An innovative AQP4 loop C-targeted monoclonal antibody reduces complement-dependent cytotoxicity and lessens experimental neuromyelitis optica.

Neuromyelitis optica spectrum disorder (NMOSD) is a neuroinflammatory disease caused by an autoantibody that targets the extracellular domain (ECD) of the astrocytic water channel aquaporin-4 (AQP4-IgG or NMO-IgG). The generation of AQP4-IgG leads to the loss of AQP4 function, destruction of astrocytes via complement-dependent cytotoxicity (CDC), antibody-dependent cell-mediated cytotoxicity, and subsequent inflammatory nerve damage. Therefore, preventing NMO-IgG-AQP4 binding may represent a feasible therapeutic strategy for NMOSD. In this study, we generated a monoclonal antibody targeting th…

Pericyte-glial cell interactions: Insights into brain health and disease.

Pericytes are multi-functional mural cells of the central nervous system that cover the capillary endothelial cells. Pericytes play a vital role in nervous system development, significantly influencing the formation, maturation, and maintenance of the central nervous system. An expanding body of studies has revealed that pericytes establish carefully regulated interactions with oligodendrocytes, microglia, and astrocytes. These communications govern numerous critical brain processes, including angiogenesis, neurovascular unit homeostasis, blood-brain barrier integrity, cerebral blood flow regu…

Cytomegalovirus Infection Affects the Differentiation and Determines the Functionality of a Human Progenitor Neural Cell Line.

Cytomegalovirus (CMV) is a leading cause of congenital infections and one of the most common causes of neurodevelopmental disabilities worldwide. Despite the critical consequences of CMV infection in the brain, little is known about the mechanisms of neuropathogenesis responsible for malformations and dysfunction in the central nervous system. Some reports point out the infection of neural precursor cells (NPCs) as a key element in this process; these cells show the greatest susceptibility to CMV in the developing brain. In order to further characterize how CMV perturbs NPCs properties, we inv

Low intensity focused ultrasound stimulation targeted on M1 ameliorates neuroinflammation in hemi-parkinsonian rats.

OBJECTIVE: Low intensity focused stimulation (LIFUS) has been proved to improve motor function in Parkinson's disease (PD) animal modules. The aim of this study is to investigate whether LIFUS target on the primary motor cortex (M1) can improve motor deficit in the PD rats. METHODS: The PD rat model was induced by injection of 6-hydroxydopamine (6-OHDA) in the medial forebrain bundle (MFB). Two weeks after the injection, LIFUS was used on PD rats for two weeks. Behavioral tests were performed including open field test and rotarod test to examine the motor ability of the rats. The activity of

Juvenile peripheral LPS exposure overrides female resilience to prenatal VPA effects on adult sociability in mice.

Autism spectrum disorder (ASD) exhibits a gender bias, with boys more frequently affected than girls. Similarly, in mouse models induced by prenatal exposure to valproic acid (VPA), males typically display reduced sociability, while females are less affected. Although both males and females exhibit VPA effects on neuroinflammatory parameters, these effects are sex-specific. Notably, females exposed to VPA show increased microglia and astrocyte density during the juvenile period. We hypothesized that these distinct neuroinflammatory patterns contribute to the resilience of females to VPA. To in

APOE4 impairs the microglial response in Alzheimer's disease by inducing TGFβ-mediated checkpoints.

The APOE4 allele is the strongest genetic risk factor for late-onset Alzheimer's disease (AD). The contribution of microglial APOE4 to AD pathogenesis is unknown, although APOE has the most enriched gene expression in neurodegenerative microglia (MGnD). Here, we show in mice and humans a negative role of microglial APOE4 in the induction of the MGnD response to neurodegeneration. Deletion of microglial APOE4 restores the MGnD phenotype associated with neuroprotection in P301S tau transgenic mice and decreases pathology in APP/PS1 mice. MGnD-astrocyte cross-talk associated with β-amyloid (Aβ) p

Actigraphic evidence of persistent sleep disruption following repetitive mild traumatic brain injury in a gyrencephalic model.

We studied the effect of multimodal traumatic brain injuries on daily sleep/activity patterns and related histology. Gyrencephalic ferrets wore actigraphs and received military-relevant brain injuries including shockwaves, strong rotational impact, and variable stress, which were evaluated up to 6 months post injury. Sham and Baseline animals exhibited activity patterns occurring in distinct clusters of high activity, interspersed with periods of low activity. In the Injury and Injury + Stress groups, activity clusters diminished and overall activity patterns became significantly more disperse

Low-intensity open-field blast exposure effects on neurovascular unit ultrastructure in mice.

Mild traumatic brain injury (mTBI) induced by low-intensity blast (LIB) is a serious health problem affecting military service members and veterans. Our previous reports using a single open-field LIB mouse model showed the absence of gross microscopic damage or necrosis in the brain, while transmission electron microscopy (TEM) identified ultrastructural abnormalities of myelin sheaths, mitochondria, and synapses. The neurovascular unit (NVU), an anatomical and functional system with multiple components, is vital for the regulation of cerebral blood flow and cellular interactions. In this stud

Fluid transport in the brain.

The brain harbors a unique ability to, figuratively speaking, shift its gears. During wakefulness, the brain is geared fully toward processing information and behaving, while homeostatic functions predominate during sleep. The blood-brain barrier establishes a stable environment that is optimal for neuronal function, yet the barrier imposes a physiological problem; transcapillary filtration that forms extracellular fluid in other organs is reduced to a minimum in brain. Consequently, the brain depends on a special fluid [the cerebrospinal fluid (CSF)] that is flushed into brain along the uniqu

Inhibition of neuronal necroptosis mediated by RIP1/RIP3/MLKL provides neuroprotective effects on kaolin-induced hydrocephalus in mice.

OBJECTIVES: Necroptosis is widespread in neurodegenerative diseases. Here, we examined necroptosis in the hippocampus and cortex after hydrocephalus and found that a necroptosis pathway inhibitor alleviates necroptosis and provides neuroprotective effects. MATERIALS AND METHODS: Hydrocephalus was induced in C57BL/6 mice by kaolin. Haematoxylin and eosin (HE), Nissl, PI and Fluoro-Jade B (FJB) staining were used for general observations. Phosphorylated receptor-interacting protein kinase 3 (p-RIP3) and phosphorylated mixed lineage kinase domain-like (p-MLKL) were measured by Western blotting a

Müller glial responses compensate for degenerating photoreceptors in retinitis pigmentosa.

Photoreceptor degeneration caused by genetic defects leads to retinitis pigmentosa, a rare disease typically diagnosed in adolescents and young adults. In most cases, rod loss occurs first, followed by cone loss as well as altered function in cells connected to photoreceptors directly or indirectly. There remains a gap in our understanding of retinal cellular responses to photoreceptor abnormalities. Here, we utilized single-cell transcriptomics to investigate cellular responses in each major retinal cell type in retinitis pigmentosa model (P23H) mice vs. wild-type littermate mice. We found a

Emerging therapeutic targets for neuromyelitis optica spectrum disorder.

Introduction: Neuromyelitis optica spectrum disorder (NMOSD) is an inflammatory demyelinating disease of the central nervous system affecting primarily the spinal cord and optic nerves. Most NMOSD patients are seropositive for immunoglobulin G autoantibodies against astrocyte water channel aquaporin-4, called AQP4-IgG, which cause astrocyte injury leading to demyelination and neurological impairment. Current therapy for AQP4-IgG seropositive NMOSD includes immunosuppression, B cell depletion, and plasma exchange. Newer therapies target complement, CD19 and IL-6 receptors.Areas covered: This re
