Holstein dairy calves: Effects on feed intake, feeding and chewing behavior, feed preference, and health criteria.

We investigated the effect of reconstitution of alfalfa hay on feeding and chewing behavior, sorting activity, and health status using 20 neonate Holstein male calves (3 d of age; 40.3 ± 1.30 kg of body weight) that were assigned randomly to 2 treatments, a starter feed containing either 10% dry (AH) or reconstituted alfalfa hay (RAH), each consisting of 10 calves. Alfalfa hay was reconstituted with water 24 h before feeding to achieve a theoretical dry mater (DM) content of 20%. Both starter diets had the same ingredients and nutrient compositions but differed in their DM content (90.9 vs. 66

Can an app designed to reduce repetitive negative thinking decrease depression and anxiety in young people? Results from a randomized controlled prevention trial.

BACKGROUND AND OBJECTIVES: Rates of mental health disorders are rising among adolescents and young adults. Therefore, scalable methods for preventing psychopathology in these age groups are needed. As repetitive negative thinking (RNT) is a risk factor for depression and anxiety disorders, targeting RNT via smartphone app promises to be an effective, scalable strategy. The current three-arm, parallel group, randomized controlled trial tested whether a self-help app designed to reduce RNT decreased psychopathological symptoms and RNT in adolescents and young adults at risk for mental disorders.

Nature lifts when feeling low: Daily high and low awe nature clips decrease repetitive negative thinking and dampening and increase subjective happiness in adults.

Exposure to nature can enhance mental well-being, making nature-based interventions promising for the treatment and prevention of mental health problems like depression. Given the decreased self-focus and sense of self-diminishment associated with awe, the present study investigated the impact of exposure to awe-evoking nature on two key risk and maintenance factors of depression-repetitive negative thinking (RNT) and dampening of positive feelings-and on subjective happiness. In a randomized controlled trial, we tested the effects of exposure to awe-evoking nature clips through a 1-week inter

"Sticky" thinking disrupts decision making for individuals with a tendency toward worry and depression.

Depressed individuals are commonly known to suffer from low mood. Less attention is paid to their decision-making deficiencies, consisting of indecisiveness and biased judgments. Many theories attempt to explain these impairments by focusing on reduced sensitivity to reward and punishment or biased information processing. Beyond these accounts, the present study explores another scenario, namely, whether the occurrence of sticky thinking-the occurrence of thoughts that are difficult to disengage from-could be a cause for the disruption of the decision-making process in individuals with depress

Impact of infectious diseases on wild bovidae populations in Thailand: insights from population modelling and disease dynamics.

The wildlife and livestock interface is vital for wildlife conservation and habitat management. Infectious diseases maintained by domestic species may impact threatened species such as Asian bovids, as they share natural resources and habitats. To predict the population impact of infectious diseases with different traits, we used stochastic mathematical models to simulate the population dynamics over 100 years for 100 times in a model gaur (Bos gaurus) population with and without disease. We simulated repeated introductions from a reservoir, such as domestic cattle. We selected six bovine infe

Multilocus Genotyping of Giardia duodenalis in Alpine Musk Deer (Moschus chrysogaster) in China.

Giardia duodenalis is the underlying cause of a significant number of outbreaks of gastrointestinal illness in humans and animals worldwide. The purpose of this study was to elucidate the prevalence and genetic diversity of G. duodenalis in captive alpine musk deer (Moschus chrysogaster) in China. A total of 202 fecal samples were collected from three farms in Gansu Province, China. Identification of G. duodenalis was conducted by nested PCR targeting the genes coding for SSU rRNA, β-giardin (bg), glutamate dehydrogenase (gdh) and triosephosphate isomerase (tpi). The overall prevalence of G. d

Deletion in KARLN intron 5 and predictive relationship with bovine tuberculosis and brucellosis infection phenotype.

Kalirin (gene: KALRN) is a Rho-GEF kinase linked to neurodegenerative diseases in humans. Unexpectedly, various polymorphisms in KALRN gene were previously associated with resistance to bacterial infections in ruminants. In this study, we evaluated the effect of the rs384223075 (RS-075) deletion in KALRN intron 5 on the occurrence of Mycobacterium bovis and Brucella abortus infections in cattle. We performed two separate case-control association analyses: one for bovine tuberculosis (bTB) using 308 Holstein and Jersey cows from three herds with prevalence between 5 and 15% for this infection; …

Cross-sectional survey for sheep leptospirosis in the northeast region of Brazil.

Sheep farming has shown expressive growth, even in the Northeast region of Brazil, with rainfall below expectations in the last decade. This growth has been observed not only in the number of herds, but also in the number of properties involved in sheep farming. Although expressive, the production of small ruminants still presents low levels of performance, mainly due to the occurrence of diseases, which constitute an obstacle to this activity. The aim of this study was to determine the factors associated with the seroprevalence of leptospirosis in sheep in Northeastern Brazil. Blood samples w…
