Galectin-3 regulates erythropoiesis and enhances the immunoregulatory properties of CD71+ erythroid cells across developmental stages.

Galectins are expressed by different immune and nonimmune cells with diverse immunomodulatory properties. However, their roles in erythropoiesis remain unknown. We investigated the expression of galectin genes in splenic CD71+ erythroid cells (CECs) from neonatal BALB/c mice at various developmental stages using bulk RNA sequencing. Our analysis revealed distinct gene expression profiles at different ages. Specifically, CECs from day-3 mice had a markedly different expression pattern compared to those from days 6, 12, and 28. Notably, Lgals1, Lgals3, Lgals4, Lgals8, and Lgals9 were constitutiv

Cytokine and metabolite networks shape T cell residency and functionality at the term human maternal-fetal interface.

Placentation presents immune conflict between mother and fetus, yet in normal pregnancy maternal immunity against infection is maintained without expense to fetal tolerance. This is believed to result from adaptations at the maternal-fetal interface (MFI), which affect T cell programming, but the identities (i.e. memory subsets and antigenic specificities) of T cells and the signals that mediate T cell fates and functions at the MFI remain poorly understood. We found intact recruitment programs as well as proinflammatory cytokine networks that can act on maternal T cells in an antigen-independ

RTF1 mediates epigenetic control of Th17 cell differentiation via H2B monoubiquitination.

A gene encoding the transcription factor RTF1 has been associated with an increased risk of ulcerative colitis (UC). In this study, we investigated its function in modulating T cells expressing interleukin-17A (Th17 cells), a cardinal cell type promoting intestinal inflammation. Our results indicate that Rtf1 deficiency disrupts the differentiation of Th17 cells, while leaving regulatory T cells (Treg) unaffected. Mechanistically, RTF1 facilitates histone H2B monoubiquitination (H2Bub1), which requires its histone modification domain (HMD), for supporting Th17 cell function. Impaired Th17 diff

Efficient expansion of tumor-infiltrating lymphocytes from gynecologic cancer.

Tumor-infiltrating lymphocyte (TIL) therapy is a type of adoptive immunotherapy potentially applicable to many types of solid tumors. Although gynecologic malignancies are promising targets for TIL therapy, its objective efficacy has not been established. Current TIL culture typically involves incubation of dissociated samples with high-dose IL-2 (HD-IL2) for weeks to enrich tumor-reactive T cells. While this protocol has been successfully used for melanoma TIL, it has not necessarily been optimized for other cancers. Here we investigated the method of efficiently expanding TILs derived from p

Activation and exhaustion of CD8 T cells in patients with chronic lymphocytic leukemia treated with ibrutinib and pembrolizumab.

Immune checkpoint blockade has been shown to restore anti-tumor T-cell function and elicit durable responses in select solid and hematopoietic malignancies. However, single-agent anti-programmed death 1 (PD-1) antibodies proved less efficacious in patients with chronic lymphocytic leukemia (CLL). In patients with high-risk or relapsed/refractory CLL, we conducted a phase 2 study testing the combination of lead-in ibrutinib and up to 2 cycles of fludarabine, followed by continuous therapy with ibrutinib and 17 cycles of pembrolizumab administered every 3 weeks. A total of 15 patients were enrol

The transcription factor RORα is required for the development of type 1 innate lymphoid cells in adult bone marrow.

Type 1 innate lymphoid cells (ILC1s) respond to infections and tumors by producing IFN-γ. Although RAR-related orphan receptor α (RORα) is required for ILC2s and some ILC3s, its role in ILC1 development remains controversial. To investigate the role of RORα in ILC1s, we analyzed bone marrow (BM) chimeras of RORα-deficient mice. ILC1s derived from RORα-deficient BM cells were significantly reduced in various tissues, including the intestine, indicating a hematopoietic cell-intrinsic need for RORα in ILC1 development. Developmental stage-specific RORα-deficient mice showed a decrease in adult li

Cell death in the brain during parasitic infections.

Programmed cell death is an integral arm of immunity, especially in response to intracellular pathogen infection, for the release of inflammatory mediators, and the clearance of pathogen-infected cells. In the central nervous system, cell death as a form of immunity may seem paradoxical; neuronal health is critical to survival and neurogenesis is nominal post gestation. However, the brain is composed of various glial cell types that have a greater regenerative capacity than neurons, thus allowing for cell death to potentially be utilized as a protective immune response. While the brain is an i

Distinct expression of CD56 and CD19 marks molecular and functional endotypes of tetanus- versus RBD-specific human bone marrow plasma cells.

Plasma cell survival is influenced by various factors, including soluble mediators, intrinsic and extrinsic signals as well as adhesion molecules defining the bone marrow microenvironment. The role of their induction, turnover and competition dynamics among different antigen-specific bone marrow plasma cell subsets is not well understood. This study addresses the co-expression of CD19 and CD56 on both antigen-specific and total human bone marrow plasma cells (BMPC) using multiparametric flow cytometry and data from a previous single-cell RNA sequencing (scRNA-seq) study. While the RNA-seq data

Role of companion dogs in cancer immunotherapy development.

Rodent models for evaluating new cancer immunotherapies often fail to fully recapitulate the complexity of immune interactions that occur in cancer patients. Companion dogs with spontaneously developing cancers more closely resemble humans immunologically than rodents. This resemblance results in part because their immune systems have been educated early by repeated vaccinations and viral and bacterial infections, with further sculpting later in life by tumors that typically take months to develop. Moreover, dogs possess all of the major leukocyte subsets present in humans, both in circulation

Regulatory T cell memory: implications for malaria.

Regulatory T cells (Tregs) can persist as memory cells (mTregs) in both infectious and non-infectious settings. However, their functional behavior, phenotypic stability, and suppressive properties upon antigen re-exposure remain poorly understood. Emerging evidence suggests that mTregs exhibit enhanced proliferation and suppressive capacity upon re-encountering the same antigen, a feature that may be critical in recurrent infections such as malaria. In malaria, Tregs are known to modulate immune responses and influence acute disease outcomes, suggesting that mTreg recall may play a significant

Impact of obesity on the CCR6-CCL20 axis in epidermal γδ T cells and IL-17A production in murine wound healing and psoriasis.

Obesity is associated with comorbidities including type 2 diabetes, chronic nonhealing wounds, and psoriasis. Normally, skin homeostasis and repair is regulated through the production of cytokines and growth factors derived from skin-resident cells including epidermal γδ T cells. However, epidermal γδ T cells exhibit reduced proliferation and defective growth factor and cytokine production during obesity and type 2 diabetes. One of the genes modulated in epidermal γδ T cells during obesity and type 2 diabetes is CCR6, which is the receptor for CCL20. CCL20 is elevated in the skin during obesit

The 4 functional segments of Factor H: Role in physiological target recognition and contribution to disease.

Factor H controls proximal complement activation, and its dysfunction leads to diseases that often manifest in the kidney. Structural and functional analyses have identified 4 distinct functional segments: an N-terminal regulatory unit, a cell binding unit, a segment with combined low-affinity C3b and heparin sites, and a C-terminal recognition or sensor unit with overlapping C3b/C3d and heparin sites. Three segments are linked to diseases. The regulatory segment is affected in C3 glomerulopathy and antineutrophil cytoplasmic antibody-associated vasculitis. The second segment includes the Y402

Adjuvant conditioning enhances neutrophil function while inducing a suppressive peritoneal macrophage phenotype.

Adjuvants are widely used to boost the immune response during vaccination protocols. Our group has previously reported that repeated intraperitoneal administration of alum in mice, known as adjuvant conditioning (AC), creates an immunosuppressive environment that delays allogeneic graft rejection through NLRP3-dependent MDSC expansion. However, little is known about the effects of AC on the reprogramming of peritoneal cavity cells, particularly the different peritoneal macrophage populations, and the impact on the adaptive immune response. We found a population-specific immune response to alum

TLR7-mediated immune response of renal myeloid-derived suppressor cells via RUNX1-KLF4 in systemic candidiasis.

TLR7 and myeloid-derived suppressor cells (MDSCs) play unique roles in determining host resistance to candidiasis. However, the precise mechanisms of TLR7 in MDSC differentiation and functionality during Candida albicans infection remain elusive. We found that compared with wild-type mice, kidney injuries and inflammation were significant in Tlr7 knockout mice. Tlr7 deficiency impeded the differentiation and maturation of mature myeloid cells and stimulated MDSC expansion. Furthermore, the absence of Tlr7 enhanced the immunosuppressive ability of infected MDSCs. Contrarily, the treatment of th

Effects of PARP1 inhibitor PJ-34 on TGFα, IL-6, and IL-1β levels in diabetic nephropathy.

Diabetic nephropathy is a severe chronic complication characterized by cytotoxicity, inflammation, and fibrosis, ultimately leading to renal failure. This study systematically investigated the effects of the PARP1 inhibitor PJ-34 on high glucose-induced cytotoxicity, inflammation, and fibrosis in HK-2 cells, as well as its improvement on neuropathic pain response and transforming growth factor β (TGFβ) expression in a type 1 diabetes mellitus diabetic nephropathy mouse model. Through cellular and animal experiments, we observed that PJ-34 significantly enhanced the proliferative capacity of ce

Memory-like NK cell differentiation, inhibitory NKG2A blockade, and improved recognition via antibody or CAR engineering combine to enhance NK cell attack against multiple myeloma.

Natural killer (NK) cells are a promising approach for cellular cancer immunotherapy and are being investigated to treat patients with multiple myeloma (MM). We found that MM patient blood NK cell frequencies were normal with increased activating receptors and cytotoxic granules, without evidence of functional exhaustion. Despite this activated state, MM target cells were resistant to conventional NK cells by unclear mechanisms. Memory-like (ML) NK cells are generated after brief activation via the interleukin (IL)-12, IL-15, and IL-18 receptors and exhibit multiple enhanced antitumor properti

High fat diet feeding impairs neutrophil phagocytosis, bacterial killing, and neutrophil-induced hematopoietic regeneration.

The prevalence of obesity and metabolic diseases have risen significantly over the past decades. Chronic inflammation in obesity is a link between obesity and secondary disease. While macrophages and monocytes are known to contribute to metabolic disease risk during diet exposure, little is known about the contribution of neutrophils. We assessed the impact of obesity on neutrophils using a 16-week model of diet-induced obesity. Bone marrow (BM) neutrophils significantly expanded with chronic high-fat diet (HFD), significantly decreased TNFɑ protein release, and impaired neutrophil regenerativ

Sterile production of interferons in the thymus.

T-cell central tolerance is controlled by thymocyte TCR recognition of self-peptides presented by thymic APCs. While thymic epithelial cells are essential for T-cell central tolerance, a variety of other traditional APCs also play critical roles in T-cell selection. Similar to how peripheral APCs require activation to become effective, thymic APCs also require activation to become tolerogenic. Recent studies have identified IFNs as an essential factor for the activation and generation of an optimally tolerogenic thymic environment. In this review, we focus on interferon (IFN) production within

CD209d/e are required for macrophage-mediated phagocytosis and activation during methicillin-resistant Staphylococcus aureus pulmonary host defense.

Staphylococcus aureus is a commensal and opportunist pathogen of the upper respiratory tract. The recognition of pathogen-associated molecular patterns through pattern-recognition receptors is crucial for eliminating microorganisms such as S. aureus. DC-SIGN (CD209) is a pattern-recognition receptor that binds to a broad range of pathogens, promoting phagocytosis. Here we aimed to study the role of mouse homologues of DC-SIGN, CD209d/e, in a methicillin-resistant S. aureus (MRSA) pulmonary infection model. CD209d/e-/- and wild-type C57BL/6 mice were infected with MRSA and inflammatory paramete

RACGAP1 promotes tumor progression by influencing neutrophil recruitment and tumor cell proliferation in colorectal cancer.

Colorectal cancer (CRC) progression is closely associated with the dynamic evolution of the tumor microenvironment. As a key component of the tumor microenvironment, tumor-associated neutrophils exhibit protumor properties and dynamically regulate malignant processes by secreting bioactive mediators that coordinate interactions among immune cells. RACGAP1, a cytoplasmic division protein, is highly expressed in various cancers. Here, we investigated the impact of RACGAP1 on CRC cells and the immune microenvironment. Bioinformatics analysis revealed that RACGAP1 was overexpressed in CRC patients
