BACKGROUND: Thyroid cancers are among the growing list of cancer types that are resistant to immune-checkpoint inhibitor monotherapy. Although T-cell infiltration is common in follicular cell-derived thyroid cancers, tumor mutation burden is low. The antigenic potential of thyroid cancers is unknown.
METHODS: To investigate the anti-tumor T-cell response in thyroid cancer, we expanded tumor-infiltrating lymphocytes (TIL) from primary thyroid tumors and tumor-involved lymph nodes (TILN). Putative neoantigens and both tissue-associated and tumor-associated antigens were identified by targeted s

Transmembrane protein TMEM119 has been implicated in tumor progression (e.g., ovarian cancer), but its role and underlying mechanism in ovarian endometriosis (EM) remain elusive. Thus, this study aimed to investigate the expression pattern and functional role of TMEM119 in ovarian EM. To this end, TMEM119 expression was first analyzed using the GSE141549 dataset, and further detected in ovarian EM tissues and ovarian ectopic endometrial stromal cells (ESCs). After TMEM119 overexpression in ectopic ESCs, gene ontology/Kyoto Encyclopedia of Genes and Genomes enrichment analyses of differentially

The regulation of T cell exhaustion within the tumor microenvironment plays a pivotal role in shaping the immune response to cancer and determining the efficacy of immunotherapy. However, the molecular factors governing this process in colon cancer remain poorly understood. This study investigates the expression characteristics and functional significance of the transmembrane protein CD82 in the colon cancer immune microenvironment, with emphasis on its regulatory role in CD8+ T cell exhaustion and clinical outcomes. Publicly available transcriptomic datasets were integrated with multiplex imm

Fragrant pear canker, caused by Cytospora pyri, is a major branch disease that reduces the productivity and longevity of Korla fragrant pear orchards in Xinjiang, China. During infection and lesion expansion in woody tissues, C. pyri must breach the host cell wall barrier, in which pectin degradation plays a central role by weakening intercellular adhesion and disrupting tissue integrity. Members of glycoside hydrolase family 28 (GH28), represented mainly by polygalacturonases and other pectin-degrading enzymes, are closely involved in fungal invasion and colonization. However, the composition

Fritillaria thunbergii Miq. has been traditionally used to treat respiratory disorders in Chinese medicine. Peimisine, a major F. thunbergii alkaloid, has anti-inflammatory and antioxidant effects, but its role in allergic asthma and mucus-driven airway remodeling remains unclear. This study investigated whether peimisine could attenuate asthmatic phenotypes and explored its mechanistic links to Th2 inflammation, epithelial activation, and oxidative stress. Female BALB/c mice were sensitized and challenged with ovalbumin (OVA) and treated with peimisine. Airway hyperresponsiveness (AHR) and lu

AIM: Periapical mucositis (PAM) is defined as inflammation of the periapical tissues and localized oedema of the maxillary sinus mucosa, typically resulting from periradicular disease. Radiographically on cone-beam computed tomography (CBCT), PAM presents as mucosal thickening or a dome-shaped soft tissue expansion along the floor of the maxillary sinus, adjacent to the affected root apex. Although several studies have evaluated PAM, their findings vary considerably. This study aims (1) to evaluate the prevalence of PAM in maxillary posterior teeth amongst patients at NYU College of Dentistry,…

The inflammation-intestinal metaplasia (IM)-carcinoma cascade has been proposed as a framework for gastric cancer (GC) development, yet the cell-level heterogeneity and microenvironmental remodeling underlying this progression remain poorly characterized. Here, we constructed a single-cell transcriptomic atlas by integrating scRNA-seq data from chronic gastritis (superficial, CGS), IM, cancer-adjacent, and tumor tissues through a unified analytical pipeline. Seven major cell lineages were resolved. Relative to CGS, IM and GC tissues exhibited a progressive contraction of epithelial compartment

Iron (Fe) deficiency is a common nutritional disorder that causes iron chlorosis in crop production, affecting plant growth, development, yield, and quality. To elucidate the growth-physiological characteristics, iron accumulation dynamics, and allocation patterns of grapevines under long-term gradient iron deficiency stress, this experiment was conducted. Using 'Red Globe' grapevines as the experimental subject, four gradient Fe-deficiency treatments (1/5, 2/5, 3/5, and 4/5 of normal iron supply) were established under tissue culture conditions, with a control (CK) group maintained at normal

BACKGROUND: Directed growth modulation is commonly utilized as a surgical treatment for early-onset scoliosis. Growing rods are instrumented on the spine and apply sustained tension on the immature spine for a substantial amount of time, with the clinical goal of accommodating axial expansion of the spine. Despite the use of growing rods in humans, the mechanobiology of the spinal tissues under tensile loading remains relatively unknown. To bridge this knowledge gap, we developed a preclinical mouse model that allows for mechanistic investigations of sustained tension on the spine.

INTRODUCTION: Kaposiform lymphangiomatosis is a lymphatic anomaly that, when symptomatic, presents in children at a median age of 6.5 years. Symptoms may include respiratory issues, palpable masses, and hemostatic abnormalities. Kaposiform lymphangiomatosis can be life-threatening due to diffuse tissue expansion and subsequent invasion of surrounding organs and tissues. Initial detection of this rare condition can be difficult due to the varied nature of its presenting symptoms and the overall lack of familiarity of clinicians with the condition. Consequently, misdiagnoses can occur, such as p…

OBJECTIVE: To develop a novel Laryngopharyngeal Reflux Disease (LPRD) model in Bama pigs through endoscopic cricopharyngeal myotomy.
METHODS: A total of eight 8-month-old Bama pigs were randomly assigned to either the control or surgery group. Prior to intervention, upper esophageal sphincter (UES) manometry and laryngopharyngeal Dx-pH monitoring were conducted to establish baseline physiological parameters for each pig. Subsequently, the surgery group underwent endoscopic cricopharyngeal myotomy, while the control group did not. Two weeks postintervention, these procedures were repeated to e

BACKGROUND: Oxidative stress is implicated in the pathogenesis and progression of abdominal aortic aneurysm (AAA). Antioxidant delivery as a therapeutic for AAA is of substantial interest although clinical translation of antioxidant therapy has met with significant challenges due to limitations in achieving sufficient antioxidant levels at the site of AAA. We posit that nanoparticle-based approaches hold promise to overcome challenges associated with systemic administration of antioxidants.
METHODS: We employed a peptide-based nanoplatform to overexpress a key modulator of oxidative stress, s

BACKGROUND AND AIMS: Autologous keratinocyte sheets constitute an important component of the burn wound treatment toolbox available to a surgeon and can be considered a life-saving procedure for patients with severe burns over 50% of their total body surface area. Large-scale keratinocyte sheet cultivation still fundamentally relies on the use of animal components such as inactivated murine 3T3 fibroblasts as feeders, animal-derived enzymes such as trypsin, as well as media components such as fetal bovine serum (FBS). This study was therefore aimed to optimize autologous keratinocyte sheets by

We have previously shown that natural killer (NK) cells expand, and increase their function after interaction with cells that exhibit a number of different knock-down genes. We hypothesized that deletion or knockdown of a variety of key genes such as RAG may cause de-differentiation of the cells which could lead to increased NK expansion and function since we have shown previously that NK cells are activated and expanded by less differentiated cells. When comparing the function of NK cells from bone marrow (BM), spleen, pancreas, adipose tissue, and gingiva from WT mice to those from Rag2-/- m

The mucosa of the body of the stomach (ie, the gastric corpus) uses 2 overlapping, depth-dependent mechanisms to respond to injury. Superficial injury heals via surface cells with histopathologic changes like foveolar hyperplasia. Deeper, usually chronic, injury/inflammation, most frequently induced by the carcinogenic bacteria Helicobacter pylori, elicits glandular histopathologic alterations, initially manifesting as pyloric (also known as pseudopyloric) metaplasia. In this pyloric metaplasia, corpus glands become antrum (pylorus)-like with loss of acid-secreting parietal cells (atrophic gas

Devastating fires in Australia over 2019-20 decimated native fauna and flora, including koalas. The resulting population bottleneck, combined with significant loss of habitat, increases the vulnerability of remaining koala populations to threats which include disease. Chlamydia is one disease which causes significant morbidity and mortality in koalas. The predominant pathogenic species, Chlamydia pecorum, causes severe ocular, urogenital and reproductive tract disease. In marsupials, including the koala, gene expansions of an antimicrobial peptide family known as cathelicidins have enabled pro

PURPOSE: Limbal niche cells (LNCs) play a vital role in the maintenance of limbal epithelial stem/progenitor cells (LESCs). Four methods have been reported to isolate and expand LNCs: digestion by collagenase alone (C-LNC), collagenase following dispase removal of the limbal epithelium (DC-LNC), dissection of dispase-isolated limbal epithelial sheets (D-LNC), and explant cultures of limbal stromal tissues (Ex-LNC). This study aimed to isolate LNCs using those four methods and to compare their capacity to maintain LESCs.
METHODS: LNCs were isolated from the rat corneal limbus by the following

Previous studies have reported robust inflammatory cell infiltration, synthesis of IgG, B-cell clonal expansion, deposition of immune complexes and complement within cerebral cavernous malformation (CCM) lesions. B-cell depletion has also been shown to reduce the maturation of CCM in murine models. We hypothesize that antigen(s) within the lesional milieu perpetuate the pathogenetic immune responses in CCMs. This study aims to identify those putative antigen(s) using monoclonal antibodies (mAbs) derived from plasma cells found in surgically removed human CCM lesions. We produced human mAbs fro

The natural history of canine visceral leishmaniasis (CVL) has been well described, particularly with respect to the parasite load in different tissues and immunopathological changes according to the progression of clinical forms. The biomarkers evaluated in these studies provide support for the improvement of the tools used in developing vaccines against CVL. Thus, we describe the major studies using the dog model that supplies the rationale for including different biomarkers (tissue parasitism, histopathology, hematological changes, leucocytes immunophenotyping, cytokines patterns, and in vi
