Scientific opportunities have never been better than today! The completion of the Human Genome project has sparked hope and optimism that cures for debilitating conditions can be achieved and tailored to individuals and communities. The availability of reference genome sequences and genetic variations as well as more precise correlations between genotype and phenotype have facilitated the progress made in finding solutions to clinical problems. While certain craniofacial and oral diseases previously deemed too difficult to tackle have benefited from basic science and technological advances ove

Experiencing auditory verbal hallucinations is a prominent symptom in schizophrenia that also occurs in subjects at enhanced risk for psychosis and in the general population. Drug treatment of auditory hallucinations is challenging, because the current understanding is limited with respect to the neural mechanisms involved, as well as how CNS drugs, such as antipsychotics, influence the subjective experience and neurophysiology of hallucinations. In this article, the authors review studies of the effect of antipsychotic medication on brain activation as measured with functional MRI in patients

Translational Medicine is an evolutional concept that encompasses the rapid translation of basic research for use in clinical disease diagnosis, prevention, treatment and finally in public health promotion. It follows the idea "from bench to bedside and back", and hence relies on cooperation between laboratory research and clinical care. Translation process is a complex process that requires both research and non-research activities. During the past ten years, there has been intense interest in the development of new clinical procedures, therapeutic molecules, and prototypes based on translati

The need for a collaborative approach to complex inherited diseases collectively referred to as laminopathies, encouraged Italian researchers, geneticists, physicians and patients to join in the Italian Network for Laminopathies, in 2009. Here, we highlight the advantages and added value of such a multidisciplinary effort to understand pathogenesis, clinical aspects and try to find a cure for Emery-Dreifuss muscular dystrophy, Mandibuloacral dysplasia, Hutchinson-Gilford Progeria and forms of lamin-linked cardiomyopathy, neuropathy and lipodystrophy.

Diarrhea is the second leading cause of death in children younger than 5 years. Enterotoxigenic Escherichia coli (ETEC) strains are the most common bacterial cause of diarrhea in young children living in endemic countries and children and adults traveling to these areas. Pathogenesis of ETEC diarrhea has been well studied, and the key virulence factors are bacterial colonization factor antigens and enterotoxins produced by ETEC strains. Colonization factor antigens mediate bacteria attachment to host small intestinal epithelial cells and subsequent colonization, whereas enterotoxins including

High-speed biplane x-ray and neutral density targets were used to examine brain displacement and deformation, as well as aortic motion and deformation within the mediastinum, during impact. Thirty-five impacts using eight human cadaver head and neck specimens and eight impacts of the intact cadaver thorax are summarized. During impact, local brain tissue tends to keep its position and shape with respect to the inertial frame, resulting in relative motion between the brain and skull and deformation of the brain. The local brain motions tend to follow looping patterns. Similar patterns are obser

Yersinia pestis (YP), the gram-negative plague bacterium, has shaped human history unlike any other pathogen known to mankind. YP (transmitted by the bite of an infected flea) diverged only recently from the related enteric pathogen Yersinia pseudotuberculosis but causes radically different diseases. Three forms of plague exist in humans: bubonic (swollen lymph nodes or bubos), septicemic (spread of YP through the lymphatics or bloodstream from the bubos to other organs), and contagious, pneumonic plague which can be communicated via YP-charged respiratory droplets resulting in person-person t

In the 40 years since the founding of the National Institute on Alcohol Abuse and Alcoholism (NIAAA), researchers have gained a better understanding of the brain circuits and brain chemical (i.e., neurotransmitter) systems involved in the development and maintenance of alcoholism and other drug dependence. This understanding has led to the identification of numerous potential targets for pharmacotherapy of addiction. For example, insight into the roles of signaling molecules called endogenous opioids and the neurotransmitter glutamate were fundamental for developing two medications--naltrexone

Forty years ago, alcohol was not commonly recognized as a teratogen, an agent that can disrupt the development of a fetus. Today, we understand that prenatal alcohol exposure induces a variety of adverse effects on physical, neurological, and behavioral development. Research supported by the National Institute on Alcohol Abuse and Alcoholism (NIAAA) has contributed to the identification of the range and prevalence of fetal alcohol spectrum disorders (FASD), as well as methods for prevention and treatment of FASD. The worldwide prevalence and high personal and societal costs of FASD speak to th

In mammals, the contribution of the Y chromosome is paramount for male sexual determination; however, the presence of a single functional X chromosome is also of importance. In contrast to females where X inactivation is seen; the X chromosome of the male stays active. When, due to meiotic non-disjunction events, males are born with a supernumerary X chromosome, the resulting 47, XXY karyotype is referred to as Klinefelter's syndrome. This frequent genetic condition is most commonly associated with infertility, hypogonadism, gynecomastia and cognitive impairments. The condition has also been a

Traditionally, the patients believed to be at highest risk of invasive aspergillosis (IA) are those who are neutropenic due to chemotherapy for hematological malignancy or those undergoing allogeneic hematopoietic stem cell transplantation. However, emerging data show that other patients are vulnerable to IA, even though some are not classically immunocompromised. These include: solid organ transplant recipients; patients with TB, chronic obstructive pulmonary disease and patients in the intensive care unit for other reasons. The conference highlighted the diagnostic and therapeutic challenges

Fundamental differences in the philosophy of history as an academic discipline are briefly explored, primarily from two perspectives. The traditional psychiatric and mental health nursing historian objectively uses primary sources in order to be able to make 'truth' claims about the past. The post-modern psychiatric nursing historian, on the other hand, constructs truth claims, rather than discovers them, and in the process of doing so creates historical discourses that are different from the past. To the postmodern psychiatric nursing historian, all histories are fictions, created with the us

The treatment of childhood leukaemia is seen as a successful historical example of the operation of the randomized controlled trial and continues to inform contemporary policy making on such trials within health research. This article analyses the scientists' 'story of success' through historical research. It tells us about the organizational and professional structures of such research post-war in the United Kingdom, and examines the history of the cancer clinical trial through this particular example. The story reveals a more complex picture than the 'heroic' one, with key developments in th

My interest in germ cells began when I first witnessed sea urchin fertilization and embryo development during a laboratory class at Hokkaido University, Sapporo, Japan, almost 60 yr ago. Weismann's concept of germ cells that I learned during my undergraduate years became the driving force of my entire research career. During the early years, my associates and I used mainly the golden hamster and the guinea pig as model animals because their spermatozoa had large acrosomes and we could readily follow changes in the acrosomes without killing or staining spermatozoa. We later used the mouse as ou

OBJECTIVES: This study explores communication within families of clinically significant genetics research results, after the death of the patient participant. BRCA2 mutations were found in several men after their death from prostate cancer. Spouses were given the results in a genetic counselling session and asked to inform relatives.
METHODS: Cross-sectional, qualitative exploratory study. Interviews with 13 relatives, including informers and recipients of the information, were analysed using interpretative phenomenological analysis.
RESULTS: Dissemination was hampered when communication cha
